A 2026 University of Vienna study proposes that an ancient reorganization of coleoid cephalopod genomes—octopuses, squid and cuttlefish—helped create the regulatory environment needed for their complex nervous systems. The work focuses on how DNA is folded in three dimensions, not on a single “intelligence gene.” It identifies a possible evolutionary mechanism, but it does not prove that any one genomic rearrangement directly caused intelligent behavior.
What the study discovered
The researchers reconstructed three-dimensional genome organization in octopus, squid and cuttlefish and compared that structure with gene activity. According to the University of Vienna’s October 9, 2026 account, a large-scale genome reorganization hundreds of millions of years ago brought DNA regions that had previously been far apart into physical contact.
Those contacts can matter because genes are regulated not only by their linear DNA sequence. When the genome folds, distant regulatory elements can come close enough to influence one another and the genes they control. The researchers propose that repeated interactions gradually became incorporated into wider regulatory networks, a process they call regulatory entanglement.
Lead author Thea Rogers, PhD, described the central idea this way: “The genome isn’t just a sequence of genes. It’s folded into a complex three-dimensional structure.”

Why three-dimensional DNA matters
Every cell contains a long DNA molecule that must be compacted inside the nucleus. That compaction is organized rather than random. DNA winds around proteins, forms loops and gathers into larger chromatin domains. The resulting shape helps determine which regulatory sequences can communicate with which genes.

The contrast is important. Stable domains can provide a durable organizational framework, while changing loops offer opportunities to adjust gene regulation in particular tissues or at particular stages of development. The reported loops were frequently near genes associated with nervous-system development and function, although proximity alone does not demonstrate that a loop controls a specific behavior.

Regulatory entanglement is the team’s name for a proposed evolutionary process. Once an ancestral genome rearrangement places formerly distant regulatory elements in contact, changes affecting one element may influence the activity of another. Over many generations, those interactions could become embedded in interconnected gene-regulation networks.

This is different from adding a new gene with a simple, isolated function. It suggests that evolutionary innovation can arise partly from rewiring when and where existing genes are switched on. In cephalopods, such rewiring could have affected developmental programs and neural tissues, giving natural selection more regulatory combinations to work with.
How this relates to octopus intelligence
Octopuses have unusually complex nervous systems for invertebrates, and cephalopods show sophisticated learning, flexible behavior and extensive sensory processing. The new study addresses a possible genomic foundation for that complexity by examining DNA architecture across related coleoid species.

However, the study did not measure intelligence in an experiment, identify a single causal mutation or show that a particular chromatin loop produces a defined cognitive ability. Its evidence is comparative: genome structure was mapped, gene activity was examined and structural patterns were associated with genes involved in cephalopod biology. The appropriate conclusion is that three-dimensional genome organization may have helped shape the regulatory landscape from which complex nervous systems evolved.

Study identification and limits
The work is led by Thea F. Rogers and colleagues in the paper “Genome reorganisation and expansion shape 3D genome architecture and define a distinct regulatory landscape in coleoid cephalopods,” published in Nature Communications in 2026 (DOI 10.1038/s41467-026-77625-7). The University of Vienna’s press release is dated October 9, 2026. ScienceDaily published a report on October 12, 2026, crediting the university and giving the same citation.

The accessible institutional and secondary accounts establish the study’s central comparisons and interpretation, but they do not provide a named quantitative effect size or a complete methods description. The journal article should be consulted for detailed sampling, sequencing, statistical analyses and the specific species, tissues and developmental stages included.
Why the finding matters beyond octopuses
The study reinforces a broader shift in genetics: evolutionary change can involve not only which genes an organism possesses, but also how the genome is folded and how regulatory elements communicate. A rearrangement that changes three-dimensional contacts may alter many gene-expression relationships at once, potentially creating new combinations without requiring entirely new genes.
For cephalopods, that idea provides a testable route for future work. Researchers can investigate whether particular loops are required for neural development, whether disrupting them changes gene activity, and whether comparable contacts recur in independent cephalopod lineages. Those experiments would be needed to move from an evolutionary association to a demonstrated causal link with nervous-system traits or behavior.

The Bottom Line
The surprising twist is not a newly discovered “intelligence gene,” but the possibility that ancient genome rearrangements rewired three-dimensional DNA contacts. That architecture may have helped cephalopods evolve complex nervous systems, while direct causation between the rearrangements and octopus intelligence remains unproven.
