Researchers identified three unusual natural products in the springtail Ceratophysella denticulata, including a structure the authors describe as the first naturally occurring fully hetero-substituted benzene. They pieced together the structures from tiny samples using several analytical methods, computer calculations and chemical synthesis. In an ant assay, only one of the three compounds was reported to significantly deter the tested predator.
What the researchers found in two springtail species
A 2024 study in Chemical Science reported three major compounds from Ceratophysella denticulata:
- 4-methoxy-5-(methylthio)benzo-1,3-dioxolane
- 5,6,7-trimethoxybenzo-1,3-oxathiolane
- 8-amino-5,6,7-trimethoxybenzo-1,3-oxathiolane
The authors describe the last compound as the first naturally occurring fully hetero-substituted benzene. That novelty claim concerns the reported molecular structure; it does not mean the substance is a medicine or an established commercial product. The same paper reports a separate compound, 2-methyl-1H-imidazo[4,5-b]pyridine, from another springtail, Hypogastrura viatica. The study appeared in Chemical Science, volume 15, pages 15332–15338.
How they identified structures from a small sample
Springtails are small soil-dwelling animals, and the amounts of chemicals available from them can be extremely limited. For the C. denticulata analysis, the team extracted material from 50 individuals—a methods sample count, not an estimate of the species’ population.

No single instrument established the answer. The researchers combined gas chromatography/mass spectrometry, high-resolution mass spectrometry and infrared spectroscopy. They also used density functional theory calculations to help prioritize possible structures. Finally, they synthesized the three major compounds, providing confirmation by comparing the products of synthesis with the natural compounds. As corresponding author Stefan Schulz put it in an interview reported by Chemistry World, “The final proof comes from synthesis.” Chemistry World’s report gives additional context on the identification work.
What the ant assay did—and did not—show
In deterrence assays involving the ant Lasius niger, the study reports significant activity for 4-methoxy-5-(methylthio)benzo-1,3-dioxolane. The result should not be generalized to the other two compounds, to other ant species or to predators as a whole: the reported finding singles out this compound in this assay.

The paper reports no antibacterial or antifungal activity for the compounds tested. Chemistry World notes the authors’ suggestion that activity against soil microbes might be disadvantageous to springtails. That is their proposed explanation, not a demonstrated mechanism. Why the discovery matters, and where its limits are
The authors place the work in a relatively understudied area: springtail chemical defenses have received less attention than insect defenses, in part because collecting enough animals and isolating measurable amounts of their chemicals is difficult. Identifying unusual compounds expands what is known about springtail chemistry and offers a specific example of how trace analysis, computation and synthesis can work together.

The study establishes chemical structures and reports a deterrent result for one compound in a particular ant assay. It does not establish a medical use, a pesticide application or another practical commercial use.
