Adding a second active pharmaceutical ingredient (API) supplier improves continuity only when it is a genuinely qualified, usable source. A second company name is not automatically a second independent supply path: both suppliers may rely on the same starting-material producer, intermediate site, testing laboratory, broker, port, utility or regional manufacturing cluster.
Qualify the specific manufacturing site and its quality system, confirm analytical and process comparability, define responsibilities in a written quality agreement, verify the regulatory path for the actual product and market, and test whether the proposed source removes or merely duplicates existing dependencies. Use a risk-based depth of review; the controls should become more stringent as API production approaches final processing, purification and packaging.
Start by defining exactly what is being diversified
Before requesting documents or scheduling an audit, write a one-page scope statement. Identify:
- The API, dosage form and finished product it will support.
- Each destination market and the relevant marketing application, master file or other filing commitment.
- The intended manufacturing site, process steps, grade, specification and impurity profile.
- Whether the source is for routine commercial supply, contingency stock, development, a specific market or a limited capacity allocation.
- Every broker, distributor, repacker, contract laboratory, warehouse and other outsourced operation involved between the API manufacturer and your site.
- The incumbent source and the precise failure that diversification is meant to mitigate: quality failure, capacity shortage, geopolitical disruption, transport interruption, single-site dependence or another risk.
Confirm which legal entity actually manufactures the API and which sites perform synthesis, fermentation or cell culture, extraction, recovery, purification, micronization, packaging, testing and release. ICH Q7 covers these API manufacturing activities, subject to its stated scope and qualifications. Its supplier-approval expectations are described in the FDA-hosted guidance ICH Q7A.

Qualify the actual manufacturer, not just the commercial supplier
Demand evidence of consistent quality
Approval should be based on evidence that the named manufacturer can repeatedly meet the agreed specification. ICH Q7 states: “Supplier approval should include an evaluation that provides adequate evidence (e.g., past quality history) that the manufacturer can consistently provide material meeting specifications.” Review the site’s relevant quality history, audit results, regulatory inspection information where available, batch records, release data and history of rejected or reworked lots. The governing Q7 guidance is available from FDA at this page.
Examine the quality system and process capability
Tailor the review to the API’s hazards and process maturity. Evidence commonly includes:
- Quality-unit independence, authority and batch-disposition controls.
- Validated or otherwise scientifically justified process controls, cleaning controls and computerized systems.
- Deviation, out-of-specification (OOS), out-of-trend and laboratory-investigation procedures, including root-cause analysis and corrective and preventive action.
- Change control, data-integrity safeguards, record retention and traceability from starting materials through shipment.
- Complaint, return, recall and market-action procedures.
- Controls for critical starting materials, intermediates, solvents, catalysts, processing aids and outsourced testing.
- Training, preventive maintenance, calibration, environmental or contamination controls where relevant, and business-continuity arrangements.
This is an applied checklist rather than a universal FDA audit form. The depth should reflect the API’s toxicity, potency, route, impurity risks, process complexity, supply criticality and the consequences of failure. The FDA’s final September 2016 Q7 guidance for industry explains the applicable GMP framework.

A different synthesis route, polymorph, particle-size distribution, impurity profile, specification, site or critical raw material can affect formulation, stability, bioavailability, downstream processing or regulatory commitments. Technical and quality teams should compare the candidate and incumbent process descriptions, controls, analytical methods, impurity fate and purge, physical attributes and stability data. Q7 establishes the need to produce API meeting its intended quality and specifications; it does not provide a product-specific comparability conclusion. That conclusion requires data for the actual API, process and finished product.

Control receipt, sampling and supplier materials
Supplier approval belongs inside the material-control system, not in procurement alone. Q7 calls for written procedures covering receipt, identification, quarantine, storage, handling, sampling, testing, approval or rejection, and purchasing to agreed specifications from suppliers approved by the quality unit. Translate those requirements into operating instructions for your receiving, warehouse, laboratory and quality teams.
Define how containers are inspected, labels and seals are verified, lots are identified, samples are taken without contamination, temperature or humidity excursions are handled, and material is released to production. Specify what happens when a shipment arrives from an unapproved site, through an unapproved intermediary or with incomplete documentation. The procedure should make it impossible for a purchasing substitution to bypass quality-unit approval.
Build an analytical qualification and COA-reliance plan
Agree the testing model before routine orders
Set the specification, analytical methods, acceptance limits, sampling plan, reference standards, method-transfer or verification work, impurity characterization and handling of discrepant results before the first commercial shipment. Decide which tests your laboratory will perform, which results may be accepted from the supplier’s certificate of analysis (COA), and how results will be reconciled when the two laboratories disagree.

When a finished-drug manufacturer relies on an API supplier’s analysis, FDA requires at least one specific identity test and requires the manufacturer to validate the reliability of the supplier’s analyses at appropriate intervals. FDA states: “Therefore, if the finished drug product manufacturer accepts the test results from an API supplier’s COA rather than performing the tests itself (other than for identity, which the manufacturer is required to perform), the manufacturer must validate the API supplier’s reliability.” The statement appears in FDA’s June 9, 2015 CGMP questions and answers on control of components.

Use the three-batch recommendation correctly
Q7 recommends complete analysis of at least three batches before reducing in-house testing, followed by complete analysis at appropriate intervals in comparison with supplier COAs and regular verification of COA reliability. Treat “three batches” as the Q7 recommendation for establishing confidence, not as a universal waiver from applicable CGMP, filing commitments or your product-specific control strategy. Document the scientific rationale, test frequency, trend review, reversion triggers and approval by the quality unit.
Put operating responsibilities in a written quality agreement
Use a quality agreement with the manufacturer and, where appropriate, with contract laboratories or other contract facilities. Keep commercial terms separate or severable from quality terms so that a price or volume dispute cannot obscure a safety or compliance obligation.
Define the subjects that cause real failures
Cover, at minimum:
- Manufacturing, laboratory, sampling, packaging, storage and release/disposition responsibilities.
- Batch records, certificates, electronic data, retention samples and record-access rights.
- Deviation, OOS, complaint, recall and investigation ownership, timelines and escalation contacts.
- Planned and unplanned changes, regulatory-impact assessments and advance-notification or approval requirements.
- Subcontracting, approved-site lists and controls over further delegation.
- Audit rights, inspection support, response to findings and corrective-action verification.
- Communication routes for shortages, allocation, transport incidents, data-integrity concerns and quality emergencies.
FDA’s final November 2016 Contract Manufacturing Arrangements for Drugs: Quality Agreements Guidance for Industry recommends documenting these CGMP-related roles. It also makes the legal boundary explicit: “Quality agreements cannot be used to delegate statutory or regulatory responsibilities to comply with CGMP.” The agreement allocates work; it does not transfer the manufacturer’s continuing obligations.

Set escalation thresholds and requalification triggers according to product criticality and risk appetite rather than copying an invented industry score. FDA’s quality-agreement guidance discusses ICH Q10 oversight and purchased-material controls and supports pre-selection assessment, ongoing performance review and improvement; the same guidance is available at FDA’s quality-agreements document.
Reassess after a major process, site, ownership, specification, analytical-method, subcontractor or upstream-material change; a significant deviation or recall; repeated delivery failures; or a prolonged period without supply. Verify that incoming material still comes through the approved supply chain, not merely from an approved company name.
Test whether the second source actually reduces correlated risk
Map the full dependency chain
For both incumbent and candidate sources, map API starting materials, key intermediates, critical reagents, manufacturing and testing sites, ownership and contract relationships, distributors, repackers, warehouses, transport lanes, ports, utilities and other dependencies relevant to the product. Ask where the two sources share a starting-material producer, a specialized laboratory, a port, a packaging component, a utility, a region exposed to the same hazard or a parent company’s allocation policy.
Compare failure scenarios, not just locations
Model realistic events: a quality-system shutdown, contamination, fire, cyber incident, export restriction, natural disaster, utility failure, transport blockage, starting-material shortage, laboratory loss or sudden demand allocation. Estimate capacity, lead time, safety stock, recovery time, substitution constraints and the time required to qualify or release material. Geographic separation can help, but it does not by itself prove independence.

FDA’s May 2022 Risk Management Plans to Mitigate Potential Drug Shortages offers a planning framework for identifying, prioritizing and mitigating hazards that can cause disruption. It is explicitly marked “Draft” and “Not for implementation,” so use it as a planning reference rather than binding regulation. The draft says, “Based on recent publications and reports, the majority of drug shortages are associated with quality issues.” It gives no percentage on the cited page; do not convert that statement into a numeric shortage statistic.

Confirm the regulatory path for the specific product
Do not infer a filing category, approval timeline or notification requirement from the fact that a second supplier is being added. Regulatory affairs must review the product’s marketing application, currently registered API source and site, process and specification commitments, proposed changes and destination jurisdictions. Determine whether the change requires a submission, prior approval, notification or another action under the applicable rules.

FDA’s Q7 guidance expressly says it is not intended to define registration or filing requirements and that commitments in registration or filing documents should be met. The final September 2016 guidance is at FDA’s Q7 page. For decisions outside the United States, verify the requirements of the relevant regulator; FDA and ICH materials do not automatically determine another jurisdiction’s filing obligations. The regulatory consequence cannot be resolved without the actual application, registered source and site, change details, commitments and markets.
A practical approval sequence
- Write the scope and risk question. Name the API, product, markets, sites, intermediaries and the disruption the second source should address.
- Collect a site-level information package. Obtain process, specifications, quality history, audit evidence, upstream-source information, capacity data and regulatory details.
- Perform a risk-based quality and technical assessment. Escalate to an audit or equivalent evaluation when the API’s risk, history or evidence warrants it.
- Complete comparability and analytical planning. Approve methods, sampling, identity testing, COA reliance, initial full testing and ongoing verification before routine supply.
- Approve the quality agreement. Assign responsibilities, change notification, subcontracting controls, records, escalation and audit rights.
- Resolve the product-specific regulatory path. Obtain the required internal and external approvals before commercial use.
- Run qualification batches and release them under the approved controls. Investigate every discrepancy and trend results against the incumbent where appropriate.
- Activate the ongoing scorecard and continuity plan. Review quality, service, dependency and change data at a defined cadence and trigger requalification when conditions change.
Common diversification mistakes
- Counting suppliers instead of independent paths: two vendors may share the same starting material, site or logistics bottleneck.
- Approving a distributor without identifying the manufacturer: the quality decision must cover the actual manufacturing and testing sites.
- Treating a COA as a substitute for qualification: identity testing and validation of supplier analytical reliability remain necessary when relying on supplier results.
- Using a quality agreement to shift accountability: contracts allocate activities but cannot delegate statutory or regulatory CGMP responsibilities.
- Reducing testing without documented evidence: the Q7 three-batch recommendation still requires a justified, approved verification program.
- Assuming a different country guarantees resilience: shared upstream materials, ownership, ports or utilities can create common-cause failure.
- Leaving regulatory review until after purchasing: a technically acceptable API may still require a filing action before commercial manufacture.
- Choosing on unit price alone: qualification, testing, inventory, freight, rejects, dual oversight and interruption costs belong in the landed-cost decision.
A second API source is worth approving when the site can demonstrate consistent quality, the material and process fit the product, responsibilities and changes are controlled, the regulatory path is clear, and the dependency analysis shows a meaningful reduction in correlated disruption risk. If any of those conditions remains unresolved, treat the source as an unqualified contingency—not as an operationally available supplier.
