De-extinction can help aging researchers compare ancient DNA, living relatives and conserved biological signals, but it has not shown a way to reverse aging or extend human life. The key is to keep three questions separate: whether an animal can be engineered to express traits inferred from an extinct species, whether biological age can be estimated, and whether an intervention increases healthspan or lifespan.

The best current evidence supports useful connections between comparative genetics and aging biology. It does not support the claim that bringing back an extinct animal—or measuring its DNA methylation age—constitutes a human longevity treatment.

What “de-extinction” usually means

In public discussion, de-extinction can sound like recovering an extinct species intact and producing an identical copy. Modern projects generally do something more limited: researchers identify genetic variants associated with selected extinct traits, edit the genome of a living relative, and raise the edited animal in a living species’ biological and environmental context.

It is not retrieval of an intact extinct genome

Ancient DNA is fragmented, chemically damaged and incomplete. Even when scientists recover enough sequence to compare an extinct animal with living relatives, gaps remain and the extinct animal’s development, behavior and ecological history cannot simply be reconstructed from a sequence. Colossal Biosciences describes its approach as making close approximations of an extinct species’ genome through genetic engineering rather than creating a clone. The company also acknowledges that the resulting animals cannot be genetically, behaviorally and psychologically identical to their extinct ancestors. That is a company description, not an independent proof that an extinct species has been restored in every biological sense.

The dire-wolf example

A University of California, Santa Cruz report dated April 14, 2025, and modified May 27, 2025, provides a concrete example. The report describes pups named Romulus and Remus as genetically modified gray wolves endowed with selected traits of the extinct dire wolf. Researchers used pieces of genetic code identified in ancient dire-wolf DNA to alter gray-wolf genomes. The university’s caption and text make clear that the pups were not exact replicas of dire wolves.

The same report says the dire-wolf genome analyzed by the study team appears to derive roughly two-thirds from an ancestor of wolves and coyotes and the remaining third from an ancient lineage related to South American dogs. The researchers present that pattern as evidence consistent with hybrid speciation about 4 to 5 million years ago. Those proportions and the evolutionary interpretation should be attributed to the reported study findings, rather than treated as timelessly settled facts about every dire-wolf population.

Beth Shapiro, an evolutionary biologist and Colossal chief science officer, said in the UC Santa Cruz report: “Ancient DNA is notoriously difficult to work with because it is often fragile and degraded, but paleogenomics has made tremendous leaps forward in the past 20 years, leading to the recent groundbreaking achievement of the world’s first de-extinction with the birth of Colossal’s dire wolves,” Shapiro said. “World’s first de-extinction” is her characterization as quoted by the university. The operational result described in the same report is narrower: edited gray wolves carrying some inferred dire-wolf traits.

What aging researchers actually measure

Aging studies can involve chronological age, biological age, molecular damage, disease risk, healthspan or lifespan. These are related but different endpoints. A method that predicts age accurately is not automatically a method that slows aging, and a molecular difference associated with mortality is not automatically a cause of mortality.

Epigenetic clocks

An epigenetic clock estimates age from patterns of chemical marks on DNA, especially DNA methylation at selected sites. Methylation patterns tend to change with age, so a statistical model can use them to estimate chronological age or derive a “biological-age” measure. A sample can therefore appear older or younger than its calendar age according to the clock, without that result proving that the person or animal has been rejuvenated or will live longer.

What the 2023 mammalian clock study found

A peer-reviewed 2023 paper in Nature Aging, “Universal DNA methylation age across mammalian tissues,” reported cross-mammal models built from 11,754 methylation arrays covering 59 tissue types and 185 mammalian species. The study’s abstract reports predictive models with a correlation greater than 0.96. Its evaluated cross-validation results also report median absolute error below one year and relative error below 3.3% for the models tested.

Those figures describe performance in the study’s datasets and validation procedures. They are not a guarantee that every species, tissue, laboratory assay or consumer test will estimate age with the same accuracy. A model can perform strongly on the organisms and samples used to build and validate it yet require recalibration for a new species, tissue or population.

The paper also reports that age deviations from its methylation predictions correlate with human mortality risk, mouse somatotropic-axis mutations and caloric restriction. These are associations involving an age-estimation framework and biological conditions. They do not demonstrate that changing a methylation score, editing a gene, or creating a de-extinct animal will extend human lifespan.

What the National Institute on Aging adds

A 2024 National Institute on Aging summary describes a separate line of human research in which DNA-methylation-based biological-age measures can help predict age-related health outcomes, chronic disease, cognitive and functional limitations, and mortality in older adults. The summary notes that associations differ somewhat by clock type. This supports the use of methylation measures as risk and aging-biology indicators; it does not turn any particular clock into a validated anti-aging therapy.

Where de-extinction and aging research genuinely connect

The connection is comparative biology. Extinct species leave partial genetic records, while living relatives provide cells, genomes and measurable physiology. Comparing those materials can help researchers ask which traits evolved, which gene networks are conserved and how environmental pressures shaped mammalian biology.

Ancient DNA can broaden the comparison set

Ancient genomes can reveal variants that disappeared from living populations or show how traits changed over millions of years. Those variants may point researchers toward pathways involved in metabolism, immune function, development or stress responses. Finding a candidate pathway is a starting point for experiments, not evidence that the pathway can safely alter human aging.

Living relatives provide the experimental system

Because extinct cells and tissues are unavailable, any engineered animal is created in a living host. Researchers can then measure development, physiology, behavior and health under controlled conditions. That may make it possible to test how a selected genetic change affects a trait in the host species. It still does not reproduce the extinct species’ complete genome, microbiome, developmental history or ecological interactions.

Cross-species clocks can reveal conserved signals

The mammalian methylation study suggests that some age-associated DNA patterns are sufficiently shared across species to support broad models. Such conservation can help identify biological processes worth investigating. It does not establish that a trait selected for de-extinction is an aging intervention, or that a methylation pattern is a lever that can be safely manipulated to increase lifespan.

What de-extinction cannot currently prove about longevity

It does not show rejuvenation

Engineering a young animal to express a physical trait—such as body size, coat characteristics or aspects of skull shape—addresses trait reconstruction. Rejuvenation would require evidence that an older organism’s age-related damage, function or risk has been reversed. The reviewed de-extinction reports do not provide that evidence.

It does not establish human lifespan extension

A claim about human longevity requires a defined intervention, a specified human or animal population, a prespecified endpoint and an appropriate study design. Depending on the question, endpoints could include survival, disease-free survival, functional capacity or validated healthspan measures. Ancient-genome reconstruction and methylation-age prediction are methods or observations, not lifespan-extension trials.

It does not erase species differences

Mice, wolves, humans and other mammals differ in development, metabolism, immune systems, reproductive schedules and causes of death. A correlation that appears in a cross-species model can identify a conserved signal while still failing to predict how a human intervention will work. Even within one species, clock performance and age-related risk can vary by tissue, sex, ancestry, health status and laboratory method.

How to compare a de-extinction claim with an aging claim

Use the same five questions for every statement. They prevent a visually dramatic animal or a precise-looking age estimate from being mistaken for a longevity result.

Comparison axis De-extinction example Aging or longevity example Why it matters
Organism and material Ancient DNA interpreted through a living relative such as a gray wolf DNA-methylation measurements from specified mammalian tissues or human samples The source material and host determine what can be inferred.
Method Ancient-genome analysis and targeted gene editing Statistical age modeling, methylation assays or an intervention study A reconstruction method is not the same as a treatment method.
Primary endpoint Resemblance to selected extinct traits Chronological-age prediction, health outcome, healthspan or lifespan Only the endpoint actually measured supports the corresponding conclusion.
Evidence type Company explanation and institutional reporting on a collaborative project Peer-reviewed model validation, government research summary or controlled intervention trial Source type affects how confidently a claim can be generalized.
Extrapolation limit Edited gray-wolf traits do not equal a complete dire wolf A cross-mammal association does not equal a human treatment effect Biological similarity does not remove species and context differences.

Could a de-extinct animal become useful for aging research?

In principle, an engineered animal could be useful if researchers define a testable biological question before interpreting its results. For example, they might compare edited and unedited animals of the same host species, measure a specified molecular pathway, and follow health or survival outcomes under an approved protocol. The informative result would be the measured difference between groups—not the animal’s extinct-species label.

Several safeguards would be essential:

  • Specify the edit: identify the exact variant or regulatory change and why ancient or comparative evidence implicates it.
  • Specify the endpoint: distinguish a visible trait, a molecular marker, a disease measure, healthspan and lifespan.
  • Use matched controls: compare edited animals with appropriate unedited or sham-edited controls raised under comparable conditions.
  • Follow the animals long enough: an early developmental measurement cannot establish an effect on late-life disease or survival.
  • Replicate and report adverse effects: unintended edits, developmental problems, fertility changes and welfare outcomes are part of the result.
  • Test translation separately: a finding in the host species would still require independent evidence before any human application.

Without those steps, “this animal looks younger,” “its clock is lower” or “it carries an ancient variant” remains an observation rather than a longevity intervention.

Common reasoning errors to avoid

Calling an edited relative an identical resurrection

The dire-wolf case illustrates why wording matters. The pups are described as genetically modified gray wolves with selected dire-wolf traits, not as exact copies. “De-extinction” can be used as a project label or a broad characterization, but the biological details should be stated alongside it.

Treating a clock score as a treatment result

If an intervention changes a methylation-based age estimate, that is evidence about the clock’s readout. It becomes evidence of slowed aging only if independent measures show improved function, reduced disease or longer survival, with a design that rules out measurement artifacts and short-term changes unrelated to aging.

Turning correlation into causation

The Nature Aging paper’s correlations and the NIA summary’s health-outcome associations identify relationships. They do not show that the measured methylation sites cause mortality, that changing them will improve health, or that an extinct-species genome contains a safe human longevity program.

Ignoring who made the claim

Separate a company’s account of its goals and methods from an institutional report of a study and from peer-reviewed evidence. The UC Santa Cruz report is useful for describing the dire-wolf work and its reported ancestry findings; the company’s explanation is useful for describing its stated approach; the Nature Aging article supplies the quantitative clock results. None of those source types alone substitutes for a human longevity trial.

A practical checklist for reading future headlines

  1. Write down the organism: human, mouse, wolf, another mammal or a mixed dataset.
  2. Identify the material: ancient DNA, edited cells, living tissue, methylation data or clinical measurements.
  3. Name the method: sequencing, gene editing, methylation modeling or an intervention.
  4. Record the endpoint and time frame: trait appearance, estimated age, disease risk, healthspan or survival.
  5. Check the evidence type and date: company statement, institutional report, peer-reviewed paper, government summary or controlled trial.
  6. Ask what comparison is missing: unedited controls, another clock, another tissue, a different species or long-term follow-up.
  7. Reject any leap from “associated with age” or “resembles an extinct trait” to “extends human life” unless a study directly measured that outcome.
