A 2023 study reported two nearly cubic molecular nanocapsules whose calcium-containing seams enclose accessible internal volumes of about 3,500 ų and 2,500 ų. The researchers also showed that the capsules could hold fluorescent reporter molecules and release them over roughly an hour. That is a laboratory proof of concept—not evidence of a working drug-delivery product.
What are calcium-seamed nanocapsules?
They are molecular assemblies made from C-ethylpyrogallol[4]arene units and calcium-containing polyatomic ions. In the structures reported by Sikligar and colleagues, the assemblies have an approximately cubic shape: the pyrogallol[4]arene units form the cube’s 12 edges, while polyatomic cations bridge its six faces. The resulting cavity is enclosed from the exterior in the crystal structures described in the study.

How large are the internal cavities?
The authors report accessible internal-volume estimates of approximately 3,500 ų for capsule 2 and 2,500 ų for capsule 3. The smaller cavity is partly occupied by the polyatomic ion in its seam. These are molecular-scale volume estimates, not dimensions of a ready-made container or a measurement of how much medicine a product could hold.

How do the two structures differ?

The second structure’s formation is not fully explained. Attempts to reproduce it by deliberately adding sodium and formate instead produced known calcium formate crystals. The study therefore establishes the capsule’s structure but does not provide a reliable general recipe for assembling that seam. The paper describes the synthesis and structural analysis.
What did the release experiments demonstrate?
The researchers tested the capsules with two fluorescent reporters, Nile red and pyranine. Both were encapsulated and released over roughly an hour, but their release profiles differed. Nile red showed a notable initial burst followed by slower release; pyranine release was steadier.

The authors discuss guest properties—including pyranine’s charge and larger size—and interactions between guest molecules and the capsule walls as possible contributors to the differing behavior. They state that detailed entrapment mechanisms and release kinetics were outside the study’s scope. The experiments demonstrate reporter encapsulation and release, not a method for precisely controlling delivery.

No. The authors suggest that future work might explore cargo such as pharmaceuticals, nutrients, preservatives, or antioxidants, but the study does not establish those uses. It reports no clinical efficacy, safe dose, patient testing, commercial product, or scale-up process. Predicting cavity size or which cargo a capsule will select from its seam composition is also not established as a general design rule.
