Gene-editing systems reach cells through three main routes: physical methods such as electroporation and microinjection, viral vectors such as adeno-associated virus (AAV), adenovirus and lentivirus, and nonviral carriers such as lipid nanoparticles (LNPs). Emerging virus-like particles and hybrid combinations add further options. The right choice depends on the target cells or tissue, whether editing occurs outside or inside the body, the cargo format and size, how long the editor should remain active, and the acceptable balance among efficiency, cellular stress, immune response, persistence and manufacturing complexity.

No delivery method is best for every experiment or therapy. A method that works well for collected cells in an ex vivo process may be unsuitable for administering an editor directly into a person, while a carrier that reaches a tissue in vivo may not be practical for producing a controlled cell product.

Why delivery is the central problem in gene editing

A gene editor is useful only if its active components reach the intended cells and cross the cell membrane. Some systems must also reach the nucleus, where genomic DNA is located. Delivery therefore has two linked jobs: transporting the cargo and determining which cells or tissues receive it.

The cargo can be supplied as DNA, RNA or a preassembled protein–guide complex. These formats do not behave the same way. DNA can instruct a cell to produce the editor, RNA can be translated for a limited period, and a ribonucleoprotein (RNP) delivers the already assembled Cas9 protein and guide RNA. The format affects how long editing activity lasts, what a carrier can package, and the risks associated with prolonged expression.

“Delivery efficiency” is incomplete unless it identifies the editor, cargo format, cell type, route of administration and measurement used. A high fraction of cells exposed to a carrier does not necessarily mean that those cells received a functional editor or achieved the intended edit.

The main delivery families

Delivery family How it gets cargo into cells Common setting Main advantages Important trade-offs
Physical delivery Temporarily opens or disrupts the membrane, or introduces material directly Ex vivo cell engineering and controlled laboratory experiments Can deliver DNA, RNA or RNP directly; does not require a viral carrier Membrane damage, cellular stress and reduced viability are possible
Viral vectors Uses engineered viral particles to carry nucleic-acid instructions into cells Ex vivo and in vivo research, depending on vector and target Efficient entry and, with suitable designs, access to selected cell types or tissues Immune responses, cargo limits, persistence or integration behavior, and prolonged editor expression
Nonviral carriers Packages or associates cargo with lipid, polymer, peptide or inorganic particles Especially transient RNA delivery and in vivo development Flexible formulation and strong interest in temporary expression Distribution depends on formulation and tissue; performance varies substantially among systems
Virus-like particles Uses particle-based delivery without relying on a conventional replicating virus Emerging research applications Potential to combine particle delivery with a different safety and cargo profile Platform maturity and evidence base are less established than for widely used approaches

Physical delivery methods

Electroporation

Electroporation applies an electrical field to cells. The field creates transient pores in the membrane, allowing editing cargo to enter before the membrane reseals. It is widely used when cells can be collected, edited under controlled conditions and then returned to a patient or used in a downstream process.

Electroporation can introduce all three common CRISPR-Cas9 formats: plasmid DNA, Cas9 messenger RNA with guide RNA, and Cas9–guide RNA RNP. That flexibility is valuable when researchers want to compare transient RNP activity with longer DNA-driven expression or when a protocol requires several components at once.

The cost is cellular stress. Electrical conditions that improve uptake can also damage membranes or reduce viability, and the best settings depend on the cell type and cargo. A useful assessment therefore measures both editing activity and the number and health of cells that remain after treatment.

Microinjection

Microinjection delivers material directly into individual cells through a fine instrument. Because the operator controls where and when the cargo is introduced, it is useful for tightly controlled experimental work. Its limitation is throughput: manipulating cells one by one is not a practical general solution for manufacturing large cell populations or treating an entire tissue.

Microfluidic squeezing, sonoporation and hydrodynamic injection

Microfluidic squeezing forces cells through narrow channels so that temporary membrane disruptions permit cargo entry. Sonoporation uses ultrasound-related mechanical effects to increase membrane permeability. Hydrodynamic injection relies on a rapid fluid volume to drive material into a target compartment. These approaches can be useful in specialized studies, but their operating windows, scale and tissue applicability are narrower or still developing compared with routine electroporation.

Viral-vector delivery

Viral vectors are engineered to carry nucleic-acid instructions into cells. The vector does not make every cell in a tissue equally accessible: delivery depends on the vector design, administration route, formulation, dose and biological context. A vector’s ability to enter a cell also does not by itself guarantee the desired editing outcome.

Adeno-associated virus (AAV)

AAV is widely considered when a system must reach cells in vivo or when a particular vector design has useful tissue access. Its packaging capacity is a major design constraint. An editor, guide sequence and donor template may not all fit in one particle, especially when the editor or repair template is large. Researchers may therefore need a smaller editor, split components or a different delivery combination.

AAV-based designs also require attention to immune response and how long the delivered instructions remain active. The appropriate assessment depends on the target tissue, route and construct rather than on the AAV label alone.

Adenovirus

Adenoviral vectors can carry genetic instructions into cells and are another viral option for research delivery. As with other viral systems, the relevant questions include which cells are reached, how much cargo the design can carry, how long expression persists and how the immune system responds.

Lentivirus

Lentiviral vectors are used for cell engineering and can provide efficient delivery of genetic instructions. Their persistence or integration behavior depends on vector design and must be considered when an editor is expressed for an extended period. That longer exposure can be useful for some engineered-cell workflows but may be undesirable when the goal is a brief editing pulse.

Viral-vector decision points

  • Immune response: pre-existing or treatment-related immunity can affect distribution and repeat dosing.
  • Cargo capacity: the editor and any donor template must fit within the selected vector’s packaging constraints.
  • Duration: prolonged expression may increase the window for unintended activity, while transient exposure may be preferred for some edits.
  • Persistence and integration: these properties vary by vector and construct and should be evaluated rather than assumed.
  • Manufacturing: reproducible particle production, purification and quality testing are part of the delivery decision.

Nonviral carriers

Lipid nanoparticles

Lipid nanoparticles package or associate with editing cargo and help it cross cell membranes. They are especially attractive for transient RNA delivery and for in vivo applications. An LNP formulation can be designed around a particular cargo and route, but LNPs are not interchangeable products: composition, particle properties, dose and administration route influence where the particles travel and which cells take them up.

Because RNA does not require the cell to transcribe a delivered DNA template, an RNA-focused LNP strategy can support temporary editor production. The practical result still depends on how much cargo reaches the intended cells, how efficiently it is released inside them and how long the RNA remains available.

Other nonviral systems

Researchers are also developing polymeric, peptide and inorganic nanoparticles, cell-penetrating peptides and DNA nanostructures. These systems may offer different loading, release or targeting properties. However, promising laboratory results should not be treated as evidence of established clinical performance. Each platform needs its own data on distribution, toxicity, immune effects, editing activity and manufacturing reproducibility.

Virus-like particles and combined strategies

Virus-like particles

Virus-like particles (VLPs) use particle-based delivery without relying on a conventional replicating virus. They are being investigated as a way to package or present editing components while potentially avoiding some limitations of standard viral vectors. Reviews describe VLPs as promising, but their maturity and evidence base remain distinct from more established delivery approaches.

Hybrid delivery

A combination can assign different jobs to different carriers. For example, cells may receive editor cargo by electroporation while an AAV supplies a donor template. Other designs combine lipid and viral components. These strategies can address a size or timing problem that one carrier cannot solve alone, but they add process steps, compatibility questions and manufacturing complexity.

Which cargo format is being delivered?

Cargo format What the cell receives Typical delivery considerations Duration implication
Plasmid DNA DNA encoding Cas9 and guide RNA Electroporation can introduce it; viral vectors generally deliver genetic instructions The cell can produce editor components after the DNA is delivered
Messenger RNA plus guide RNA Cas9 mRNA and a separate guide RNA Electroporation can deliver it; LNPs are commonly considered for RNA cargo RNA supports temporary production rather than a DNA template that remains available for transcription
RNP Preassembled Cas9 protein–guide RNA complex Electroporation is a practical route for direct delivery The cell receives active protein directly, enabling a transient editing exposure

The table describes compatibility, not a ranking. A method can handle a cargo format technically yet still be unsuitable for a particular cell type, tissue, scale or safety requirement.

Ex vivo versus in vivo delivery

Ex vivo editing

In an ex vivo workflow, cells are collected, exposed to the editor outside the body, assessed and then used or returned. This setting gives the team control over cell concentration, exposure conditions, washing, enrichment and release testing. Electroporation is therefore a common choice, particularly when RNP, RNA or DNA must be introduced directly.

  1. Collect and prepare the cells: establish the cell population and conditions needed to keep it viable during handling.
  2. Select the cargo: choose DNA, mRNA plus guide RNA, RNP or a combination based on the intended duration and edit.
  3. Apply the delivery method: set physical or vector conditions appropriate for the cell type and cargo.
  4. Recover the cells: allow membrane recovery or post-vector expression under controlled conditions.
  5. Measure both performance and health: assess the intended edit, the fraction of cells reached and cell viability; do not rely on a single uptake measurement.
  6. Decide whether the product is usable: include quality, reproducibility and safety criteria before the cells are used further.

In vivo editing

In vivo delivery administers the editor or its instructions directly to the organism. The central challenge is access: the carrier must reach the relevant tissue, enter the intended cells and release functional cargo without unacceptable distribution or immune effects. Route, formulation, vector design and biological context all influence the result.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

LNPs are of particular interest for in vivo RNA delivery, while viral vectors may be selected when their distribution or expression characteristics fit the target. Physical methods are generally less suited to whole-body delivery because they require direct manipulation of cells or a defined compartment. A tissue described as “targeted” should therefore be understood as preferentially reached under stated conditions, not as an absolute guarantee that no other tissue receives cargo.

How to choose a delivery method

  1. Define the setting: decide whether cells can be collected and returned, or whether administration must occur in vivo.
  2. Name the target: identify the cell type or organ and the route through which it can realistically be reached.
  3. Choose the cargo: compare DNA, mRNA and RNP according to required activity duration and component size.
  4. Check size and packaging: confirm that the editor, guide and any donor template fit the carrier or delivery process.
  5. Set the exposure goal: decide whether a short editing pulse or longer expression is acceptable.
  6. Define effectiveness: measure intended-cell access, editing activity and selectivity, including where the cargo distributes.
  7. Set safety limits: evaluate immune response, cellular stress, viability and unintended genome effects.
  8. Plan production: consider scale, formulation, reproducibility, release testing and whether cells must be handled outside the body.

This framework prevents a common mistake: selecting a platform because it performs well in a different cargo, cell type or administration setting. The same carrier can produce different results when any of those variables changes.

Common failure modes and how to interpret them

Many cells take up cargo, but few are edited

Uptake does not prove that cargo escaped into the right intracellular compartment or remained functional. Check the cargo format, intracellular release and the assay used to measure editing, not just the percentage of cells associated with particles or fluorescent material.

Editing is strong, but cell recovery is poor

This pattern is particularly relevant to physical delivery. Revisit electrical or mechanical conditions, exposure time and recovery handling, then compare editing per surviving cell with total usable-cell yield. A protocol that maximizes editing in a small surviving fraction may be less useful than a moderate-editing protocol that preserves more cells.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

The editor reaches the wrong tissue or persists too long

For viral and nanoparticle systems, distribution is shaped by route, formulation and vector design. Reconsider those variables and measure unintended distribution. If prolonged expression is not required, a transient RNA or RNP strategy may be more appropriate, provided the chosen cells can be reached.

The complete construct does not fit

AAV packaging constraints can become decisive when the editor and donor template are large. Options include reducing cargo size, separating components, changing the vector or using a hybrid strategy in which different carriers deliver different elements.

What is established—and what still needs qualification

Electroporation is a practical, widely used approach for controlled ex vivo cell engineering. Viral vectors have established research roles and can provide efficient entry, but their immune, persistence, integration and packaging characteristics must be evaluated for each construct. LNPs and other nonviral carriers are active areas of development, especially for transient RNA and in vivo delivery, yet results from one nanoparticle formulation should not be generalized to all others. VLPs and several physical alternatives remain promising but more specialized or less mature.

These platform descriptions do not establish that a particular construct is clinically available or approved. Clinical status depends on the specific editor, carrier, dose, route, manufacturing process, geography and regulatory decision, and it can change over time.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Bottom line

Gene-editing cargo can enter cells through electroporation and other physical techniques, viral vectors, lipid and other nonviral carriers, VLPs, or combinations of these approaches. Start with the target and setting, then match the cargo format, size and desired duration to the delivery system. Evaluate functional editing, cell or tissue selectivity, viability, immune response, persistence and manufacturing together; no single delivery family wins on every criterion.
