“Differentiate” can mean two different things here: determine whether one dataset is approximately normal, or compare two or more datasets. A normal distribution does not by itself tell you which comparison test to use. First define the quantity that matters—mean, variance, or the overall distribution—then account for whether observations are independent or paired and how many groups you have.
First clarify what “differentiate” means
In statistics, the wording may refer to:
- Diagnosing normality: checking whether one sample is reasonably consistent with a normal population.
- Comparing datasets: deciding whether groups differ in their means, variances, or broader distributional shape.
- Calculus: calculating a derivative from an ordered series. A probability distribution being normal does not make a derivative meaningful; the observations must have a meaningful order and a function-like relationship.
The guidance below addresses the first two, which are the usual statistical interpretations.
How to check whether a dataset is approximately normal
Use a normal probability plot
A normal probability plot places the ordered observations against theoretical normal order-statistic medians. If the sample is approximately normal, the points should follow a roughly straight line. Systematic curvature or other departures can indicate skewness, unusually short or long tails, or other distributional features. NIST describes this diagnostic and the kinds of departures it can reveal in its normal probability plot guidance.
The plot is evidence about approximate fit, not proof that the population is exactly normal. Inspect the pattern, the size of the departures relative to the application, and whether a few extreme observations are driving the result. A formal normality test, if used, should supplement—not replace—the plot and subject-matter reasoning.

Do not treat normality as the comparison question
Normality is an assumption that can affect a method’s reliability. It is not the outcome you are automatically trying to compare. Two normal datasets can have different means, different variances, or different locations and shapes despite both being normal.
Define what difference you want to measure
| Question | Quantity (estimand) | What the result should report |
|---|---|---|
| Do typical values differ? | Difference in means (or another location measure) | Estimated difference, confidence interval, and practical meaning |
| Is one process more variable? | Ratio or difference of variances | Estimated variance contrast and uncertainty |
| Do the distributions differ in any important way? | Broader distributional comparison | Where the distributions differ, not only a single p-value |
| Does one sample resemble a normal population? | Approximate normality | Probability-plot pattern and the type and size of departures |
NIST’s comparison guidance emphasizes using tests and confidence intervals to assess differences, while keeping the scientific or operational meaning of the estimated effect in view. See NIST Technical Note 2106, Comparing Instruments, published September 30, 2020.

Match the method to the data design
Independent samples
Use this design when measurements in one group are unrelated to measurements in the other—for example, randomly selected units processed by two separate methods. The analysis must account for each group’s sample size and variability.
Paired or repeated observations
Use a paired analysis when the same units are measured twice or when observations are deliberately matched. Analyze the within-pair differences; treating paired observations as independent discards the design and can distort uncertainty.

More than two groups
With several groups, define the overall comparison first and then identify which contrasts matter. Adjust any follow-up comparisons as appropriate for the number of comparisons and the study design.
When equal variances matter
For normally distributed populations, some mean-comparison procedures use an equal-variance assumption. Do not silently presume that assumption: compare the group spreads, examine the design, and choose a procedure whose assumptions fit the data. NIST discusses this issue in its process-comparison material at “Assuming the observations are normal, do the processes have the same variance?”.

Bartlett’s test
Bartlett’s test evaluates the null hypothesis that several normal populations have equal variances. It is sensitive to departures from normality, so a significant result can reflect non-normal shape rather than a genuine variance difference.
Levene’s test
NIST presents Levene’s test as a less-sensitive alternative when normality is uncertain. It is often a more defensible variance check when the probability plot shows meaningful departures from a normal shape. The same NIST page covers Bartlett’s test and the Levene comparison.

A variance test is not a substitute for deciding what substantive difference matters. A result about spread does not answer a question about average level, and neither alone describes every distributional difference.

- State the scientific question. Write down whether the target is a mean, a variance, an overall distribution, or resemblance to normality.
- Describe the design. Record the number of groups, sample sizes, and whether observations are independent, paired, or repeated.
- Plot the data. Use a normal probability plot for each relevant sample and inspect group-specific spread, skewness, tails, and unusual observations.
- Assess variance assumptions. If a method requires equal variances, evaluate that assumption. Bartlett’s test is sensitive to non-normality; consider Levene’s test when normality is uncertain.
- Select the comparison procedure. Choose a method that matches the estimand and design rather than selecting a test solely because the data look normal.
- Quantify uncertainty. Report the estimated difference and a confidence interval, alongside any test result.
- Interpret practical importance. Explain whether the size of the difference matters in the real application; statistical significance and practical importance are not interchangeable.
How to choose the right comparison axis
| Decision point | Why it changes the analysis |
|---|---|
| Mean, variance, or full distribution? | Each answers a different substantive question and requires different summaries or tests. |
| Independent or paired observations? | Pairing changes the error structure and the correct estimate of uncertainty. |
| Two groups or several? | Multiple groups require an overall strategy and, often, controlled follow-up contrasts. |
| Is approximate normality credible? | Non-normality can affect procedures and makes Bartlett’s variance test especially unreliable. |
| Are equal variances plausible? | Some normal-population mean comparisons rely on that assumption; others do not. |
| What effect is large enough to matter? | A confidence interval and domain threshold make the conclusion useful beyond a binary p-value. |
Common mistakes to avoid
- Using “normal” as a test-selection rule. Normality alone does not identify the estimand or the design.
- Calling a probability plot proof. A straight-looking plot supports an approximation; it cannot establish exact normality.
- Comparing variances when the real question is about means. Decide the target before running a variance test.
- Applying Bartlett’s test automatically. Its sensitivity to non-normality makes it a poor default when the shape assumption is doubtful.
- Ignoring pairing. Matched measurements should be analyzed through their within-pair differences.
- Reporting only significance. Include the estimated effect, uncertainty, and practical consequences.
If you literally mean calculating a derivative
A dataset can be differentiated only when its observations represent an ordered variable—such as measurements over time or position—and a derivative is meaningful for the relationship being studied. Normality describes the distribution of observed values; it does not supply an order, a function, or a smoothing rule. For an ordered series, specify the independent variable, handle measurement noise, and choose an appropriate numerical-differentiation or modeling method separately from any normality assessment.
