In a 2019 experiment, researchers used light confined by a metallic tip to map vibrational patterns inside a single molecule at angstrom-scale resolution. The technique revealed where a molecule’s atoms move together—not by filming them in motion, but by measuring and imaging their vibrational signals.

What does it mean to see a molecule vibrate?

Atoms in a molecule are not stationary. They move in coordinated patterns called normal modes: characteristic ways the molecule’s atoms shift relative to one another. A vibrational spectrum can identify those motions by their frequencies, but it does not by itself show where within a molecule each mode is strongest.

In their 2019 Nature paper, Joonhee Lee, Kevin T. Crampton, Nicholas Tallarida and V. Ara Apkarian reported a way to record vibrational spectra from an individual molecule and produce spatial images of its normal modes. The work addressed a longstanding challenge: resolving molecular motion requires angstrom-scale spatial detail, so normal modes had generally been inferred through theory rather than directly visualized at that scale. Nature’s paper, published 3 April 2019, reports the method and results.

How did the researchers measure the vibrations?

The team used tip-enhanced Raman spectromicroscopy (TER-SM). A metallic tip concentrates light into an extremely small region near its apex, while surface-enhanced Raman scattering amplifies the light signal associated with a molecule’s vibrations. By scanning the tip over an individual molecule and recording the localized Raman response, the researchers could map how the signal associated with a vibrational mode varied across the molecule.

The experiment took place in a cryogenic, ultrahigh-vacuum scanning tunnelling microscope (STM). The metallic tip and molecule formed a carefully controlled junction; the paper describes operation at subatomic separation in the quantum-tunnelling regime of plasmons. The authors reported angstrom-scale spatial resolution. That is nearly three orders of magnitude smaller than the diffraction limit in optics, according to the paper’s abstract, though the abstract does not give a specific diffraction-limit distance.

What did the images show—and what did they not show?

The instrument measured a spatially localized Raman signal, then used the signal’s variation to image the molecule’s vibrational normal modes. The study also analyzed vibration-driven intramolecular charges and currents at atomic scale. These results reveal spatial structure in molecular vibrations; they are not a real-time movie of atoms moving.

This distinction matters: the achievement was not simply detecting that a molecule vibrates, but mapping the characteristic patterns of motion within one molecule. The setup was also not an ordinary optical microscope. Its result depended on the specialized metallic-tip junction inside a cryogenic ultrahigh-vacuum STM.

Why could atom-scale vibration maps matter?

Knowing where vibrational behavior is concentrated could help researchers investigate how molecules behave in more complex systems. Chemistry World’s 2019 coverage identified catalyst design and superconductivity as possible areas of relevance, not outcomes demonstrated by this experiment. The study established a measurement and imaging technique; it did not produce a new catalyst, a superconductor or a commercially available instrument. Chemistry World’s report by Tim Wogan was published 11 April 2019.

How does this differ from conventional vibrational spectroscopy?

Many vibrational spectroscopy methods characterize materials by measuring spectra, without producing an angstrom-scale map of each mode within one molecule. In this study, the spatially confined light and STM junction let the researchers pair vibrational spectra with a map of mode intensity at molecular scale. That comparison describes this particular demonstration; Raman spectroscopy in general does not require a cryogenic ultrahigh-vacuum STM.

The result is a specialist research technique rather than a consumer microscope or a ready-to-use laboratory kit. Its significance lies in the new spatial detail it made measurable, not in replacing spectroscopy as a whole.
