A new cross-species atlas shows that mitochondria still contain a large amount of biology scientists cannot explain. By comparing mitochondrial protein inventories from parasites, a plant, humans and yeast, the project identifies pathogen-specific protein families that could eventually support selective drug discovery while also sharpening questions about how mitochondria evolved.
What the atlas mapped
Nature reported the atlas on 16 October 2026. The researchers assembled mitochondrial protein catalogues from six newly analysed organisms: five single-celled parasites and one plant. They then compared those data with established human and yeast mitoproteomes, creating a cross-lineage reference rather than a catalogue for one species.
A mitoproteome is the set of proteins associated with a cell’s mitochondria. Those proteins are encoded in two places: the mitochondrion retains a small genome of its own, while the much larger nuclear genome supplies most of the organelle’s components. Comparing both canonical mitochondria and mitochondrion-like organelles can expose proteins shared across lineages as well as adaptations found only in particular organisms.

The figures describe protein families as well as individual proteins. A family can contain related sequences from several organisms, so the family count is not a second estimate of the number of proteins.
What the atlas found in Giardia lamblia
Giardia lamblia is a particularly useful test case because it infects the oxygen-poor environment of the human gut and relies on reduced mitochondrion-like organelles that are difficult to study with methods developed for textbook mitochondria.
The project identified 59 mitochondrial proteins in Giardia. Thirty-two corresponded to previously known proteins and 27 were newly identified. The report also classified 34 of the Giardia proteins as having unknown function; that functional annotation is a separate category from whether a sequence itself was newly detected.

The organism infects about 280 million people worldwide each year, according to the epidemiological figure cited by Nature. Its unusual organelles therefore matter both for basic cell biology and for understanding a widespread parasite.
Cell biologist Pavel Doležál of Charles University in Prague said the uncharacterized proteins suggest “it means that there is some new metabolism that we don’t know about. And it was overlooked”.
Why parasite mitochondria matter for medicine
Parasites cannot simply be treated as small versions of human cells. Their mitochondria or mitochondrion-like organelles may run different pathways, use different enzymes or operate under environmental conditions that human mitochondria do not encounter. Those differences can reveal vulnerabilities that are absent from the host.

The strongest therapeutic idea in the atlas is selectivity. The 33 protein families conserved in eukaryotic pathogens but missing from humans could, in principle, be attacked without hitting a human counterpart. Vamsi Mootha, a mitochondrial biologist at the Howard Hughes Medical Institute, described such pathogen-specific families as potentially “ideal drug targets” for therapies directed at parasite proteins rather than human ones.

The atlas does not show that any compound blocks one of these proteins, that a target is essential for parasite survival, or that a candidate drug is safe in animals or people. It also does not establish that a malaria or sleeping-sickness cure is imminent. Each lead would need genetic validation, biochemical and structural work, selectivity testing, toxicology and clinical trials.
Why so many proteins remain mysterious
About 43% of the protein families in the combined catalogue were not present in Pfam, a widely used database of protein families. Absence from Pfam does not prove that a protein is unique to mitochondria or that it has no relatives; it means the sequence did not match a family represented there at the time of the comparison.

Unclassified proteins are scientifically valuable because they can point to overlooked metabolism or lineage-specific adaptations. They are also difficult starting points for drug development: researchers first have to determine where each protein sits, what it binds, whether it is essential and whether its structure offers a tractable site for intervention.

The broad evolutionary model remains endosymbiosis. Roughly two billion years ago, an archaeal cell engulfed a bacterium; instead of digesting it, the host retained the bacterium as a permanent symbiont that became the mitochondrion.

Mitochondria still carry small genomes distinct from nuclear DNA, even though most mitochondrial proteins are now encoded in the nucleus. The new atlas does not overturn endosymbiosis. Its value is that proteins from parasites, plants, animals and yeast can be compared across many branches of the eukaryotic tree, allowing scientists to ask which features are ancient, which appeared later and which may have been lost.

Those comparisons reopen debates about the timing and details of the transition from free-living bacterium to integrated organelle. A protein shared by distant lineages may trace back to an early mitochondrial ancestor, while a protein found only in a parasite may reflect a later adaptation to its host or environment.
What readers should take from the atlas
- It is a comparative map of mitochondrial proteins, not a single-organism genome announcement.
- Its most concrete medical contribution is a list of parasite-associated families for testing, especially those with no human counterpart.
- Its largest basic-science contribution is evidence that substantial mitochondrial biology remains unannotated.
- Its evolutionary contribution is a denser set of cross-lineage comparisons for studying how an engulfed bacterium became an organelle.
Turning the atlas into medicines will require experiments that connect sequence families to real cellular functions and then show that disrupting those functions harms parasites more than human cells. Until those steps are completed, the atlas is best understood as a powerful map of possibilities rather than proof of a new therapy.
