RNA editing is a chemical change made to an RNA molecule after it has been copied from DNA. In the form best documented in octopuses, squid, and cuttlefish, ADAR enzymes convert adenosine (A) to inosine (I). During translation, cells usually read inosine as if it were guanosine (G), so an edited codon can specify a different amino acid without any change to the animal’s DNA.
What makes coleoid cephalopods unusual is not simply that they edit more RNA overall. Their distinctive feature is the extensive use of editing to recode protein-coding messages, especially in neural tissue. Reviews published in 2023 summarize recoding across several octopus, squid, and cuttlefish lineages, while noting that the evolutionary cause and the benefits of most individual edits remain unresolved.
RNA editing changes the message, not the genome
DNA is the relatively stable archive of genetic information. When a cell needs to make a protein, it transcribes the relevant DNA sequence into messenger RNA (mRNA). RNA editing acts on that transcript after transcription. It can alter the RNA sequence that a ribosome reads, while leaving the DNA sequence from which the transcript was copied unchanged.

- An ADAR enzyme recognizes a suitable adenosine in an RNA molecule.
- The enzyme chemically converts adenosine (A) into inosine (I).
- The translation machinery generally interprets inosine as guanosine (G).
- If the edited base is in a protein-coding codon, the codon may now specify a different amino acid.
This creates a temporary, regulated way to produce a protein variant. It is not a mutation inherited by changing the genome, and it is not an octopus “rewriting” its DNA.
Three kinds of editing sites matter
Not every edit has the same consequence. An edit in a noncoding region can influence RNA structure, processing, localization, stability, or regulation without changing a protein sequence. A coding edit can be synonymous, meaning the amino acid remains the same, or recoding, meaning the amino acid changes. The headline cephalopod finding concerns the abundance of recoding events, particularly in neural transcripts.
Across animals, noncoding editing is more common overall. This is why “cephalopods have more RNA editing” is too imprecise. The more accurate statement is that coleoids have an unusual propensity to use editing to recode mRNAs.
Why the coleoid pattern is unusual
Octopuses, squid, and cuttlefish belong to the coleoid cephalopods. Comparative work summarized by Joshua J. C. Rosenthal and Eli Eisenberg in their 2023 Annual Review of Animal Biosciences article describes orders-of-magnitude more protein-recoding sites in coleoids than in the other taxa included in its comparisons. The same review emphasizes that overall editing levels are not exceptionally high; the unusual property is how often editing changes protein-coding messages.

The pattern appears in multiple coleoid lineages rather than in one exceptional species. Studies discussed in the 2023 reviews include Octopus bimaculoides, Octopus vulgaris, the cuttlefish Sepia officinalis, Hawaiian bobtail squid (Euprymna scolopes), striped pajama squid (Sepioloidea lineolata), and other squid. In the comparisons summarized, extensive recoding was not observed in the nautilus Nautilus pompilius. That contrast suggests an innovation associated with the coleoid branch, but it does not by itself identify the evolutionary cause.
How extensive is neural recoding?
A 2023 review in Briefings in Functional Genomics reports that close to 60% of squid, cuttlefish, and octopus brain transcripts contain at least one recoding site. This is a review-level estimate drawn from the studies it cites. It is not a percentage of all RNA molecules, not a universal value for every species, and not a claim that 60% of every transcript is edited.
The tissue distribution is also important. The 2023 Annual Review of Animal Biosciences review summarizes measurements in which recoding activity in nonneural tissues of Octopus bimaculoides was three to six times lower than in neural tissues. Neural enrichment is therefore a central part of the cephalopod pattern, although the exact ratio should not be generalized to every cephalopod or tissue.

What happens inside an edited neuron?
Neurons depend on proteins that control electrical excitability and communication. The cephalopod reviews discuss recoding in transcripts for ion channels, neurotransmitter receptors, and components of synaptic signaling. Changing an amino acid in one of these proteins could alter properties such as channel behavior, receptor responses, or interactions among signaling proteins.

Editing can also be spatially regulated. Research on squid neurons has examined editing within axons, showing that RNA processing need not be confined to the cell body. The practical implication is that a neuron may regulate some transcripts near the location where their protein products are needed. The existence of spatial control does not mean every edited site has a known function; it demonstrates that editing can be organized within a cell.
A useful comparison of the evidence
| Comparison | What the evidence shows | How to interpret it |
|---|---|---|
| Total editing versus recoding | Cephalopods are not reported to have unusually high overall editing; their recoding propensity is unusual. | Count protein-changing edits separately from all edits. |
| Neural versus nonneural tissue | In the summarized O. bimaculoides measurements, nonneural recoding activity was three- to sixfold lower than neural activity. | The comparison is species- and study-specific, not a universal cephalopod ratio. |
| Coleoids versus nautilus | Extensive recoding is documented in octopuses, squid, and cuttlefish; it was not observed in N. pompilius in the cited comparisons. | The pattern points to coleoid evolutionary history without proving one cause. |
| Neural transcript prevalence | Close to 60% of squid, cuttlefish, and octopus brain transcripts were reported to contain at least one recoding site. | This is a review-summarized estimate with defined taxa and tissue scope. |
| Environmental response | Many measured octopus recoding sites vary with temperature. | Observed temperature sensitivity supports a possible acclimation role; adaptive benefit is not established for every site. |
Why might cephalopods use so much recoding?
The leading functional idea is transcriptome plasticity: editing lets a cell adjust selected protein sequences without waiting for a DNA mutation and its spread through a population. A single gene can therefore yield related protein variants under different cellular or environmental conditions. This is especially intriguing in a nervous system that must tune channels, receptors, and synaptic machinery.
That idea is plausible, not a complete explanation. A high editing level can reflect biochemical constraints, historical evolution, or a mixture of beneficial and neutral effects. The fact that an edit is conserved or common does not automatically prove that it improves survival. Some edits may have small effects, operate only in particular tissues, or be tolerated because the surrounding RNA structure permits them.
Temperature provides the strongest direct test so far
A 2023 PNAS study titled “Temperature-dependent RNA editing in octopus extensively recodes the neural proteome” experimentally documented temperature-related editing in octopus. The reviews summarize many recoding sites as temperature-sensitive, making acclimation a credible role: as body temperature changes, editing could adjust neural proteins without altering the genome.

Temperature sensitivity should still be stated precisely. It shows that editing changes with temperature under the tested conditions. It does not show that RNA editing is the only mechanism of thermal adaptation, that every temperature-responsive site is useful, or that the process explains the behavioral sophistication of octopuses.
Other proposed influences remain hypotheses
Researchers have discussed oxygen availability, light, pH, and social conditions as possible influences on editing. These are research directions rather than established general explanations. The most important open question is whether the many recoding events have distinct physiological functions or whether a smaller subset produces most of the measurable benefit.
Does RNA editing explain octopus intelligence?
No such conclusion is established. Coleoids have complex nervous systems and extensive neural recoding, but correlation does not demonstrate that editing caused intelligence, learning ability, camouflage, problem solving, or any particular behavior.
RNA editing could contribute to neural flexibility by modifying proteins involved in electrical signaling. That is a biologically reasonable hypothesis, and the concentration of recoding in neural transcripts makes it worth testing. However, nervous-system complexity also depends on anatomy, development, gene regulation, synaptic organization, behavior, and ecology. Current reviews treat the relationship between recoding and complex cephalopod behavior as an open problem, not a solved explanation.

How scientists distinguish real recoding from a misleading percentage
Claims about “RNA editing levels” can refer to different measurements. A careful reading asks four questions:
- What is being counted? All edited positions, only coding edits, or only edits that change an amino acid?
- Which tissue was sampled? Brain, another neural structure, muscle, or a mixed specimen can produce different results.
- Which species and conditions were tested? A result from one octopus species at one temperature is not a universal cephalopod value.
- What does the measurement show? A change in editing frequency demonstrates molecular response; it does not by itself demonstrate a beneficial adaptation.
The often-quoted comparison that 11–13% of global editing activity in the cited cephalopod neural measurements was recoding activity, versus less than 1% in mammals, is a review-reported comparison of particular underlying measurements. It should not be treated as a constant for all cephalopods, all mammals, or all tissues.
What remains unknown
Why are coleoid transcripts so prone to recoding?
The biochemical basis is unresolved. The reviews identify possible contributions from ADAR enzyme structure, associated proteins, and the shapes formed by editable RNA sequences. RNA must fold into suitable structures for ADAR recognition, so changes in transcript sequences and secondary structure could influence which sites are editable. No single explanation has been established as sufficient.

Some recoding events are likely consequential, particularly when they alter neural proteins, but the function of many individual sites remains uncertain. Establishing a role requires more than detecting an A-to-I change: researchers need to connect the edited protein form to a measurable effect in a cell, neuron, organism, or environmental condition.
Why did extensive recoding evolve in coleoids?
Comparative studies show a striking distribution across octopuses, squid, and cuttlefish and a different pattern in nautilus. They do not yet reveal whether the shift was driven mainly by ecological pressures, neural demands, genome evolution, molecular constraints, or several factors acting together. A 2017 Science study framed this issue as a trade-off between transcriptome plasticity and genome evolution: reliance on conserved editable sites may provide flexibility while also imposing constraints on DNA changes that would disrupt the required RNA structures.
Common misconceptions, corrected
- “Octopuses edit their DNA.” The documented process changes RNA after transcription; the underlying DNA is not rewritten.
- “Every RNA edit changes a protein.” Many edits are noncoding, and some coding edits are synonymous. Recoding is the protein-changing subset.
- “All cephalopods edit RNA at the same rate.” The strongest pattern is associated with coleoids, and nautilus provides an important contrasting lineage.
- “A 60% figure means 60% of all octopus RNA is edited.” The figure concerns brain transcripts containing at least one recoding site in the review’s summarized studies.
- “Temperature-responsive editing proves adaptation.” It supports a possible acclimation function, but adaptive value must be demonstrated site by site or system by system.
- “RNA editing is the reason octopuses are intelligent.” Neural recoding may be relevant to neuronal flexibility, but causation has not been established.
What a careful answer should say
RNA editing is a post-transcriptional way to diversify cellular messages. In A-to-I editing, ADAR enzymes change selected adenosines to inosines, which are generally read as guanosines. Coleoid cephalopods stand out because they use this mechanism extensively to recode neural protein transcripts, across several octopus, squid, and cuttlefish lineages. The reviewed evidence includes a close-to-60% brain-transcript estimate, strong neural enrichment, and temperature-dependent editing in octopus.
The central scientific question is still open: why did coleoids evolve such a strong bias toward protein recoding, and what do the many individual edits do? Temperature response makes acclimation a well-supported possibility, while claims about intelligence or universal adaptation go beyond the evidence.
