There is no universal sample-preparation recipe for lithium-ion battery microcalorimetry. The correct sequence depends on the cell format and chemistry, whether the experiment targets total heat generation, parasitic reactions, or SEI formation, whether the cell is dry or already filled, and the calorimeter holder and control program.
Published pouch-cell work commonly combines controlled drying when cells are opened, inert-atmosphere handling, measured electrolyte filling and sealing, a defined wetting and formation history, optional degassing, and thermal equilibration in the calorimeter before measurement. The temperatures, times, voltages, and handling steps below are documented examples, not a universal standard operating procedure.
Start by defining what the measurement must represent
Microcalorimetry measures heat flow while the cell is held at a controlled temperature and electrical condition. The preparation must therefore create a known electrochemical starting state and preserve it while the cell is installed. A protocol for high-voltage parasitic reactions is not automatically suitable for a study of SEI formation or full-cell heat generation.
State the cell architecture and chemistry
Record whether the specimen is a pouch, wound cell, coin cell, or another format; the electrode materials; nominal capacity; electrode loading if relevant to the experiment; and whether it is a full cell or anode-free design. The published examples include NMC442/graphite pouch cells and anode-free pouch cells. Their different purposes produce different preparation choices.

Define the reaction or operating window
Write down whether the calorimetry will follow formation, a voltage-dependent hold, cycling heat, or parasitic reactions at a selected state of charge. The planned voltage and current program determines which formation steps, rest periods, and electrical connections must be completed before the heat-flow run.
Inspect and document the incoming cell
Before opening or connecting anything, identify the cell’s condition:
- dry and factory-sealed;
- already filled with electrolyte and sealed;
- assembled in-house but not yet filled;
- previously formed, cycled, or degassed.
Record the maker or assembly route, format, chemistry, capacity, lot or cell identifier, mass if measured, and any prior storage or cycling. A dry cell that is opened for filling requires a different moisture-control sequence from a prefilled cell that can be transferred directly to the holder. Do not describe a prefilled-cell procedure as equivalent to a refill procedure.

Opening a dry pouch exposes internal surfaces to moisture and oxygen. If the design requires drying, perform it before electrolyte filling and define how the cell moves from the dryer to the filling environment.
Documented NMC442/graphite example
Downie, Hyatt, and Dahn described machine-made 260 mAh NMC442/graphite pouch cells that were opened, dried in a heated vacuum oven at 80 °C overnight (approximately 14 hours), and transferred directly to an argon-filled glove box without exposure to ambient air. Electrolyte filling took place in the glove box. The article was published online in 2015 and is commonly cited as a 2016 publication.

That 80 °C, approximately 14-hour schedule belongs to that cell preparation and should not be treated as a universal moisture specification. A different separator, binder, electrode coating, pouch laminate, or calorimetry question may require a different validated drying process. If the cell was never opened, do not add a drying step merely because it appears in a published refill protocol.
Control the transfer
For an opened cell, specify the oven conditions, duration, pressure or vacuum description used by the laboratory, transfer route, glove-box atmosphere, and the time between drying and filling. If ambient exposure occurs, record it as a deviation rather than silently combining the cell with cells handled under inert gas.
Fill and seal with a measured electrolyte charge
Electrolyte identity and quantity are part of the sample definition. Record the formulation, supplier or preparation route where applicable, volume or mass added, filling temperature, and any differences between cells. The cited NMC442/graphite procedure filled the dried pouch cells in argon and vacuum-sealed them. A separate manufacturer-described battery calorimetry example also used electrolyte filling followed by vacuum sealing.
Use the same sealing method for every comparison group. Note whether the first seal is temporary or final, the dimensions of the seal area, and whether a tab or fill edge remains available for later gas release. These details affect internal pressure and the mechanical state seen by the calorimeter.
Wetting, formation, and degassing establish the starting state
Electrolyte does not instantly produce the same wetting and interphase condition in every cell. The wetting hold, formation sequence, gas handling, and rest periods must be treated as part of the calorimetry protocol, not as informal assembly details.
Wetting hold
In the NMC442/graphite example, filled cells were held at 40 °C and 1.5 V for 24 hours to promote complete wetting before staged formation. In Logan et al.’s 2021 anode-free pouch-cell study, an approximately 24-hour hold at 1.5 V was used to ensure positive-electrode wetting and to prevent corrosion of the copper current collector. The similar time and voltage do not make the two procedures interchangeable: the cell architectures and failure risks differ.
State which electrode condition the hold is intended to establish, the temperature, voltage, duration, and whether the cell is connected to a current source, left at open circuit, or controlled by another program.

Formation
Report every formation stage: voltage limits, current or C-rate, rest periods, temperature, and whether the cell is charged or discharged between stages. The NMC442/graphite study used staged formation. Formation history changes the interphase and residual gas, so two calorimetry datasets cannot be considered directly comparable when one cell was freshly filled and the other had completed a multistage formation sequence.
Do these 3 things before closing this tab:
1Clear out junk files and repair common Windows errors2Fix the driver behind crashes, sound loss and screen glitches3Repair Windows errors before they cause bigger problemsDegassing and resealing
Gas evolution can be part of formation. In the NMC442/graphite protocol, cells were opened under argon to release gas and then resealed after formation steps. The manufacturer-published example likewise reports opening cells to release gas and resealing them. Degassing is not automatically required for every lithium-ion cell: include it only when the cell design and study require it, and record when it occurred relative to formation and calorimetry.
Install the cell and allow thermal equilibration
The calorimeter measures the assembled specimen, holder, electrical leads, and thermal surroundings as a system. Before collecting data, specify the holder, contact arrangement, pressure or mechanical constraint, lead routing, and any compression or stack fixture. A pouch cell measured with a constrained stack is not the same sample as one measured freely supported.
Published TAM III example
Downie, Hyatt, and Dahn used a TAM III isothermal microcalorimeter set to 40.0000 ± 0.0001 °C. After formation, the cells were left at open circuit for 24 hours to thermally equilibrate, then connected to an automated battery tester. This is a documented set of conditions for that experiment, not a required setpoint for all instruments or chemistries.
Set the calorimeter temperature, stabilization criterion, and equilibration time before the run. If the instrument software reports a baseline or drift check, preserve those records with the cell file. A voltage or current program should not begin while the cell is still approaching thermal equilibrium.
What a reproducible preparation record contains
A useful laboratory record lets another operator reconstruct the cell’s chemical, mechanical, thermal, and electrical history. Include:
- cell format, electrode chemistry, nominal capacity, and identifier;
- incoming state: dry sealed, prefilled, assembled in-house, formed, or previously cycled;
- opening procedure, drying temperature and duration, vacuum or pressure description, and transfer atmosphere;
- glove-box atmosphere or other moisture-control conditions;
- electrolyte formulation, amount, filling method, and sealing method;
- wetting temperature, voltage, duration, and purpose;
- formation stages, current or C-rate, voltage limits, rests, and temperatures;
- gas release or degassing steps, atmosphere during opening, and resealing sequence;
- holder type, stack constraint or pressure, cell orientation, and lead arrangement;
- calorimeter model or configuration, setpoint, equilibration duration, and stabilization check;
- the connected battery-tester program, including cycling or voltage holds.
How to compare two preparation protocols
When deciding whether datasets can be compared, align the following axes rather than comparing only the calorimeter temperature:

Instrument and equipment considerations
The method requires an isothermal microcalorimeter and an electrochemical control system capable of the intended current, voltage, and rest program. TA Instruments’ technical e-book describes a TAM calorimeter used in battery heat-flow experiments; it is an example of a reported workflow, not evidence that one instrument is best for every sample.
Laboratories that open and refill pouches generally need a moisture-controlled work area, such as an argon glove box, together with a suitable vacuum oven, filling tools, sealing equipment, and a holder compatible with the calorimeter. Select the holder and instrument from the cell dimensions, heat-flow range, electrical connections, and required mechanical constraint. No single holder pressure, temperature, or calorimeter model has been established as universal.

Common preparation mistakes and how to prevent them
Applying a published drying schedule to every cell
Problem: Treating 80 °C overnight as a general requirement can alter materials or remove a step that a sealed cell does not need.
Prevention: Tie drying to the incoming state, cell construction, and validated laboratory procedure. Label the 80 °C, approximately 14-hour condition as the NMC442/graphite example it is.
Allowing an uncontrolled air exposure
Problem: Moving an opened cell through ambient air breaks the intended moisture history.
Prevention: Plan a direct transfer into the inert environment and record any exposure as a deviation. Do not merge those cells with an argon-handled group without an explicit experimental justification.

Leaving wetting and formation undocumented
Problem: A cell described only as “formed” may have a different interphase and gas history from another cell with the same label.

Prevention: Preserve the complete voltage, current, temperature, duration, and rest sequence, including the reason for a wetting hold.
Degassing at an undefined point
Problem: Gas release before or after formation changes internal pressure and the state entering calorimetry.
Prevention: State whether degassing was performed, the atmosphere used for opening, the resealing method, and the formation stage at which it occurred.
Starting the measurement before stabilization
Problem: Early heat flow can reflect thermal settling rather than the electrochemical event being studied.
Prevention: Set a defined equilibration period or stability criterion. The cited TAM III procedure used 24 hours at open circuit after formation.
Changing the holder between comparison groups
Problem: Different pressure, contact, or lead routing can change both heat transfer and cell behavior.
Prevention: Keep the holder configuration constant when possible and report every mechanical difference when it is not.

The following sequence shows how a published pouch-cell preparation can be written as an auditable protocol. It is an example to adapt, not a universal recipe.
- Identify a dry, sealed 260 mAh NMC442/graphite pouch cell and document its identifier and capacity.
- Open the cell as specified by the laboratory procedure.
- Dry the opened cell in a heated vacuum oven at 80 °C overnight, approximately 14 hours, as in the Downie, Hyatt, and Dahn study.
- Transfer it directly into an argon-filled glove box without ambient-air exposure.
- Add the defined electrolyte formulation and quantity, then vacuum-seal the pouch.
- Hold the filled cell at 40 °C and 1.5 V for 24 hours for wetting, matching the cited NMC442/graphite example.
- Run the documented staged formation program.
- If the study requires gas release, open the pouch under argon, release gas, and reseal it before the next formation stage or measurement.
- Install the cell in the specified holder and mechanical constraint.
- Set the TAM III example temperature of 40.0000 ± 0.0001 °C, allow 24 hours of open-circuit thermal equilibration, and then connect the automated battery tester.
- Begin the planned cycling or voltage-hold program only after the thermal and electrical starting conditions have been logged.
If the experiment uses anode-free pouches, a different chemistry, a prefilled cell, or another calorimeter, replace the example values with the validated conditions for that design and explain the change.
Pre-run checklist
- The scientific question and voltage/current program are written down.
- Cell format, chemistry, capacity, and incoming state are identified.
- Any opening, drying, atmosphere transfer, filling, and sealing steps are recorded.
- Electrolyte formulation and amount are known for each cell.
- Wetting, formation, and degassing history is complete.
- Holder, pressure or constraint, orientation, and electrical connections match the comparison plan.
- Calorimeter setpoint, baseline or stability check, and equilibration duration are documented.
- Any deviation from the intended atmosphere, temperature, timing, or electrical program is preserved with the data.
The most defensible preparation is the one that makes the cell’s prior history explicit. Published work supports a controlled sequence of moisture handling, filling, wetting, formation, optional gas release, and thermal equilibration, but it does not establish one recipe for every lithium-ion cell.
