DNA testing can recover biological clues from some ancient Egyptian remains, including maternal mitochondrial lineages, biological sex, possible close kinship, and—when enough authentic nuclear DNA survives—genetic affinities relevant to population history. It cannot reliably produce a complete genetic profile from every mummy, and a result from one burial or site cannot stand for all ancient Egyptians.
What scientists can say depends on preservation, contamination controls, the amount and type of DNA recovered, and the reference populations used for comparison. The strongest conclusions are therefore tied to a named individual, archaeological context, date and laboratory method.
What kinds of information can mummy DNA provide?
Ancient-DNA laboratories may analyze two broad sources of human genetic material. Mitochondrial DNA (mtDNA) occurs in many copies per cell and is inherited along the maternal line. It can support comparisons of maternal lineages and sometimes survives when nuclear DNA is too degraded for broader analysis. Nuclear DNA is distributed across the chromosomes and, when sufficient authentic sequence is available, can support biological-sex inference, kinship tests and genome-wide ancestry comparisons.

These tests describe biological evidence, not modern national, ethnic or racial identities. An affinity to a reference population is a statistical comparison, shaped by which ancient genomes are available and by the model used.

What the 2017 Abusir el-Meleq study says—and how far it reaches
The 2017 investigation is valuable because it combined a relatively large archaeological sample with different levels of genetic recovery. Researchers examined 151 mummified individuals from Abusir el-Meleq, a site in Middle Egypt, spanning the late New Kingdom to the Roman period. They obtained complete mitochondrial genomes from 90 samples, but only three male individuals yielded genome-wide SNP data that met the study’s quality criteria.
In the study’s comparison, the ancient Abusir el-Meleq individuals shared more ancestry with Near Eastern reference groups than present-day Egyptians did. The authors related the difference to additional sub-Saharan ancestry entering the present-day comparison population during more recent history. That is an interpretation of the sampled people, dates and reference panels—not a timeless description of Egypt’s population.
The contrast between 90 complete mtDNA results and three usable genome-wide datasets is instructive. A project can recover a reliable maternal lineage from many remains while obtaining too little nuclear DNA for broad ancestry modeling. Reporting the denominator and the quality threshold prevents a mitochondrial result from being mistaken for a complete genome.

What the 2025 Nuwayrat genome adds
Morez Jacobs and colleagues reported a 2.02×-coverage whole genome from one adult male excavated at Nuwayrat. Radiocarbon dating placed him at 2855–2570 calibrated years BCE, reaching back to the Old Kingdom era. In the authors’ ancestry models, most of the individual’s ancestry was best represented by North African Neolithic sources, with approximately 20% modeled as related to genomes representing the eastern Fertile Crescent.
The authors interpret the result as consistent with contact that included human movement as well as exchange of objects, imagery, domesticated plants and animals, and writing systems. Their abstract says that contacts between Egypt and the eastern Fertile Crescent “were not limited to objects and imagery … but also encompassed human migration.” That statement applies to the one genome and the comparative models used. The authors also emphasize that more genomes are needed to describe early Egyptian genomic diversity.
At 2.02× coverage, many genomic positions are represented by relatively few reads. Statistical modeling can still extract useful signal, but uncertainty is greater than it would be with a high-coverage modern genome. The result is therefore evidence about one person in a particular time and place, not an estimate that 20% of every Old Kingdom Egyptian’s ancestry came from the same source.

How scientists infer biological sex and kinship
Biological sex
Sequencing reads can be counted against the X and Y chromosomes. A pattern containing substantial Y-chromosome sequence alongside X-chromosome sequence supports a male biological-sex assignment; reads concentrated on X chromosomes with little or no authenticated Y signal support a female assignment. Laboratories must account for low coverage, damaged DNA and contamination before making the call.
The 2018 mummy-head report demonstrated this approach on a roughly 4,000-year-old Egyptian specimen using shotgun sequencing. Its mtDNA work also helped assess authenticity. The case reported mitochondrial contamination estimates of 0.6% in one library (95% confidence interval 0.08–1.8%) and 11.6% in another (95% confidence interval 9.7–13.6%). Those values describe the two libraries in that study, not a standard contamination rate for Egyptian mummies.
Kinship
Close biological relationships can be tested with shared nuclear variants, runs of homozygosity and, where available, Y-chromosome or mitochondrial-line evidence. A parent-child or sibling hypothesis needs enough independent markers and a suitable comparison sample. If only a short mitochondrial sequence survives, the result can address maternal-line compatibility but generally cannot prove a close relationship by itself.

How an ancient-DNA investigation works
- Define the archaeological question and permissions. Researchers document the site’s date, burial context and specimen identity, then obtain authorization to sample curated human remains. DNA extraction and sequencing are specialized procedures, not consumer-kit services.
- Select tissue likely to preserve DNA. Studies may sample teeth, dense bone or, when appropriate, soft tissue. Heat, humidity, burial chemistry, embalming treatment and elapsed time all affect survival, so no tissue guarantees success.
- Extract DNA in a dedicated facility. Ancient-DNA rooms are physically and procedurally separated from modern-DNA work. Workflows include extraction blanks and other negative controls to detect laboratory-introduced molecules.
- Build sequencing libraries. DNA fragments are prepared for high-throughput sequencing. Researchers may sequence all fragments (shotgun sequencing) or use targeted approaches, depending on the question and quantity of material.
- Measure authenticity and contamination. Ancient molecules commonly show characteristic damage patterns and fragment-length distributions. Teams compare those patterns with controls, estimate the proportion of modern human DNA and check whether results are reproducible across libraries or extracts.
- Classify the recovered data. Analysts report whether they obtained mtDNA, targeted markers, sex-chromosome information or genome-wide sequence. A sample that fails a genome-wide quality threshold can still yield a useful mitochondrial result.
- Compare with appropriate references. Ancestry models use statistical fits to ancient and present-day reference genomes. The chosen references, coverage, missing data and model assumptions are reported because they affect the inferred affinity.
- Integrate genetics with archaeology. Genetic similarity can be consistent with movement or admixture, but it cannot by itself identify a language, cultural practice, political allegiance or the exact cause and route of migration.
Why preservation and contamination determine the answer
Ancient DNA is not evenly distributed through a mummy. Temperature, moisture, soil chemistry, microbial activity, embalming materials and handling history can destroy or chemically alter molecules. A failed extraction means that usable DNA was not recovered at adequate quality; it does not show that the person lacked a particular lineage or trait.
Modern human DNA can enter during excavation, conservation, sampling and laboratory work. Dedicated ancient-DNA facilities, extraction blanks, damage-pattern analysis, independent libraries and statistical contamination estimates are used together. A high percentage of human reads is not automatically a high-quality ancient genome: those reads must also display authentic damage, map reliably and survive contamination checks.

An ancestry percentage is the output of a model that represents an individual’s genome as a mixture of similarities to selected reference groups. It is not a literal measurement of modern nationality or a direct label for ethnicity. “Related to the eastern Fertile Crescent,” for example, identifies the reference affinity in the model; it does not name a single ancient tribe or prove that a person migrated along one known route.

Reference panels are incomplete. Some ancient populations are unsampled, and genetic variation often changes gradually across geography rather than stopping at political borders. Different models can therefore emphasize different source groups. Strong reporting states the archaeological context, the reference genomes, the coverage and the uncertainty alongside the headline affinity.
Why one mummy cannot represent all ancient Egyptians
Egypt’s population changed across thousands of years and included communities in different ecological and trade settings. Abusir el-Meleq is one Middle Egyptian site, while Nuwayrat’s 2025 result comes from one adult male. Even the 2017 dataset, with 151 sampled individuals, reflects the people buried at one location and the periods represented there.
A responsible comparison records five dimensions: the site’s geography and date; the tissue sampled; whether the output is mtDNA, targeted markers or genome-wide sequence; the authentication and contamination controls; and whether the conclusion concerns one person or a community sample. Comparing the 2017 mitochondrial dataset directly with the 2025 low-coverage genome without those distinctions would create a false sense of equivalence.

What current institutional capacity means for readers
The National Museum of Egyptian Civilization describes ancient-DNA laboratories supporting work on human kinship, disease and ancient ecology. Its institutional information also reports a capacity-building grant to procure next-generation sequencing equipment for metagenomic analysis of ancient Egyptian biological remains and artifacts. Such facilities represent research infrastructure and permitted scientific study, not a service where the public can mail in a mummy or order a consumer ancestry test.
How to read a headline about “mummy DNA”
- Ask whether the result came from mtDNA, a few targeted markers or a genome-wide dataset.
- Check the number of individuals sampled and the number that passed quality control.
- Look for the archaeological site, date and tissue type.
- Look for damage-pattern and contamination evidence, not just the word “sequenced.”
- Separate a modeled regional affinity from a modern identity label.
- Read migration claims alongside archaeological and historical evidence.
DNA can therefore add a powerful biological dimension to the study of Egyptian mummies: maternal-line history, sex-chromosome evidence, possible kinship and carefully modeled population affinities. The result is strongest when its limits are stated with equal precision—what survived, how it was authenticated, how many people it represents and which comparisons made the inference possible.
