A newly described bacterial defense system can make a specific DNA repeat in an unusual way: one of its proteins directs DNA synthesis without an RNA or DNA template. That expands the known chemistry of DNA synthesis, but it does not show that a protein’s sequence is copied back into nucleic acid or overturn biology’s central dogma.
What did the researchers discover?
Two 2026 studies describe DRT3, a bacterial system that helps defend against bacteriophages—viruses that infect bacteria. It contains two reverse transcriptases, Drt3a and Drt3b, and a noncoding RNA. The studies describe a defense response that can lead to abortive infection, in which an infected bacterium’s activity is curtailed to limit the phage’s spread.
The two enzymes make complementary strands of a defined, alternating DNA repeat, but they do so by different mechanisms.
Drt3a uses an RNA template
Drt3a performs a familiar kind of templated synthesis: it uses the sequence of an RNA molecule to guide DNA production. The Cell study reports that Drt3a makes poly-(dTdG) from a 5′-ACACAC-3′ RNA template.

Drt3b directs DNA synthesis without a nucleic-acid template
Drt3b makes the complementary repeat strand without using an RNA or DNA sequence as a template. The studies describe protein-directed nucleotide selection that produces a defined alternating pattern—not an arbitrary DNA sequence. The Science study describes alternating poly(GT/AC) double-stranded DNA and a protein-primed poly(AC) strand made by Drt3b.
How does this relate to the central dogma?
The central dogma is often simplified as DNA → RNA → protein. That shorthand can misleadingly suggest that proteins cannot influence DNA synthesis at all. The important question is whether information encoded in a protein’s sequence is transferred back into nucleic acid.

In DRT3, Drt3b directs production of a particular repeat pattern, but the reported findings do not show that the protein’s own sequence is read out and reconstructed as DNA or RNA. The distinction is between a protein helping determine a limited DNA product and a protein conveying its encoded sequence information back into nucleic acid. The studies establish the former, not the latter. An explanatory article about the finding discusses the central-dogma framing and attributes its wording to Francis Crick; that article is useful context, while the DRT3 papers establish the mechanism.

Why is the finding significant?
Drt3b’s template-free synthesis is an unusual extension of known DNA-synthesis chemistry. It shows that a protein can direct nucleotide selection to produce a defined DNA repeat without a nucleic-acid template. That makes the result scientifically notable, while leaving the central-dogma claim narrower than the headline might imply: the work does not demonstrate protein-to-nucleic-acid recovery of a protein’s encoded sequence.

The biological setting also matters. The studies concern bacterial defense against bacteriophages, not a process demonstrated in human cells or a medical treatment. The Science study reports cryo-electron microscopy structures at 2.6 Å resolution; that describes structural detail, not how common DRT3 is or how effective it is across bacterial populations.
