In a laboratory assay reported in 2004, acid-rich tripeptides were more susceptible to radical damage than other sequences tested. The researchers suggested that these peptides bind iron more strongly, bringing radical formation close to them. That result concerns a large library of short peptides under specific laboratory conditions—not a general rule about whole proteins in the body.
What did the study find?
A 2004 Chemistry World report by Caroline Evans summarized work by University of Basel researchers Helma Wennemers and Matthias Nold. They tested a combinatorial library of more than 29,000 tripeptides—short chains of three amino acids—and reported greater damage among acid-rich sequences.
The report describes radical generation under Fenton conditions, involving electron transfer from Fe2+ to oxygen or hydrogen peroxide. The explanation offered was that acid-rich peptides bind Fe2+ more tightly, so radicals form closer to those peptides. The library size is not an effect size: the report does not quantify how much more damage occurred or rank individual sequences.

Iron can participate in reactions that generate highly reactive species. If a peptide binds iron, the location of radical formation relative to that peptide could influence its chance of being modified. In the reported interpretation, stronger Fe2+ binding by acid-rich peptides helps explain their greater damage in the assay.

This is a proposed explanation for a pattern in a model system, not proof that acidic sequences are always more vulnerable. Results can depend on the radical-generating system, peptide sequence, damage measurement, and surrounding conditions.
What does this mean for proteins in living systems?
Reactive species can oxidize, nitrate, or chlorinate amino-acid residues, potentially changing protein function. Cells can process some oxidized proteins through quality-control machinery; oxidative stress can also contribute to protein inactivation, unfolding, or aggregation. Which outcome occurs depends on the reactive species, the affected residue, protein structure, and cellular context.

A tripeptide library does not reproduce the folded structure of an intact protein or the complexity of a cell. The 2004 news report does not establish that acid-rich proteins are broadly more susceptible in organisms, nor does it show that this mechanism causes a disease or contributes to aging.

What the report does—and does not—establish
- Reported: More than 29,000 tripeptide sequences were examined, and acid-rich peptides showed greater damage under the reported Fenton conditions.
- Proposed explanation: Tighter Fe2+ binding may bring radical formation closer to acid-rich peptides.
- Not specified in the accessible report: Exact assay conditions, quantitative effect sizes, a ranked list of sequences, or a direct comparison among intact proteins.
- Not demonstrated: A universal vulnerability rule for proteins in organisms or a causal link to human disease or aging.
The report cites the primary paper as M. Nold and H. Wennemers, Chemical Communications (2004), page 1800. The news summary alone does not supply enough experimental detail to assess the assay quantitatively.

Why radical measurements need careful interpretation
Researchers use methods such as electron spin resonance and spin trapping to study protein-centered radicals and oxidative changes. A review of immuno-spin trapping describes interpretive challenges: competing reactions can influence what is captured and detected, and the review noted that a standard for quantitatively measuring DMPO nitrone adducts had not been produced. A detected radical signal therefore is not, by itself, a complete measure of protein damage.
For further context, see the review on immuno-spin trapping and its methodological limitations, and reviews of oxidative protein damage and protein oxidation and cellular consequences.
