In a rat study, researchers produced a sciatic nerve block lasting two to three weeks after one injection of tetrodotoxin carried in specially designed liposomes. That is a substantial preclinical result—but it does not mean scientists have developed a weeks-long local anesthetic that people can receive. Human safety, dose, duration and effectiveness have not been established.
What did the researchers develop?
The team designed liposomes—tiny lipid-based carriers—to hold and slowly release hydrophilic drugs, which dissolve readily in water. The formulation used the unsaturated phospholipid 1,2-dioleoyl-sn-glycero-3-phosphocholine, or DOPC, and carried tetrodotoxin, the active drug used in the nerve-block experiment. The study was published in Nature Biomedical Engineering on September 23, 2026. Read the paper.
Tetrodotoxin is not a commercially used local anesthetic for patients. The experiment therefore tested an investigational drug-and-carrier formulation, not an available version of a familiar anesthetic.

What happened in the rat experiment?
After a single administration, the DOPC liposomes loaded with tetrodotoxin produced sciatic nerve blockade lasting two to three weeks in rats. The study reported no systemic toxicity in that animal experiment. This finding describes the tested rats and formulation; it does not establish safety in people or rule out risks under other conditions.

Why did the DOPC liposomes release drug slowly?
In matched laboratory comparisons with saturated phospholipid analogues, the study reported that DOPC liposomes had approximately threefold higher drug loading, elevenfold lower initial release and fourfold lower release at seven days. These are results under the researchers’ experimental conditions, not universal performance figures for liposomes.
The authors observed complex multilamellar and multivesicular structures in the carriers made with unsaturated lipids. They propose that the multiple compartments create diffusion barriers that slow drug escape. The release behavior also applied to several hydrophilic drugs, including small molecules, peptides and proteins, but not amphiphilic molecules. The study describes the formulation and experiments.

Does this mean people can get a nerve block that lasts for weeks?
No. The weeks-long result was demonstrated in rats using tetrodotoxin-loaded liposomes. The study does not establish a human treatment, clinical dose, duration, safety profile or effectiveness.
