Co-ASS is an experimental aspirin-derived cobalt–alkyne compound—not ordinary aspirin with cobalt added. Studies have reported activity in cancer cell cultures and proposed mechanisms that include cyclooxygenase (COX) inhibition, but these results do not show that Co-ASS prevents or treats cancer in people. No human efficacy, patient-use dose, clinical approval or clinical safety profile is established by the sources discussed here.
What is the aspirin–cobalt complex?
The name refers to Co-ASS, identified in a 2005 study as [2-acetoxy-(2-propynyl)benzoate]hexacarbonyldicobalt. It is an organometallic compound built from an aspirin-derived component and a cobalt–alkyne structure. It is chemically distinct from acetylsalicylic acid, the medicine commonly called aspirin. What did the cell studies find?
Co-ASS and breast-cancer cell models
The 2005 study examined Co-ASS and related compounds in MCF-7 and MDA-MB-231 breast-cancer cells. The researchers investigated cellular uptake, lipophilicity, DNA binding, nuclear drug content, glutathione reductase inhibition, COX inhibition, estrogen-receptor binding and apoptosis. The authors reported that the most antitumour-active compounds were potent COX-1 and COX-2 inhibitors, and concluded that COX inhibition probably plays a major role in the compounds’ action. Chlorinated derivatives in three cell lines
A 2018 study tested chlorinated aspirin-derived cobalt–alkyne complexes in cell assays. It reported antiproliferative activity across the tumour-cell lines HT-29 and MDA-MB-231, with lower activity in COX-1/2-negative MCF-7 cells. The reported IC50 ranges—the concentrations associated with a 50% reduction in the measured response under those assay conditions—were:

The same article reported no effect on growth of the non-tumorigenic HS-5 cell line under its assay conditions. That finding is limited to the tested cells and experiment; it does not establish safety in humans. The figures are for the chlorinated derivatives and their specified cellular systems, not a dose for patients or a potency comparison with unchlorinated Co-ASS. Royal Society of Chemistry, 2018
Co-ASS in mesothelioma cell models
A 2013 study investigated Co-ASS in malignant pleural mesothelioma cell lines. Its abstract reports stronger activity in sarcomatoid than in epithelioid models. The authors suggested that, in this cell-model context, Co-ASS might be better considered a carbon-monoxide-releasing NSAID than an antitumour drug candidate. How should the findings be interpreted?
- The compounds are related but not interchangeable. The 2018 potency figures refer to chlorinated derivatives, while the 2005 and 2013 studies examined Co-ASS or Co-ASS-type complexes.
- The models and endpoints differ. Results from particular cultured cell lines do not establish what happens in a tumour in a person. Enzyme inhibition and cell-growth assays also answer different questions.
- COX is a proposed part of the explanation. The 2005 authors linked activity to COX inhibition, while the later mesothelioma paper emphasized a possible CO-releasing NSAID interpretation. Neither interpretation demonstrates clinical benefit.
In their 2005 paper, Ingo Ott and coauthors wrote that their results indicated Co-ASS-type cobalt–alkyne complexes represented a new class of organometallic cytostatics, with COX inhibition “probably” playing a major role. The qualification matters: the statement summarizes experimental research, not evidence that the compound works as a cancer medicine in patients. Ott et al., 2005, PubMed
Does aspirin with cobalt treat cancer?
No. The studies described here concern a distinct experimental compound and related derivatives tested in cells. They do not establish that adding cobalt to aspirin is safe or effective, nor do they provide a patient-use dose. Ordinary aspirin and Co-ASS are different substances; evidence or clinical trials involving ordinary aspirin cannot be treated as evidence for Co-ASS. What is known about clinical status?
The sources discussed establish preclinical cell research, not human outcomes. They do not establish human efficacy, pharmacokinetics, a clinical safety profile, approval or a treatment dose for Co-ASS. A cell-culture result is a reason for further investigation, not a basis for taking or recommending the compound to prevent or treat cancer.
