The method does not make the material transparent: it uses the light that gets through. It cannot work through a barrier that absorbs all light or when no target-linked signal can be measured.
Why scattered light can still be focused
A scattering layer scrambles coherent light into a complex speckle pattern. Although the output looks random, the relationship between the incoming optical field and the outgoing field is structured by the sample. If that relationship is measured or optimized, the input phases can be adjusted so light traveling along different paths arrives in phase at one selected location. Their interference produces a focus within the scattered output.
In this context, “opaque” usually means visually opaque or strongly scattering, not completely light-impermeable. Some light must reach the target region or produce a detectable signal associated with it. Wavefront shaping redistributes that transmitted light; it does not create photons that the material has absorbed. Three ways to find a useful wavefront

These approaches are related but not interchangeable: feedback optimization searches using a target signal, phase conjugation reverses a measured field, and a transmission matrix models many input-output relationships. What a transmission-matrix setup needs
A typical laboratory demonstration uses coherent illumination, an optical system that controls the input field, a way to measure output fields, and a stable sample during calibration and use. The SLM must suit the laser wavelength, polarization, beam aperture, and required update rate. Field measurements often use a camera together with a phase reference and an interferometric or holographic arrangement.
- Coherent light: a stable source at the wavelength used for calibration and focusing.
- Wavefront control: commonly an SLM, which changes the phase of controlled portions of the incident beam.
- Field measurement: a camera and phase-retrieval or holographic arrangement to record output amplitude and phase for known inputs.
- Computation and feedback: software to assemble the matrix, calculate the input pattern, and assess the resulting focus and background.
- Sample-specific calibration: measurements must correspond to the same optical arrangement and medium state as the eventual focusing step.
There is no universal parts list or single SLM setting: optical geometry, wavelength, sample, target region, and available feedback determine the setup.

Transmission-matrix focusing, step by step
- Define the experiment. Choose the coherent illumination wavelength, scattering sample, target output region, and field of view.
- Relay controlled light to the sample. Arrange the optics so the SLM or other wavefront-control device illuminates the sample with known input patterns.
- Measure output fields. For each known pattern, record the complex output field—amplitude and phase—using a camera and a phase reference, often through interferometry or holography.
- Assemble the transmission matrix. Each measured input-output response contributes a mapping between controlled input modes and observed output modes.
- Calculate the target pattern. Use a conjugate-field solution to reinforce the target, or a matrix pseudoinverse when suppressing selected background modes is also desired.
- Display and test the correction. Send the calculated pattern through the sample and measure both the target focus and its surrounding background.
- Recalibrate after decorrelation. If the sample or optical system changes enough that the measured mapping no longer applies, acquire an updated calibration.
One 2017 ground-glass experiment used a 532 nm, 150 mW diode-pumped laser, a phase-only HOLOEYE PLUTO SLM, four-step phase-shifting holography, and 128 Hadamard input patterns to measure its transmission matrix. Those are details of that study’s apparatus, not a universal recipe or a complete laser-safety specification; see the study for its experimental context.
What the reported results do—and do not—show
In that ground-glass experiment, the authors reported a peak-to-background ratio (PBR) of 21 using phase conjugation and 35 after transmission-matrix pseudoinversion selectively darkened bright background channels. They described the difference as a 66% PBR improvement. This comparison applies to their apparatus, sample, and field of view, not to wavefront shaping in general.

Suppressing background can improve focus contrast in a selected field of view, but it does not come for free. Degrees of freedom used to darken background can reduce peak intensity, and a phase-only SLM cannot reproduce an arbitrary computed amplitude-and-phase field exactly. The authors of the 2017 paper emphasize that accurate matrix measurement is important; air turbulence, photon shot noise, and mechanical drift can degrade it.
Biological-tissue demonstrations also need to be read as individual experiments rather than general penetration promises. A 2017 study reported polarization-modulation-based generalized digital optical phase conjugation focusing through a 3-mm-thick chicken-breast sample (study). A 2016 paper reported digital optical phase conjugation through particular biological tissue and tissue-mimicking phantom samples up to 9.6 cm thick; that result does not establish performance through arbitrary tissue at that depth (study).

In a 2013 preprint, Jian Wei Tay, Puxiang Lai, Yuta Suzuki, and Lihong V. Wang described ultrasound encoding as a virtual guide star for non-invasive, dynamic optimization. They reported that light intensity at the acoustic focus increased “by an order of magnitude” in their experiment. That is a result from their particular setup, not a standard enhancement guarantee; see Digital ultrasonically encoded (DUE) optical focusing into random media.

- Choose feedback optimization when a target-linked signal can be measured repeatedly and the system can iterate quickly enough to improve it.
- Choose phase conjugation when a guide star can emit a field that can be recorded and returned as its conjugate, and the optical system can maintain alignment and stability.
- Choose a transmission matrix when measuring many input-output responses is practical and a model of the current medium is useful—for example, to calculate a focus or suppress selected background channels.
In all three cases, calibration speed must be considered alongside sample motion. If the medium changes between measurement and correction, the stored wavefront may no longer focus where intended. The number of controlled modes, measurement noise, target field of view, and hardware’s ability to control amplitude as well as phase also bound the result. No one method guarantees an arbitrary image or unlimited contrast.
