If COVID-19 symptoms begin, take a test immediately. If you were exposed but still have no symptoms, wait until at least five full days have passed after the exposure before testing. A negative home antigen result is only preliminary, so repeat it 48 hours later. FDA guidance calls for at least two antigen tests total when you have symptoms and three total when you do not.

The timing applies to U.S. CDC and FDA guidance and assumes you are using an authorized test exactly as its label directs. Test instructions, expiration information and authorized products can change, so check the package and the agencies’ current information before testing.

COVID-19 testing timing at a glance

Situation When to begin If the first home antigen test is negative
Symptoms of COVID-19 Test immediately when symptoms begin. Repeat after 48 hours. FDA recommends at least two tests total for people with symptoms.
Known exposure, no symptoms Test at least five full days after the exposure. If symptoms appear sooner, test immediately instead of waiting. Repeat after 48 hours, then 48 hours later if still negative, for three tests total.
You need one test with a more reliable negative result Ask a clinician about a molecular test such as PCR or another NAAT. CDC identifies PCR as an alternative to repeating home antigen tests; FDA says molecular tests are generally more accurate than at-home tests.

These are agency recommendations, not estimates of your personal probability of infection. Your symptoms, the exposure, recent infection, health risks and the particular test’s labeling can change the appropriate decision.

If symptoms start, test right away

Do not wait for symptoms to become severe or for several days to pass. CDC self-testing guidance says to test immediately when COVID-19 symptoms begin. Early testing can give you an answer while it is most relevant to decisions about staying home, contacting people you may have exposed and seeking medical advice.

What counts as a reason to test

Use the symptom-triggered timing even if the symptoms are mild or resemble a cold, allergies or another infection. A recent negative test does not replace a new test when a new illness begins. If you are unsure whether what you are experiencing could be COVID-19, testing is a reasonable way to reduce uncertainty rather than relying on the symptom pattern alone.

What a negative first test means

A home antigen test can miss an infection at the beginning of illness. FDA explains that antigen tests are less likely than molecular tests to detect infection early, and a negative result does not rule out COVID-19. Treat the first negative as preliminary and use the repeat schedule in the table rather than assuming the result settles the question.

After an exposure without symptoms

When you have no symptoms after an exposure, CDC says to test at least five full days after the exposure. The waiting period gives an infection more time to reach a level that a rapid antigen test can detect. Testing earlier can be useful if symptoms appear, but an early negative is especially weak evidence when you feel well.

How to count the waiting period

Use the exposure as day zero and allow five complete days to pass before the first asymptomatic test; in many calendars that means testing on day six. If the exposure happened repeatedly or you cannot identify a single exposure time, ask a health-care professional which event should anchor the count. Do not postpone testing until the scheduled day if symptoms develop first.

If symptoms appear during the wait

Switch to the symptom plan immediately. Test as soon as symptoms begin, even if fewer than five full days have elapsed since the exposure. The symptom-based instruction takes priority over the asymptomatic waiting period.

Why repeat testing matters

Antigen tests detect viral proteins and are designed to produce a fast result at home. They are convenient, but a single test has limited sensitivity early in infection and in people without symptoms. A negative result therefore answers only what that particular test detected at that particular time.

FDA advises repeating a negative antigen test after 48 hours, whether or not you have symptoms, to reduce the chance of a false-negative result. The recommended series is at least two tests total for someone with symptoms and three total for someone without symptoms. Keep the intervals close to 48 hours; taking all tests back-to-back does not provide the same information about an infection that is still developing.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

How to interpret the series

  • A positive antigen result is meaningful evidence of current infection. Follow the test’s instructions and contact a clinician if you need advice about treatment or next steps.
  • A negative first result is not a clearance certificate. Continue the repeat schedule and consider your symptoms and exposure history.
  • Repeated negative antigen results lower concern but cannot make the possibility of infection zero, particularly when symptoms persist or the exposure was substantial.
  • If you need a single, more sensitive result, discuss a molecular test with a clinician instead of relying on one rapid antigen test.

Antigen versus molecular tests

The best choice depends on whether speed, convenience or sensitivity is most important. Neither category eliminates the need to read the product’s authorized use and instructions.

Feature At-home antigen Molecular test (PCR/NAAT)
Detection method Detects viral proteins. Detects viral genetic material.
Speed and convenience Designed for rapid, at-home results and is widely available online, in pharmacies and in retail stores. Usually requires a laboratory, clinic or an authorized molecular home procedure; turnaround and availability vary.
Early or asymptomatic infection More likely to miss infection early or when no symptoms are present. Generally more accurate than at-home antigen testing, according to FDA.
After a negative result Use serial testing at 48-hour intervals. CDC identifies PCR as an alternative to serial antigen testing.
Authorization and use Check the FDA’s current authorized-test list, intended use, eligible ages and expiration information. Verify the specific test’s authorization, collection method and reporting process.

A molecular test may be the practical choice when one negative result must be more informative, when symptoms continue despite negative antigen tests or when a clinician advises it. It is not automatically available faster, so check local access before assuming it will provide a same-day answer.

Step-by-step plans

Plan A: You have symptoms now

  1. Take an authorized antigen test immediately, following every collection, timing and readout step in the package instructions.
  2. Record the date and time of the test and the result. This makes the 48-hour repeat interval clear.
  3. If the result is negative, perform a second antigen test 48 hours later. FDA recommends at least two tests total for symptomatic people.
  4. If you need a more reliable single negative result, or if symptoms continue while antigen tests remain negative, ask a clinician whether PCR or another molecular test is appropriate.
  5. Contact a health-care professional promptly if symptoms worsen, you have a condition that puts you at higher risk, or you are uncertain how to interpret the results.

Plan B: You were exposed but have no symptoms

  1. Mark the exposure as day zero and wait until at least five full days have passed.
  2. Take the first antigen test on the recommended day, using the product exactly as labeled.
  3. If it is negative, repeat after 48 hours.
  4. If the second test is also negative, repeat once more 48 hours later. This produces the three-test series FDA recommends for people without symptoms.
  5. If symptoms begin at any point, stop waiting and test immediately under Plan A.

Plan C: You need a single test with greater sensitivity

  1. Ask a clinician, laboratory or testing site about an authorized molecular test, such as PCR or another NAAT.
  2. Confirm when the specimen will be collected, how results will be delivered and whether the test is intended for symptomatic or asymptomatic use.
  3. Follow the collection and reporting instructions for that specific test; molecular tests are not interchangeable in procedure or turnaround time.

Use the test exactly as authorized

Testing accuracy depends on more than the technology. Before opening a kit, check that it is authorized for your situation and that the expiration information is current. FDA maintains a regularly updated list of authorized at-home tests and product-specific details. A package may specify eligible ages, whether an adult must collect the sample, the timing for reading the result and whether the test is for people with or without symptoms.

  • Read the complete instructions before collecting a sample; do not rely on a demonstration for a different product.
  • Use the supplied components and the stated sample type. Substituting swabs, tubes or solutions can invalidate the result.
  • Observe the exact processing and read window. Reading too early or long after the stated interval can produce a misleading interpretation.
  • Check the lot and expiration information on the package and consult the FDA’s current notices if the expiration date has been extended or questioned.
  • Keep the test and its instructions available until you have recorded the result and completed any repeat testing.

Where to obtain tests and how many to buy

CDC says at-home tests are available online and through pharmacies and retail stores. Because a negative antigen result commonly requires repeat testing, a multi-test package can be practical. That is a logistical advantage, not evidence that one brand is medically preferred.

Choose a product from the FDA’s current authorized list and verify its intended use, age range and expiration details. Availability, packaging and authorization status can change, so do not assume a kit purchased previously has the same labeling today.

When to involve a health-care professional

Contact a clinician when you are unsure whether to choose molecular testing, when symptoms persist despite negative antigen tests, or when your health history makes an accurate early answer especially important. A clinician can help decide whether PCR or another NAAT is a suitable alternative to serial antigen testing and can explain what to do with a positive result.

Seek medical advice for worsening symptoms rather than waiting for another home test. Testing guidance is general; the right action can differ with recent infection, the timing and intensity of an exposure, pregnancy, age, immune status or other health risks.

Guidance can change

CDC and FDA update self-testing recommendations, authorized products and expiration information. Before testing, check the current CDC self-testing guidance and FDA COVID-19 test information, then follow the instructions enclosed with your specific kit. The agency recommendations summarized here establish the key timing: immediate testing when symptoms begin, a five-full-day wait after exposure when you remain asymptomatic, and 48-hour repeat testing after a negative antigen result.
