A synthetic cell is an engineered, cell-like system that reproduces selected features of living cells. It may contain a membrane, genetic instructions, molecular machines, energy-producing reactions or sensing circuits, but it is not automatically a living organism. Scientists use the term for several overlapping designs—including protocells, artificial cells, cell mimics, artificial organelles and minimal cells—and judge each system by the functions it actually demonstrates.
What a synthetic cell is—and what it is not
A natural cell is an integrated system. It keeps a boundary around its chemistry, takes in resources, transforms energy, stores and uses information, makes molecules, responds to conditions and, in many cases, grows and divides. A synthetic cell imitates some of those jobs in an engineered compartment.
The word “synthetic” does not necessarily mean that every atom was manufactured from simple chemicals. Some projects assemble purified molecules; others use cell extracts, natural molecular machines or a living cell whose genome has been reduced. The shared goal is control: researchers want to build, simplify or reconfigure cellular functions so they can observe how the parts work together.
That is why a liposome filled with a fluorescent dye is not automatically a synthetic cell, and why a cell-free reaction in a test tube is not automatically one either. A credible claim identifies the compartment, the internal information or reaction system, the energy source and the life-like functions that were measured.

Are synthetic cells alive?
Most current systems are not considered fully living cells. They can perform individual life-like activities—such as gene expression, metabolism, sensing, selective exchange or division—without combining all of those activities into a self-maintaining, autonomous organism.
“Alive” is not a single laboratory test. Biologists commonly look for coordinated properties including a boundary, energy use, information processing, self-maintenance, adaptation and reproduction. A constructed system may satisfy some criteria while failing others. For example, a vesicle might express a gene but lack a way to repair its membrane; another might divide physically without copying an information system into both daughters.
The National Academies’ 2025 discussion of synthetic-cell research emphasizes that these systems have not been shown to function in open, real-world environments. Demonstrations remain dependent on controlled laboratory conditions, supplied nutrients or energy, and carefully selected components.

Projects are often grouped by where the starting material comes from. The boundaries overlap, but the three routes clarify what has actually been engineered.
Bottom-up: assemble a system from molecular parts
Bottom-up construction starts with non-living components and combines them into a compartment that develops cell-like behavior. Researchers may choose lipids, polymers, peptides, nucleic acids, purified enzymes, ribosomes, messenger RNA, DNA templates and energy donors. The components can organize themselves through chemical self-assembly or be arranged with physical supports and interfaces.
This route gives unusually fine control over composition. A team can omit a pathway, alter one reaction, or add a sensor without carrying along thousands of unrelated cellular components. It also exposes a major difficulty: a natural cell contains many interacting processes whose timing, concentrations and feedback loops have evolved together. Recreating one pathway is much easier than making all pathways support one another.
Top-down: simplify a natural organism
Top-down work begins with a living cell. Scientists remove genes, refactor a genome or otherwise reduce the organism to a minimal set of functions needed for survival under defined conditions. The resulting cell is genetically engineered, but it is not assembled from scratch: it retains a natural membrane, molecular machinery and evolutionary history.

Top-down systems are useful for asking how much of a genome is essential and for creating organisms with fewer competing pathways. Their dependence on a living cellular framework is also their limitation. A reduced genome does not reveal how to build every cellular component from isolated molecules, and the cell may still rely on natural processes that have not been redesigned.
Middle-out: combine natural modules with synthetic parts
Middle-out designs mix the two strategies. A researcher might place a natural cell extract or purified translation machinery inside a synthetic vesicle, then add an engineered membrane, energy module or genetic circuit. The natural module supplies complex functions that would be difficult to rebuild, while the synthetic parts provide control over the surrounding architecture.
This approach can produce useful demonstrations faster than a completely bottom-up design. It also makes interpretation important: a system that uses a cell extract may be highly engineered while still depending on many natural proteins and metabolites.
How scientists construct a synthetic cell
A typical project proceeds through a series of design and measurement decisions rather than one recipe. The exact order varies, but the following stages show what has to be solved.
The Tool Desk
Outbyte Driver Updater FREEScan for outdated or missing drivers - takes under a minuteDriver Scan →Outbyte PC Repair FREEClear out junk files and repair common Windows errorsFree Scan →1. Build a boundary
The boundary separates an internal chemical environment from the outside. Lipid membranes are common because they resemble biological membranes and can allow selective transport. Polymers, peptides, nucleic-acid structures and hybrid materials can provide different durability, permeability or assembly behavior.
In self-assembly, the ingredients organize spontaneously because of their chemical properties. In templated assembly, a support, interface, microfluidic device or other physical constraint helps shape the compartment. Researchers must control size, membrane composition, leakage, mechanical stability and the routes by which nutrients, waste and signals cross the boundary.
Compartmentalization matters for more than appearance. It concentrates enzymes and substrates, keeps incompatible reactions apart, permits gradients and creates an inside-versus-outside relationship. Without a boundary, a reaction mixture may show biology but lacks one of the defining organizational features of a cell.

2. Add an internal reaction space
The compartment can receive purified proteins and enzymes, ribosomes, nucleic acids, cell extracts or a complete cell-free transcription-and-translation system. A DNA template inside the vesicle can be transcribed into RNA and translated into protein when the mixture contains the required enzymes, ribosomes, nucleotides, amino acids and an energy donor.
Researchers select the internal chemistry according to the question they want to test. A minimal enzyme set may reveal how a metabolic pathway behaves in confinement. A richer extract can support gene expression but introduces many unknown interactions. Either way, the team must provide enough energy and raw materials while preventing waste products or pH changes from stopping the reactions.
3. Add specific cell-like functions
Functions are usually added as modules and tested individually before integration. Common modules include:
- Sensing: receptors or molecular circuits detect a chemical, light or other environmental change.
- Selective exchange: pores, transporters or responsive membranes regulate what enters and leaves.
- Gene expression: DNA or RNA instructions are copied and translated into proteins.
- Metabolism: linked reactions convert supplied nutrients into useful molecules or energy.
- Energy management: chemical fuels or light-driven systems maintain an energy currency for internal work.
- Information processing: genetic circuits or reaction networks switch between states and produce outputs.
- Movement: motors, shape changes or chemical gradients drive motion.
- Growth and division: material is added to a compartment and the boundary is remodeled into daughter compartments.
Demonstrating one item does not establish the others. A system may express a protein but have no metabolism; it may divide mechanically without duplicating its information; or it may sense a signal without maintaining itself afterward.
4. Control conditions and measure behavior
Microfluidic devices can produce uniform compartments, exchange solutions and expose many individual systems to the same conditions. Microscopy tracks membrane shape, cargo location, movement and division. Biochemical assays measure RNA, protein, metabolite, energy or leakage levels. Researchers also vary nutrient supply, temperature, pH and osmotic conditions to determine whether a behavior is robust or occurs only in a narrow setup.

A convincing result connects a measured output to a defined component. Controls might omit the DNA template, disable an enzyme, alter membrane permeability or remove the energy donor. Those comparisons show whether the observed signal comes from the proposed synthetic-cell mechanism rather than from a free reaction outside the compartment.
What counts as a protocell versus a synthetic cell?
“Protocell” usually refers to a model of an early, prebiotic or primitive cell. Protocell research often asks how compartments, metabolism and information could have emerged before modern biology. A protocell may be very simple and need not reproduce the architecture of a contemporary cell.

“Synthetic cell” is broader. It can include a protocell, but it also covers engineered artificial organelles, cell mimics, minimal cells and systems built to perform a practical task rather than to model the origin of life. In everyday scientific usage, the terms overlap, so the useful question is not which label appears in a paper but which functions and materials are present.
| Comparison | Protocell | Synthetic cell |
|---|---|---|
| Typical purpose | Explore early-cell or origins-of-life chemistry | Reproduce, study or use selected cellular functions |
| Starting materials | Often simple or prebiotic chemistry in a compartment | May use purified parts, cell extracts, engineered modules or a reduced organism |
| Complexity | Can be deliberately primitive | Ranges from a single artificial organelle to a multi-module cell-like system |
| Relationship | One category within the wider cell-like-systems field | Umbrella term with overlapping categories |
How to compare synthetic-cell demonstrations
Headlines can make unlike systems sound equivalent. Use the following comparison axes when reading a paper, press release or demonstration.

This framework prevents a membrane-only vesicle from being compared directly with a genetically reduced organism. It also distinguishes a one-time reaction from a system that maintains itself through repeated cycles.
Independent reader supportYour contribution helps us test, update, and keep practical guides available for everyone. Why researchers build synthetic cells
Understand how cells work
Reconstituting a process from selected parts can reveal which components are necessary, how reactions are coupled and how spatial organization changes behavior. A compartment lets scientists test questions that are difficult to isolate in a full organism, where thousands of pathways operate simultaneously.
Study the origins of life
Protocell experiments explore how a boundary, metabolism and information might have become linked. Researchers can test whether simple chemistry supports sustained reactions, selective exchange or primitive reproduction under plausible conditions. These experiments do not recreate the historical first cell; they evaluate mechanisms that could have contributed to early cellular organization.
Create miniature bioreactors
Encapsulated enzymes and cell-free pathways can make proteins, metabolites or other materials in a small reaction volume. Separating reactions into compartments may improve control, protect unstable molecules or allow several chemistries to operate side by side.
Deliver therapeutic molecules
Artificial compartments can be designed to carry a payload, respond to a local signal or release contents under a chosen condition. A delivery system that mimics one cellular function is not the same as a living therapeutic cell, so safety, stability, targeting and clearance remain separate engineering problems.

Sense chemicals or pathogens
A synthetic compartment can contain a molecular sensor that produces a measurable output when it encounters a target. Because the sensor can be assembled without maintaining a whole organism, it may be useful in controlled diagnostic or environmental formats.
Support biomanufacturing
Cell-free or cell-mimicking systems can produce proteins and other molecules without the growth requirements of a complete organism. Their practical value depends on reaction lifetime, energy costs, yield, containment and the ability to scale production.
The hardest unsolved integration problems
The central challenge is coordination. A natural cell continuously balances membrane repair, nutrient transport, energy generation, information processing, waste removal, growth and division. In an engineered system, each function may work in isolation but fail when combined.
Do these 3 things before closing this tab:
1Fix the driver behind crashes, sound loss and screen glitches2Repair Windows errors before they cause bigger problems3Scan for outdated or missing drivers - takes under a minute- Compartmentalization versus exchange: the membrane must retain useful molecules while admitting resources and exporting waste.
- Energy supply: internal reactions consume fuels, and the energy system must be replenished without disrupting the membrane or chemistry.
- Metabolic balance: pathways need compatible rates, concentrations and by-product handling.
- Information control: genetic instructions must be copied and expressed reliably under changing conditions.
- Growth and division: the boundary, contents and information system must be distributed to descendants in a coordinated way.
- Robustness: a system that works for minutes under ideal laboratory conditions may fail when temperature, dilution, nutrients or contaminants change.
These constraints explain why a fully synthetic, autonomous organism remains a research objective rather than an established technology.
How close are scientists to creating life in the lab?
Scientists can already build compartments with selected biological functions, and top-down work can reduce living organisms to compact genomes. However, no demonstrated system combines the full package of autonomous maintenance, open-environment operation, reliable information replication, metabolism, growth and division in a generally accepted artificial cell.
The most accurate description is therefore “increasingly capable cell-like systems,” not “life created from scratch.” Progress is measured module by module: a better membrane, longer-lasting gene expression, a more efficient energy cycle, a controllable sensor or a more reliable division process. The scientific milestone will be integration, when those modules support one another over repeated generations without continual laboratory rescue.
A 2023 Nature Reviews Molecular Cell Biology viewpoint, published as the version of record for the 2024 volume, captured the motivation: “Scientists are captivated by the prospect of creating a fully synthetic cell, offering the potential to revolutionize biology, medicine and biotechnology.” The aspiration is clear, but the quotation describes a prospect, not a completed artifact.
How to read a synthetic-cell claim carefully
- Look for the exact boundary material and how the compartments were formed.
- Check whether the internal system is purified, cell-free, extract-based or derived from a living organism.
- Separate a function that was measured from one that was merely proposed.
- Ask how energy and nutrients were supplied, and for how long activity continued.
- Verify whether “division” means membrane splitting, replication of genetic information, or both.
- Look for controls that show the signal came from the compartment and its designed components.
- Note the containment conditions before inferring real-world readiness.
The National Academies describes bottom-up construction as methods that “assemble non-living molecular components into cell-like systems exhibiting emergent properties.” That definition highlights both the ambition and the qualification: the system can display new behavior that emerges from its parts while still falling short of a self-sustaining living cell.
