Use dead-end filtration when your main job is straightforward pass-through filtration and the feed will not quickly foul the membrane. Use tangential-flow filtration (TFF, or crossflow) when you must retain a target while concentrating it, exchanging buffer, removing small solutes, fractionating by size, or processing a feed that would rapidly build a cake on a dead-end membrane. The deciding factors are flow path, fouling behavior, target retention, sample volume, viscosity and solids, equipment, and the recovery you need—not the name of the membrane alone.

Both modes can use microfiltration (MF) or ultrafiltration (UF) membranes. TFF is not automatically faster or better: crossflow can preserve flux under suitable conditions, but the result depends on the feed, membrane, pressure, temperature, crossflow, and process sequence.

Flow direction is the fundamental difference

In dead-end, or direct-flow, filtration, the feed is pushed toward the membrane and the intended product stream passes through it. Material that the membrane retains stays at the surface. As that layer thickens, resistance rises and permeate flow (flux) commonly falls.

In TFF, the feed travels parallel to the membrane. A portion crosses the membrane as permeate while the remainder leaves as retentate and normally returns to the feed vessel. The moving retentate creates shear along the membrane surface and can carry some deposited material back into the flowing stream.

Decision axis Dead-end filtration Tangential-flow filtration
Flow path Feed moves perpendicular to the membrane and is driven through it. Feed moves along the membrane; permeate crosses it and retentate remains in the loop.
What happens to retained solids or molecules They can accumulate at the membrane face and form a restrictive layer. Crossflow can sweep material away from the face and reduce rapid surface buildup.
Typical objective Pass-through filtration when the feed and filter are compatible. Concentration, diafiltration, fractionation, and selected clarification or cell-handling operations.
Equipment A holder and suitable filter, selected for the actual duty. A membrane module, recirculation path, pump, pressure controls, tubing and a feed vessel; some cassettes require a compatible pump head.
Scale Must be sized for the expected volume and solids load. Available from benchtop to production scale, with membrane area and flow ranges that vary by system.

The Cytiva/Pall technical guide describes these as the two principal membrane-filtration modes and notes that either mode can use MF or UF membranes. Therefore, “TFF versus UF” is not a valid comparison: TFF describes the flow arrangement, while UF describes a membrane separation range.

What dead-end filtration is good at

Simple clarification and sterile or particulate removal

Dead-end filtration is often the economical choice when the feed is relatively clean, the required volume is modest, and the filter can process the run before fouling causes an unacceptable pressure rise or flow decline. The permeate is the intended pass-through stream; retained solids remain on the filter and are discarded or collected.

When a short, disposable flow path matters

A dead-end capsule, cartridge or syringe filter can have fewer connections and less hold-up volume than a recirculating TFF assembly. That can simplify setup and cleaning. The trade-off is that every retained particle increases the resistance seen by the pump or operator, so capacity must be established for the actual feed rather than inferred from a clean-water rating.

Where dead-end filtration becomes a poor fit

A high concentration of protein, cells, colloids or other retained material can create a cake quickly. Viscous feeds and cold samples can further reduce flux. If the process must keep a valuable molecule in the retentate while removing liquid or small solutes, dead-end filtration does not provide the continuous retention-and-wash loop that TFF does.

What TFF adds to the process

Concentration

During concentration, the membrane retains the target while liquid and sufficiently small solutes leave as permeate. The retentate volume decreases and target concentration rises. A UF membrane is normally selected with a molecular-weight cut-off (MWCO) below the target’s molecular size. The cited technical guide gives a starting rule of choosing an MWCO about three to six times lower than the target molecular weight; that is guidance, not a recovery guarantee. If substantial diafiltration follows, a lower MWCO may be advisable to limit product loss.

Diafiltration and buffer exchange

Diafiltration removes permeable salts, solvents or other small molecules while the larger product remains in the retentate. In continuous diafiltration, fresh buffer enters the feed vessel as permeate leaves, keeping the retentate volume approximately constant while changing its composition. The guide gives an example in which five diafiltration volumes reduce ionic strength by approximately 99 percent. That figure applies to the stated continuous-diafiltration example and should not be treated as a universal result.

Fractionation

With an appropriate membrane and operating window, TFF can separate larger retained species from smaller permeable species. Fractionation depends on the actual size distribution, shape, concentration and interactions of the molecules, so a nominal MWCO does not by itself predict a sharp split.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Clarification and cell-handling steps

Some TFF configurations are used for clarification or cell removal. Whether this works depends on the membrane pore range, cell concentration, shear sensitivity and solids loading. A process that is called “clarification” in one system may be a very different duty from UF concentration in another.

How fouling and flux differ in practice

Dead-end filtration continually directs retained material into the membrane face. The resulting layer can narrow pores, increase cake resistance and force the operator to raise pressure or replace the filter. TFF introduces a parallel velocity that can limit the rate of surface accumulation, which is why crossflow may avoid the rapid flux decline seen in direct-flow filtration.

That benefit is conditional. High protein concentration, particulates, adsorption to the membrane, high viscosity and low temperature can still reduce flux in TFF. Excessive crossflow can also increase shear, foaming or pump stress. The correct comparison is therefore not “TFF never fouls,” but “TFF gives the process another variable—crossflow—to manage deposition while permeate is removed.”

Membrane selection: do not confuse MWCO with a guarantee

MWCO is a practical screening label, not a promise that every molecule above the number will be retained or every molecule below it will pass. Retention changes with molecular shape, aggregation, concentration, membrane chemistry, pressure and adsorption. Start with the target’s behavior in the actual formulation and confirm recovery experimentally.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.
  • For concentration: begin with a UF MWCO below the target size. The three-to-six-times-lower rule from the cited guide is a starting point.
  • For planned diafiltration: consider a lower MWCO if losing target through the membrane would be costly.
  • For particulate removal: select the MF or other pore range required for the particle-size duty; a UF MWCO is not interchangeable with a pore-size rating.
  • For sensitive products: evaluate adsorption, shear, temperature and residence time, not just nominal retention.

Membrane chemistry, area and module geometry also affect recovery. A membrane that performs well in a small screening device may need a different area or flow path at production scale.

Equipment and documented laboratory specifications

A TFF run needs a controlled recirculation path. Typical components are a feed vessel, membrane cassette or module, pump and pump head, tubing, pressure sensors or gauges, a permeate collection path and a way to control or measure retentate volume. The exact connections and acceptable pressure range belong to the selected module’s instructions.

For context, the Sartorius Vivaflow 200 research-use cassette page accessed October 13, 2026 lists one configuration with the following specifications:

Specification Listed value Qualification
Membrane area 200 cm² Value for the cited Vivaflow 200 configuration.
MWCO 10 kDa Value for the cited membrane configuration.
Maximum operating pressure 4 bar Do not exceed the module’s stated limit.
Minimum retentate volume 20 mL Configuration-specific minimum needed to maintain the loop.
Recommended feed volume 1,000–2,500 mL Vendor recommendation for that configuration.
Recommended flow rate 200–400 mL/min Vendor recommendation for that configuration.

These figures are not universal TFF settings. Confirm the current membrane, tubing, sample range, pump compatibility and availability before use. Sartorius describes its wider TFF portfolio as spanning benchtop through production systems and single-use as well as multi-use configurations; membrane area and flow ranges vary by system.

The Thermo Fisher Pierce cassette FAQ states that its cited setup requires a peristaltic pump and pump head, and that these are not supplied with the cassette. That requirement applies to that setup, not automatically to every TFF product.

A practical TFF workflow

1. Define the separation before choosing hardware

  1. Identify the molecule or particle you must retain and the species you want to remove.
  2. Set the target volume, concentration factor, diafiltration volume or fraction cut.
  3. Record feed concentration, solids, viscosity, temperature, pH and any known adsorption or shear sensitivity.
  4. Choose MF or UF and a starting MWCO or pore range based on the duty.

2. Size the membrane and verify the loop

  1. Use the manufacturer’s area and flow guidance for the selected module.
  2. Check that the pump, pump head and tubing can deliver the recommended flow without exceeding pressure limits.
  3. Confirm that the intended final retentate volume remains above the module’s minimum working volume.
  4. Account for hold-up volume in tubing and the cassette so product is not stranded at shutdown.

3. Start gently and establish a baseline

  1. Install the membrane and wet or condition it according to the module instructions.
  2. Check all connections, permeate routing and pressure instruments.
  3. Start recirculation at a conservative flow and verify that retentate returns to the feed vessel.
  4. Record baseline pressure, temperature, retentate flow and permeate flux before adding the sample.
  5. Introduce the feed and adjust crossflow and transmembrane pressure within the module’s documented range.

4. Run concentration or diafiltration

  1. For concentration, remove permeate while monitoring retentate volume and product concentration.
  2. For continuous diafiltration, add compatible fresh buffer at the rate required to replace permeate and track the number of diavolumes.
  3. Watch for rising pressure, falling flux, foaming, temperature drift or an unexpected change in retentate appearance.
  4. Stop at the defined endpoint rather than at an arbitrary time.

5. Recover product and document the run

  1. Reduce pressure and recover the retentate using the module’s recommended flush or recovery method.
  2. Measure final volume, concentration and recovery; sample permeate if product loss is a concern.
  3. Record membrane, area, MWCO, flow, pressure, temperature, feed composition and endpoint.
  4. Follow the module’s instructions for cleaning, sanitization or disposal.

A practical dead-end workflow

  1. Define the required pass-through quality and the solids load the filter must retain.
  2. Select a membrane material and pore rating compatible with the fluid and target.
  3. Size the filter area for the actual volume, viscosity and expected fouling burden.
  4. Install the filter in the specified orientation and wet it if required by the instructions.
  5. Begin at the permitted pressure or vacuum, then monitor flow and differential pressure.
  6. Stop when the endpoint is reached or the filter approaches its pressure, capacity or integrity limit; do not force a fouled filter to maintain flow.

Troubleshooting guide

Symptom Likely contributors Checks and corrective actions
Dead-end flow falls rapidly Surface cake, excessive solids, viscous or cold feed, undersized area. Check differential pressure and feed temperature; pre-clarify if appropriate, reduce loading, or select more area or a different filtration step.
TFF permeate flux declines Fouling, adsorption, concentration polarization, high viscosity or low temperature. Check feed condition, pressure, crossflow and temperature within validated limits; assess whether the membrane chemistry is compatible.
Pressure rises unexpectedly in TFF Blocked tubing, closed valve, precipitate, excessive concentration or flow outside the module range. Stop safely, inspect the flow path, verify valve positions and compare conditions with the cassette’s pressure limit.
Target appears in permeate MWCO too open, aggregation state changed, pressure or chemistry altered retention. Assay permeate and retentate, reassess membrane selection and operating conditions, and lower MWCO if justified by testing.
Recovery is lower than expected Adsorption, hold-up volume, incomplete recovery, degradation or passage through the membrane. Perform a mass balance, rinse or recover the loop as specified, and test membrane-product compatibility.
Foaming or product damage occurs Excessive shear, air entrainment, leaks or unsuitable pump settings. Inspect connections and vessel level, remove air, reduce stress within validated limits and confirm product sensitivity.

Decision checklist

  • Choose dead-end when the feed is suitable for a one-pass filter, the retained load is manageable and simplicity is more valuable than recirculation control.
  • Choose TFF when you need concentration, diafiltration, buffer exchange, size fractionation or sustained processing of a fouling-prone feed.
  • Choose neither by membrane label alone: MF and UF membranes can be used in both flow modes.
  • Treat MWCO, pressure, flow and volume values as process- or module-specific starting points, then validate retention, flux and recovery with the real feed.
  • Include pump, pump head, tubing, pressure control, minimum working volume, hold-up and cleaning or disposal in the equipment decision.

The most defensible choice is the one that meets the separation objective while preserving product recovery at an acceptable pressure, flux and handling burden. A small feasibility run with the actual feed is more informative than assuming that crossflow or direct flow will behave ideally.
