Scientists observe atomic-scale change by exposing a sample to a controlled condition or stimulus and measuring how its structure responds. In-situ and time-resolved electron microscopy can produce images of that evolution, while ultrafast X-ray scattering measures changes in a scattering signal. “Real time” can mean very different time windows—from microseconds to femtoseconds—and each method works only under particular sample and instrument conditions.
What does “watching atoms move” mean?
It does not always mean filming individual atoms continuously. In electron microscopy, researchers form images of a sample and compare structural changes under specified conditions. In scattering experiments, they measure how a signal changes after a stimulus and use it to study atomic-scale motion. The output and the inference are different: an image shows structure in real space, while a scattering measurement provides evidence about structural change without being the same kind of direct image.
Time resolution describes how quickly a method can distinguish events; it is not, by itself, proof of a reaction mechanism. The sample environment, how the stimulus is applied, beam exposure, and the kind of signal measured all affect what an observation can establish.

Which methods can observe structural change?
| Method | What it measures or enables | Useful sample context | Key qualification |
|---|---|---|---|
| In-situ or environmental TEM | Real-space images of structural evolution; imaging can be combined with diffraction or spectroscopy | Materials exposed to controlled gas, liquid, temperature, or other conditions | The electron beam and sample environment can affect the process being observed (npj Materials Degradation, 2025). |
| Time-resolved or pump-probe TEM | Time-dependent imaging after a stimulus | Nanoscale chemical and physical dynamics | A 2023 review reports microsecond temporal resolution using direct-electron detectors and femtosecond regimes using pump-probe microscopy. These are different implementations, not specifications for every TEM. |
| Femtosecond X-ray scattering | Scattering measurements used to study atomic-scale motion and early steps in material transformations | Ultrafast materials dynamics | It measures a scattering signal, not the same real-space output as TEM imaging (Annual Review of Materials Research, 2017). |
| Liquid-cell TEM | Imaging of nanomaterials in a contained liquid environment | Processes that require liquid conditions | A sealed liquid cell is integrated with the TEM sample rod; cell geometry, beam damage, and image-data processing are important considerations (Nano X. Nano, 2024). |
| Time-resolved cryo-EM | Near-atomic imaging of timed biological samples, including protein dynamics | Biological specimens and initiated molecular processes | A 2024 review describes microsecond temporal and near-atomic spatial resolution as technique-level capabilities, not guarantees for every experiment. The method requires specialized sample preparation. |
How does in-situ TEM follow a material changing?
In-situ TEM studies the sample while it is exposed to controlled conditions or a stimulus. Environmental TEM can introduce gases or liquids, and researchers can investigate changes over time and temperature. The 2025 review in npj Materials Degradation describes this approach for atomic-scale dynamics in metal oxidation and corrosion. Depending on the experiment, images may be paired with diffraction or spectroscopy to provide complementary structural or chemical information.

That controlled setting is also part of the interpretation. The conditions inside a microscope setup are not automatically identical to those outside it, and the electron beam can influence a sensitive sample. Researchers therefore need to distinguish the change visible in the measurement from a claim about what caused it.
What changes in a liquid-cell experiment?
A liquid-cell holder contains the liquid in a sealed cell fitted to the TEM sample rod, allowing imaging of nanomaterials in an environment that would not be available in an ordinary open-vacuum setup. This can make liquid-dependent behavior accessible, but the cell is part of the experiment: its geometry and the electron beam can affect what is observed. A 2024 review also identifies beam damage and image-data processing as ongoing concerns. Compatibility and experimental limits depend on the particular setup.

How fast can researchers measure a change?
There is no single time scale for “real-time” microscopy. The 2023 review by Alcorn, Jain, and van der Veen in Nature Reviews Chemistry reports microsecond temporal resolution with direct-electron detectors and femtosecond regimes with pump-probe microscopy. Those figures describe advances in particular approaches, not a universal capability of electron microscopes.
In pump-probe work, an experiment initiates a process and measures its response at chosen delays. That differs from observing an evolving sample under ongoing conditions. Femtosecond X-ray scattering is another way to investigate very fast events: a 2017 review by Lindenberg, Johnson, and Reis describes scattering studies of atomic-scale motion and early material-transformation steps. Its measurement is not interchangeable with a sequence of TEM images.

- Used Book in Good Condition
Time-resolved cryo-EM applies timed measurements to biological specimens. A 2024 review in Current Opinion in Structural Biology describes microsecond temporal and near-atomic spatial resolution for the technique. These are reported method capabilities; the actual information available depends on the sample and experiment.

An observed structural change establishes what the measurement detected under its experimental conditions. Explaining why it happened requires considering those conditions and the limits of the signal. In particular, beam exposure may heat or damage a sample, and a liquid-cell geometry may shape the environment being imaged. A time-resolved result also needs to be read in light of whether it came from imaging, diffraction, or scattering, and whether it followed a controlled stimulus.

Work on single-atom dynamics under controlled reaction conditions, reviewed in Accounts of Chemical Research in 2019, illustrates the value of studying change at very small scales. It does not remove the need to separate an observed motion from an explanation of its cause. Researchers must interpret the measurement in context rather than treating temporal resolution alone as a mechanism.
