Regioselective hydroformylation is the catalytic conversion of an alkene, carbon monoxide (CO) and hydrogen (H2) into an aldehyde while favoring one constitutional isomer over another. For an unsymmetrical alkene, the catalyst can favor the linear (normal, or n) aldehyde or the branched (iso, or i) aldehyde. The linear-to-branched ratio, written L:B or n:i, is the usual practical measure of that preference.

The reaction matters because the position of the aldehyde group determines what can be made next. Industrial processes often seek linear aldehydes for downstream plasticizer, detergent and chemical manufacture, whereas synthetic chemists may need a branched, stereodefined or enantioenriched aldehyde for a particular target molecule.

What hydroformylation does

Hydroformylation, also called the oxo process, adds the elements of a formyl group (–CHO) and hydrogen across an alkene C=C bond. In simplified form, an alkene reacts with CO and H2 to produce an aldehyde:

alkene + CO + H2 → aldehyde

Because an unsymmetrical alkene has two different carbon atoms in its double bond, the formyl group can end up at either carbon. Those two pathways produce constitutional isomers rather than merely different conformations. Catalyst structure and reaction conditions are used to make one pathway faster, more stable or more readily completed than the other.

Terminal-alkene example

For a terminal 1-alkene written as R–CH=CH2, the linear product is the normal aldehyde R–CH2–CH2–CHO. The branched product is the iso aldehyde R–CH(CH3)–CHO. “Linear” and “branched” describe the carbon skeleton around the new aldehyde; they do not describe whether the product is optically active.

Why regioselectivity matters

Hydroformylation is an atom-economical C–C-bond-forming reaction: the carbon monoxide supplies the aldehyde carbon, and hydrogen supplies the added hydrogen atoms. Bernhard Breit described it in 2003 as “an ideal atom economic C/C-bond forming reaction.” The process is industrially important for producing aliphatic aldehydes and intermediates used in plasticizers, detergents, fine chemicals, pharmaceuticals and agrochemicals.

Selectivity has practical consequences at every stage:

  • Separation: a higher proportion of the desired isomer reduces the amount of aldehyde mixture that must be separated.
  • Process design: the preferred regioisomer can have different boiling points, reactivity and purification requirements.
  • Downstream chemistry: oxidation, hydrogenation, condensation and other reactions preserve or exploit the aldehyde’s position.
  • Synthetic planning: a branched aldehyde may create a useful stereocenter, while a linear aldehyde may provide a more accessible chain for further functionalization.

There is no universal L:B percentage. The observed ratio depends on the alkene, catalyst, ligand, solvent, gas pressures, temperature, support or confinement and the rates of several competing elementary steps.

How the catalytic cycle works

The following cycle is the commonly discussed homogeneous rhodium model. Other metals, supports and reactor environments can alter individual steps, but the sequence explains how CO and H2 become an aldehyde and where regioisomeric pathways arise.

1. Formation of an unsaturated metal hydride

The active species is a coordinatively unsaturated Rh(I) hydride. “Unsaturated” means that a coordination site is available for the alkene. The ligand set around rhodium controls how open that site is, how the alkene approaches and which transition state is favored.

2. Alkene coordination and insertion

The alkene binds to rhodium and inserts into the Rh–H bond. Insertion can occur in either orientation. One orientation gives a linear Rh–alkyl intermediate; the other gives a branched Rh–alkyl intermediate. This is the first obvious branching point in the mechanism and often a major source of regioselectivity.

The two pathways are not necessarily locked at this stage. Alkyl intermediates can interconvert, and their relative rates of carbon monoxide insertion or other subsequent reactions can change the final product ratio. Consequently, the measured L:B value is a kinetic outcome of the entire catalytic network, not a direct readout of one single bond-forming event.

3. Carbon monoxide insertion

CO coordinates to the metal and inserts into the Rh–alkyl bond. The result is an acyl complex. This step creates the metal-bound carbonyl functionality that will become the aldehyde carbon.

4. Hydrogen activation

H2 undergoes oxidative addition at the metal center. The rhodium complex now contains the hydrogen atoms needed to convert the acyl group into an aldehyde and regenerate the hydride chemistry.

5. Aldehyde release and hydride regeneration

Reductive elimination releases the aldehyde and regenerates a rhodium hydride species. The catalyst can then bind another alkene. Product release, gas availability and the stability of each intermediate all affect the turnover rate and the distribution of products.

Where the linear-versus-branched choice is made

Regioselectivity is best understood as a competition among pathways rather than as a property of the metal alone. The 2026 ACS Catalysis review by Lei Wang, Shihang Zhao and Dehui Wang identifies several design levers, especially for heterogeneous systems.

Ligand sterics and electronics

Ligands determine the size, shape, electron density and flexibility of the catalyst pocket. Bulky groups can block one alkene orientation, while electronic effects change the strength of alkene, CO and hydride binding. A ligand that improves L:B for one substrate can reduce activity or favor the opposite isomer for another.

Diphosphine bite angle and catalyst geometry

For a chelating diphosphine, the bite angle changes the geometry available around rhodium and can alter the relative energies of linear and branched alkyl transition states. Monophosphines provide a different coordination environment from diphosphines, so “more phosphorus” is not itself a selectivity rule. The relevant question is how the complete ligand arrangement shapes the reactive pocket.

Substrate substitution and alkene class

Terminal and internal alkenes present different steric and conformational choices. Substituents near the double bond can favor one insertion orientation, slow alkene binding or make secondary reactions more competitive. Functional groups can also coordinate to the catalyst or change solubility, so selectivity established with a simple 1-alkene should not be assumed for a multifunctional substrate.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Solvent and gas composition

Solvent affects catalyst solvation, ligand behavior, mass transfer and the stability of charged or polar intermediates. CO and H2 pressures influence the concentrations of hydride, alkene, carbonyl and acyl species. Changing the CO:H2 balance can therefore change both conversion and L:B, even when the catalyst formulation is unchanged.

Support, confinement and defects

In supported or heterogeneous catalysts, the surface can impose a different geometry from a freely diffusing homogeneous complex. Pore size, confinement, neighboring sites and surface defects can favor one alkene orientation or alter the lifetime of intermediates. The 2026 review emphasizes geometric, electronic, synergistic and defect effects, together with ligand and confinement effects, as independent design considerations. It also notes that controlling regioselectivity in heterogeneous hydroformylation remains a persistent challenge.

Normal (linear) versus iso (branched) hydroformylation

Feature Normal or linear pathway Iso or branched pathway

Typical reason to target it Industrial aldehyde streams and linear downstream products Branched building blocks, target-oriented synthesis or stereochemical design

The L:B ratio should always be reported with the substrate and catalyst system. A ratio measured for a terminal alkene under one set of conditions does not establish the ratio for an internal alkene, a different ligand or a supported catalyst.

Homogeneous and heterogeneous catalyst approaches

In a homogeneous process, the metal complex and substrate are in the same phase, commonly a solution. This makes ligand changes and mechanistic studies comparatively direct, and the rhodium hydride cycle is the standard teaching model. The trade-off is that separating dissolved metal and ligand from the aldehyde product can be difficult.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Heterogeneous or supported catalysts immobilize the active metal complex, place it in a porous environment or use a surface-bound active site. Potential advantages include catalyst recovery and a different local environment for directing the alkene. However, diffusion, nonuniform sites, metal leaching and poorly defined active structures can complicate both activity and selectivity. Support effects are therefore part of the catalyst design, not an afterthought.

Cobalt and rhodium represent important metal choices, but the metal label alone does not predict the result. A meaningful comparison records the metal and phase together with ligand environment, substrate, gas pressures, temperature, conversion, aldehyde selectivity, L:B ratio and catalyst lifetime.

How to compare a catalyst system

When evaluating published data or planning an experiment, use the same checklist for every candidate:

  1. Identify the substrate: record whether it is terminal or internal, its substitution pattern and any coordinating functional groups.
  2. Record the active phase: distinguish a dissolved complex from a supported, confined or otherwise heterogeneous catalyst.
  3. Describe the ligand environment: note monophosphine versus diphosphine, steric bulk, electronic character and diphosphine bite angle when reported.
  4. Separate outcomes: report alkene conversion, total aldehyde selectivity and L:B independently. High conversion does not automatically mean high regioselectivity.
  5. Include operating conditions: capture temperature, CO and H2 pressures, solvent, residence time and catalyst loading because each can change the observed ratio.
  6. Check durability and recovery: for a practical process, catalyst lifetime, metal recovery and resistance to deactivation matter alongside the first-batch L:B value.
  7. Test the intended substrate: do not extrapolate from a model alkene without checking the actual functional-group and steric environment.

Regioselectivity is not stereoselectivity

Regioselectivity answers where the aldehyde group is placed: linear or branched. Stereoselectivity answers which three-dimensional arrangement is formed, and enantioselectivity asks whether one enantiomer is favored over its mirror image. A catalyst can be highly regioselective but produce a racemic branched aldehyde, or it can be designed to address regio- and enantioselectivity at the same time.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Enantioselective and substrate-directed hydroformylation are valuable for target-oriented synthesis, but they face a coupled challenge: the catalyst must control activity, regioselectivity and stereochemical outcome simultaneously. Improving one metric can compromise another, so all relevant selectivities must be reported rather than summarized by a single percentage.

Examples and evidence boundaries

Supramolecular catalyst studies illustrate how confinement can change regioisomer formation. A 2018 review reported outer-to-inner product ratios of 1:9 for trans-2-octene and 1:4 for trans-3-octene with an encapsulated rhodium complex. These are system-specific ratios for that encapsulated catalyst and those internal alkenes; they are not universal L:B benchmarks and should not be transferred to ordinary solution catalysts without evidence.

Hydroformylation has been described as “one of the most intensively explored reactions in the field of homogeneous transition metal catalysis,” a statement from a van Leeuwen and Kamer review reproduced in a 2018 supramolecular-catalysis article. The depth of the literature explains why apparently small changes in ligand, support or substrate can produce different conclusions: many mechanistic and engineering variables are coupled.

Common interpretation and troubleshooting mistakes

Treating one L:B value as universal

Problem: a reported ratio is quoted without its substrate, catalyst or conditions. Correction: carry the full experimental context with the number and compare only genuinely similar systems.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Assuming insertion alone fixes the product

Problem: the first alkene-insertion drawing is treated as the complete explanation. Correction: consider alkyl-intermediate equilibration, CO insertion, hydrogenolysis and product release; differences in later rates can change the final ratio.

Confusing conversion with aldehyde selectivity

Problem: high alkene conversion is interpreted as clean hydroformylation. Correction: measure unreacted alkene, aldehydes and side products separately, then report conversion and selectivities as distinct values.

Ignoring phase and recovery

Problem: a homogeneous result is compared with a supported catalyst using only L:B. Correction: include catalyst lifetime, metal recovery, mass-transfer behavior and possible leaching when judging process suitability.

Calling a branched product “enantioselective”

Problem: the presence of a branched aldehyde is taken as proof of stereochemical control. Correction: determine regioisomer distribution and enantiomeric composition independently.

Why the field remains active

Hydroformylation combines a simple overall equation with a demanding selectivity problem. Industrial users want reliable activity, long catalyst life, easy metal recovery and a predictable aldehyde stream. Synthetic chemists may instead prioritize a branched or enantioenriched product, functional-group tolerance and access to a specific molecular architecture.

Current catalyst design therefore moves beyond choosing a metal in isolation. Researchers tune ligand geometry and electronics, exploit cooperative or synergistic sites, engineer defects and pore environments, and use confinement to control how the alkene approaches the active center. The central practical lesson is that regioselectivity is an emergent property of the substrate, catalyst architecture and operating environment.

Key takeaways

  • Hydroformylation adds –CHO and H across an alkene using CO and H2.
  • Regioselective hydroformylation favors either the linear (normal) or branched (iso) aldehyde.
  • The L:B or n:i ratio is useful only when reported with substrate and operating conditions.
  • In the homogeneous rhodium model, alkene insertion creates linear and branched Rh–alkyl pathways, but later intermediate behavior also affects the final ratio.
  • Ligand sterics and electronics, diphosphine bite angle, catalyst geometry, substrate structure, solvent, gas pressures, support and confinement can all change selectivity.
  • Regioselectivity, stereoselectivity and enantioselectivity are separate measurements.
  • No single metal, ligand or percentage is optimal for every alkene and process.
