Molecular complexity may help scientists distinguish life from nonliving chemistry, but it is not a standalone life detector. Living systems build and maintain organized chemistry; abiotic processes also create organic molecules, and scientists have not established how complex those nonliving products can become under habitable conditions. A possible signal must therefore be interpreted alongside the sample’s environment and plausible nonbiological ways it could have formed.
What molecular complexity means
Complexity is more than molecular size. A large molecule is not automatically a biological one, and a useful assessment considers several properties together:
- Composition: the kinds and ratios of atoms, including the diversity of elements other than carbon and hydrogen.
- Structure: how atoms are arranged, including geometry, functional groups, and possible folding or binding features.
- Assembly: how many steps are required to construct a molecule from simpler pieces, as captured by a defined measure such as molecular assembly index.
- Organization: patterns across molecules, such as polymerization, a preference for particular isomers, or relationships among molecules in an ensemble.
- Context: whether the observed pattern is unusual given the sample’s chemistry and environment.
That last comparison is the interpretive goal, not a settled universal formula. The National Academies discusses these dimensions as areas for investigation, not as a single validated test or cutoff.

Life uses energy and raw materials to build and sustain molecular complexity and information processing. On Earth, biological chemistry depends on a solvent and conditions that let molecules form, persist, and interact. It is therefore plausible that biological activity can produce organized chemical patterns that differ from the local abiotic background.

This idea also fits the search for agnostic biosignatures: features that might indicate life without assuming that extraterrestrial organisms use Earth’s molecules or familiar metabolisms. Molecular complexity is a candidate because it focuses on broader properties of organized chemistry. That makes it promising, but does not make it conclusive.
How scientists might measure complexity
Molecular assembly index
Molecular assembly index (MA) is one proposed metric. It describes the minimum number of steps needed to construct a molecular graph from basic pieces. A 2021 NASA Technical Reports Server record for research on assembly theory and mass spectrometry reports calculations involving several million molecules and experimental determination using mass spectrometry. This offers a possible route from an abstract idea to instrument-based measurements; it does not establish a universal MA threshold that proves biology.

Multiple dimensions and a baseline
An assembly index cannot by itself capture every feature relevant to a sample. Scientists may also examine composition, structure, folding or binding, polymerization, isomeric preference, and organization among molecules. Those measurements need comparison with an appropriate abiotic baseline: a way to judge whether the observed pattern is exceptional for the local chemistry rather than merely complicated in isolation.
Why nonliving chemistry complicates the interpretation
Abiotic and prebiotic processes can generate organic molecules. The unresolved question is how much molecular complexity and organization can arise without life under habitable conditions, and how that range compares with prebiotic experiments. The National Academies notes that natural terrestrial samples are difficult places to establish the baseline because biology dominates and can mask abiotic or prebiotic signals.

Controlled laboratory work, field settings with low biological activity, modeling, and samples from other planetary environments can help clarify what nonliving chemistry can produce. For a potential biosignature, the practical bar is to rule out plausible abiotic and prebiotic formation mechanisms before calling the result unambiguous evidence of life. An established high-complexity abiotic endmember would help test such claims.
Why environmental context matters
A complexity measurement is more informative when paired with the sample’s geochemistry and plausible reactions. Scientists need to ask what materials were available, what energy sources and catalysts were present, and which processes might have produced or destroyed the measured molecules.

For life as observed on Earth, NASA’s summary of life’s requirements identifies an energy source such as light or chemical energy; elemental raw materials including carbon, hydrogen, oxygen, nitrogen, phosphorus, and sulfur; liquid water; and suitable ranges of temperature, pH, salinity, and pressure. The authors caution that the requirements for life’s origin may have been stricter and remain incompletely understood. Finding habitable conditions is not, by itself, evidence that a sample is inhabited.

How to assess a reported complexity signal
When evaluating a claim about a sample, look for a clear account of both the measurement and its alternatives:

- Identify the metric. Find out whether the claim concerns molecular size, assembly index, structural features, organization across molecules, or a combination.
- Examine the sample’s chemistry. Consider its molecular composition, structure, functional groups, possible folding or binding, polymerization, and isomeric patterns.
- Check the baseline. Ask what abiotic or prebiotic chemistry could produce in that environment, and whether the comparison is suitable for the sample.
- Consider the setting. Review available raw materials, energy sources, catalysts, and conditions that affect molecular formation and stability.
- Look beyond abundance. Abundance and distribution may inform the interpretation, but abundance alone does not establish a biological origin.
- Check how alternatives were tested. A strong life-detection claim should account for plausible nonliving formation pathways rather than treating complexity as proof.
The National Academies’ 2023 report recognizes these comparison dimensions but does not provide a universally accepted cutoff. As the report puts it, “The degree of molecular complexity reflects the extent of organic chemical evolution in an environment and can possibly serve to discriminate biotic from abiotic systems.” The key word is “possibly”: complexity can contribute evidence, but its meaning depends on the chemistry and context in which it is found.
