Filter paper can serve as a reaction platform for making squaramide-based compounds, according to a 2025 study by University of Naples Federico II researchers. Their method uses an ethanol-and-water solvent mixture and reports reduced water-removal work-up and fewer purification steps than the batch comparison they tested. It is a promising laboratory approach, not a universal replacement for conventional synthesis.
How does the paper-based method work?
The researchers deposited reagents in a 1:1 ethanol-to-water mixture onto filter paper. The paper distributes the reagents across the reaction area, while solvent evaporates as the reaction proceeds. The authors say this setup avoids the water-removal work-up used in their batch comparison and reduces purification steps. Those benefits describe the tested method; they should not be assumed for other reactions or laboratory conditions. The full study, published in Green Chemistry on 9 April 2025, gives the experimental details.
A 3D-printed housing held the paper in place. The authors selected filter-disc size according to reaction scale: a 2 cm disc for the 0.14 mmol setup and a 6 cm disc for the 1.4 mmol setup. The paper identifies the material as filter paper, but does not establish a particular brand or grade.

What results did the researchers report?
The paper reports results across a set of squaramide-based compounds, with yields varying by reaction. Its headline figures are maxima, not outcomes guaranteed for every substrate.

Chemistry World reported 97% yield after five minutes for small-scale benzylamine tests at room temperature. That account is a news summary; experimental context and substrate-specific details should be taken from the primary paper.
Which compounds were tested?
The reported scope includes symmetric and asymmetric squaramides made using substituted benzylamines, anilines, and selected aliphatic amines. Examples include methoxy substituents that donate electrons, as well as trifluoromethyl and nitro groups that withdraw electrons, plus halogens and other substituents. Results differ across the examples, so the maximum yield should not be read as typical for the entire scope.

How does it compare with batch synthesis?
The article compares the paper platform with batch reactions under the study’s tested conditions. Its reported practical distinctions are the paper-based reaction setup, evaporation during the reaction, and less water-removal work-up and purification than in the batch comparison. This is a comparison within one study, not evidence that paper is faster, cleaner, or simpler for every squaramide synthesis.

The study also reports one scale-up from 0.14 to 1.4 mmol. That is useful evidence that the setup can be adjusted beyond its smaller format, but it does not establish industrial-scale feasibility.

What are the method’s limitations?
The aqueous solvent system may not suit every compound. Corresponding author Margherita Brindisi told Chemistry World: “A potential limitation is the use of water-based solvents, which may not suit all compounds. However, this can be further optimised through ongoing investigation.” Solvent compatibility and reaction performance therefore need to be assessed for each intended substrate.
The authors present the platform as a promising approach, not a universal substitute for conventional synthesis. Brindisi described its possible broader use as case-dependent: “Each reaction setup has its own challenges, which should be evaluated case by case. In general, this approach represents a promising starting point that could be exploited for further applications in chemical synthesis.”

What the study means for laboratory use
This work demonstrates that filter paper can function as a reaction platform for a defined set of squaramide syntheses using ethanol and water. It reports encouraging yields, a favorable EcoScale maximum, and a tenfold laboratory scale increase, while also identifying aqueous-solvent compatibility as a constraint. It does not establish suitability for every compound, scale-up beyond the reported experiment, or industrial production.
