Single-use and reusable tangential flow filtration (TFF) systems use the same filtration principle; the difference is what happens to product-contact components after a run. A single-use configuration discards specified wetted parts, while a reusable configuration retains them for additional cycles after cleaning, validation and storage. The right choice depends on process compatibility, batch and campaign size, changeover frequency, facility utilities, qualification work and total lifecycle cost—not on the TFF label alone.

What TFF does before the single-use-versus-reusable decision

In tangential flow filtration, feed moves parallel to a membrane surface. Pressure drives part of the fluid through the membrane as permeate, while retained molecules continue through the process stream. This differs from normal-flow filtration, where feed is pushed directly toward and through the membrane. TFF is used for concentration, buffer exchange (diafiltration) and related separations in bioprocessing.

“Single-use TFF” and “reusable TFF” describe the lifecycle of the product-contact hardware, not a different filtration mechanism. A single-use system might include only a disposable membrane capsule, or it might include the entire wetted flow path: tubing, reservoir or bag, pump head, pressure sensors and connectors. A reusable system generally uses cassettes, holders, tubing and vessels that are cleaned and prepared for another batch. Always define which surfaces are disposable before comparing systems.

Single-use and reusable TFF at a glance

Decision axis Single-use TFF Reusable TFF Questions to verify
Product-contact parts Specified wetted components are discarded after the run; coverage varies by configuration. Components remain in service for additional cycles. Which membranes, tubing, sensors, reservoirs and connectors actually contact product?
Preparation and changeover Can remove or reduce cleaning, cleaning validation and storage steps. Requires validated cleaning, inspection, storage and release between uses. Measure setup, teardown, cleaning, sampling and release time at your site.
Economics Recurring capsule, tubing and bag purchases; waste-disposal costs may rise. Equipment and membranes are spread over cycles, but labor, water, chemicals, utilities, storage and validation add cost. Use a site-specific total-cost model rather than a unit-price comparison.
Performance Can match a reusable format in a particular qualified application. May fit established equipment, recipes and automation. Compare flux, yield, product quality, residual clearance and membrane compatibility at target scale.
Utilities and materials Often reduces process cleaning-fluid demand; disposable materials create a recurring waste stream. Cleaning and flushing consume water and agents; fewer disposable parts are discarded. Quantify actual site utility use and waste treatment. No universal environmental winner is established.
Facility fit Can support multiproduct operation and rapid changeovers when the complete flow path is disposable. Can align with high-throughput campaigns and existing fixed infrastructure. Check pump capacity, piping, hold-up volume, automation, floor space and scale-up evidence.
Qualification and safety Requires material compatibility and extractables/leachables assessment for disposable wetted parts. Requires cleaning validation and controls against carryover and bioburden. Use product- and process-specific risk assessments and supplier documentation.

How a single-use TFF workflow operates

1. Define the disposable boundary

List every product-contact surface, not just the membrane. A capsule-only arrangement may still leave reusable tubing, a pump head or sensors in the flow path. A complete single-use assembly can include preassembled tubing, bags, pressure sensors and connectors. The boundary determines how much cleaning and changeover work is actually removed.

  • 12-Month Warranty: Backed by a 12-month manufacturer warranty for peace of mind. Designed to meet strict quality control standards for reliable everyday performance.

2. Qualify materials and the membrane

Confirm that membrane chemistry, support materials, tubing and bags tolerate the product, buffers, pressure, temperature and cleaning or sanitization exposure that still exists elsewhere in the system. Obtain extractables and leachables information for the selected disposable materials and assess whether the data cover your process conditions.

3. Install and inspect the assembly

Operators connect the disposable flow path according to the supplier’s instructions, verify orientation and sensor connections, inspect seals and tubing, and document lot numbers. Integrity testing, wetting and flushing requirements depend on the specific capsule and process.

4. Run concentration or diafiltration

Operate within qualified transmembrane-pressure, crossflow, temperature and feed limits. Track permeate flow, retentate flow, pressure, volume and product concentration. A disposable capsule is not automatically interchangeable with a reusable cassette: membrane area, channel geometry and hold-up volume can change operating behavior.

5. Recover product and dispose of the flow path

After product recovery and any required sampling, disconnect and dispose of the designated product-contact components under the site’s waste procedures. Retain batch records and component traceability. If any reusable hardware remains, it still needs the cleaning and storage controls applicable to that hardware.

How a reusable TFF workflow differs

Cleaning and inspection

Reusable cassettes, holders, tubing and vessels must be cleaned with a defined recipe that removes product, process residues and cleaning agents. Operators inspect for damage, blocked channels, gasket wear and other defects. Cleaning acceptance limits should be based on the products and residues handled, not on a generic recipe.

Validation and release

Cleaning validation demonstrates that the procedure consistently controls carryover, residues and bioburden. The site must define sampling locations, analytical methods, acceptance criteria, maximum dirty and clean hold times, and the conditions under which a component is released for another batch.

Storage and reuse tracking

Clean components need protected storage that preserves their condition and prevents recontamination. Track use count, cleaning history, inspection results and any retirement limits. A nominal reuse life is not proof that a component remains suitable after every cycle; the process must establish how performance and integrity are monitored.

Changeover execution

Reusable systems can require disassembly, cleaning connections, flushing, drying or sanitization, inspection and documentation before the next product. These activities may be straightforward in a dedicated campaign, but they become a scheduling constraint when products change frequently.

What the evidence says about performance

A peer-reviewed study in the Journal of Chemical Technology & Biotechnology (2022) compared a tested single-use capsule with reusable cassettes while purifying activated polysaccharides for conjugate vaccines. In that application, the study reported similar TFF process performance and similar post-purification product attributes. It also reported a smaller flush requirement for the capsule and no need for a compression holder or pre-sanitization in the tested setup.

Those are application-specific findings, not a universal equivalence claim. Molecule properties, membrane material, channel design, operating conditions, scale and recovery method can all change the result. Qualification should compare flux, throughput, yield, concentration factor, diafiltration efficiency, residual clearance, aggregate or impurity levels and final product quality at a representative scale.

Supplier documentation can provide useful operating ranges and risk information. For example, the Sartoflow 4500 product page describes a single-use flow path and states benefits including increased turnaround efficiency, reduced cleaning validation, and minimized bioburden risk and product carryover in a multiproduct facility. These are manufacturer claims for that system; they are not a guarantee for every process.

Turnaround, water and labor: where differences appear

Single-use equipment can shorten the interval between batches when the disposable boundary removes cleaning, cleaning validation and storage tasks. The saving is largest when campaigns are short, products change often or a facility lacks spare cleaning capacity. If only the membrane device is disposable while tubing and vessels remain reusable, the reduction may be modest.

Reusable systems shift work into cleaning, flushing, inspection, documentation and release. They may be efficient for long campaigns in which one setup serves many batches, especially when the facility already has validated cleaning skids, water-for-injection capacity and trained operators.

Merck/Sigma-Aldrich gives an illustrative manufacturer comparison in which a cited single-use process takes less than 2.5 hours versus 7 hours for a multi-use process. The same material gives an example of 20 litres per square metre of fluid use for its capsule versus 130 litres per square metre for multi-use cassettes. These figures describe the manufacturer’s examples and conditions; they are not industry averages or guaranteed results for another membrane area, molecule or facility.

How to model total cost of ownership

Compare the cost of a complete campaign, not the purchase price of a capsule or cassette. Build separate lines for materials, labor, utilities, qualification and waste, then test the model against realistic annual volumes.

Cost category Single-use items to include Reusable items to include
Materials Capsules, bags, tubing, connectors, sensors and packaging. Cassettes, holders, gaskets, replacement parts and any disposable subassemblies.
Labor Assembly, inspection, testing, setup and disposal. Disassembly, cleaning, sampling, inspection, drying, documentation and release.
Utilities Process buffers, limited flushing and waste treatment. Water, cleaning agents, compressed air, steam or other sanitization and drying utilities.
Quality and validation Supplier qualification, extractables/leachables assessment and incoming inspection. Cleaning-validation studies, analytical testing, change control and reuse tracking.
Facility and scheduling Storage for consumables and waste-handling capacity. Cleaning-skid capacity, controlled storage and time reserved for turnaround.
Waste and disposal Disposal, treatment and any special handling for contaminated plastics. Spent cleaning solutions, filters and parts at retirement.

Calculate cost per batch and per campaign, then vary annual runs, product changes, labor rates, utility prices, waste charges and realistic reuse cycles. A historical TangenX Technology Corp. model (2008) placed a break-even point at approximately 20 reuse cycles for a 10-square-metre operation. That threshold depended on the model’s assumptions and is not a current quote or a universal rule.

Choosing by process and facility conditions

Single-use is often favored when

  • Campaigns are short or products change frequently.
  • Fast changeover is more valuable than minimizing recurring consumables.
  • The site has limited cleaning capacity, water or validated storage space.
  • A complete disposable flow path can be qualified and reduces carryover risk.
  • Development, clinical or multiproduct work makes fixed infrastructure difficult to justify.

Reusable equipment is often favored when

  • The same product runs repeatedly in long campaigns.
  • Existing cassettes, holders, automation and cleaning utilities are already qualified.
  • Annual volume is high enough to spread hardware costs over many cycles.
  • Disposable material supply, waste handling or extractables data create unacceptable risk.
  • Process scale or flow rates exceed the practical capacity of available single-use assemblies.

These are screening conditions, not automatic decisions. A small multiproduct facility may save more with disposables even when each capsule costs more, while a high-volume dedicated line may recover reusable-equipment costs quickly.

A practical selection procedure

  1. Map the wetted path. Mark the membrane device, tubing, pump head, reservoir, sensors, connectors and any fixed piping as disposable or reusable.
  2. Set process requirements. Define molecule and membrane compatibility, operating-pressure range, target flux, yield, residual-clearance limits, product-quality attributes and acceptable hold-up volume.
  3. Describe the production pattern. Record batch size, annual batches, campaign length, product changes, peak throughput and a realistic number of reuse cycles.
  4. Measure the actual workflow. Time setup, integrity checks, cleaning, flushing, inspection, documentation, storage and waste handling at the intended facility.
  5. Build two lifecycle models. Include materials, labor, water, chemicals, utilities, validation, storage, waste and downtime. Run sensitivity cases for higher and lower volume.
  6. Check qualification evidence. Request membrane performance data, scale-up information, extractables/leachables documentation for disposable wetted materials, and cleaning-validation support for reusable parts.
  7. Run a representative comparison. Use the actual molecule or a justified surrogate at relevant concentration, pressure, temperature and membrane area. Compare yield, flux stability, quality and recovery.
  8. Document the decision and revisit it. Reassess when annual volume, product mix, facility utilities, supplier availability or campaign strategy changes.

Common mistakes and how to avoid them

  • Comparing only the membrane price. Add labor, utilities, validation, storage and waste to both options.
  • Assuming “single-use” means everything is disposable. Obtain a flow-path drawing and identify every wetted surface.
  • Generalizing one performance study. Treat the 2022 vaccine-purification result as evidence for its devices and process, then qualify your own application.
  • Using vendor examples as benchmarks. The 2.5-hour-versus-7-hour and 20-versus-130-litre-per-square-metre examples are illustrative, not industry averages.
  • Ignoring cleaning capacity. A reusable system can become the schedule bottleneck if cleaning, sampling or release takes longer than production.
  • Overlooking materials risks. Disposable assemblies need extractables/leachables assessment; reusable assemblies need carryover and bioburden controls.
  • Choosing a reuse count from a brochure. Establish cycle limits using performance, integrity and cleaning data from the actual process.

Bottom line

Single-use TFF trades recurring disposable consumption for simpler changeovers and less cleaning work when the disposable boundary is broad enough to matter. Reusable TFF trades cleaning, validation and storage effort for the ability to spread equipment and membrane costs across repeated cycles. Neither format is universally superior. Select the configuration that meets your product and scale requirements after a process-specific performance qualification and a total-cost model based on your facility’s real campaign pattern.
