Saliva can be tested for molecules linked to inflammation, but the studies behind a proposed “spit test” do not establish a clinically validated way to diagnose heart disease or a heart attack. The work ranges from a 2005 laboratory assay for C-reactive protein (CRP) to later feasibility studies; it is not evidence that consumers can screen themselves at home.
What the proposed saliva test measures
The central biomarker in the early work is C-reactive protein (CRP), a marker of inflammation. Researchers have investigated whether CRP and other proteins can be measured in saliva and whether those measurements might eventually contribute to cardiovascular screening. Measuring a biomarker is not the same as showing that it can reliably identify heart disease in patients.
CRP is not specific to heart disease. Inflammation from other conditions can affect it, and the 2005 study also reported higher salivary CRP in people with chronic periodontal disease. A salivary CRP result by itself therefore cannot establish that someone has cardiovascular disease.

2005: a sensitive laboratory microchip assay
Christodoulides and colleagues used a microchip assay to measure CRP in human saliva. The paper reported sensitivity of 10 pg/mL with saliva diluted 1,000-fold. That is an assay measurement under the study’s laboratory conditions—not a validated threshold for diagnosing heart disease. The authors discussed potential relevance to chronic inflammatory conditions, including cardiovascular and periodontal disease. Read the 2005 study record on PubMed.

- Compact & Travel-Friendly: The ultra-compact design allows the adapter to stay plugged into your device without blocking adjacent ports or adding bulk, reducing wear and tear on your original USB ports.
- 12-Month Warranty: Backed by a 12-month manufacturer warranty for peace of mind. Designed to meet strict quality control standards for reliable everyday performance.
2009: a preliminary study involving acute heart attacks
A feasibility study measured 21 proteins in serum and unstimulated whole saliva from 41 patients with acute myocardial infarction and 43 apparently healthy controls. In that study’s sample and analysis, a salivary panel of CRP, myoglobin, and myeloperoxidase had an area under the curve (AUC) of 0.85; the panel combined with an electrocardiogram (ECG) had an AUC of 0.96. These figures describe the study results, not established performance for general screening or a stand-alone saliva test. The study does not show that a saliva result can replace clinical assessment. Read the full feasibility study.
2011: another exploratory CRP assay
A separate study evaluated a rapid salivary CRP assay in 55 healthy volunteers and 28 cardiac patients. The authors reported higher mean salivary CRP in the cardiac group and a correlation between paired serum and saliva results. They proposed further investigation for screening; those findings do not establish diagnostic accuracy for routine clinical use. Read the 2011 study abstract.

2024: microfluidic assay compared with ELISA
A 2024 paper described an integrated microfluidic assay for salivary CRP, interleukin-6 (IL-6), and procalcitonin. Its reported comparison with standard ELISA on real saliva samples had an R² of 0.952. That indicates agreement in the paper’s assay comparison; it does not, by itself, demonstrate that the test detects heart disease accurately in patients. The paper describes household cardiovascular screening as an opportunity, but the available evidence does not establish that the assay is commercially available or clinically authorized. Read the 2024 paper abstract.
Why these results do not add up to a home heart test
The studies ask different questions and use different methods. The 2005 result concerns analytical sensitivity for CRP in a laboratory assay. The 2009 study tested a multi-protein panel in a small acute myocardial infarction case-control sample, including a version combined with ECG. The 2011 work explored salivary CRP in cardiac patients and healthy volunteers. The 2024 paper compared a microfluidic assay with ELISA. Their figures are not directly comparable as if they came from one head-to-head clinical trial.

Clinical use requires more than detecting a molecule or matching another laboratory method. A test needs evidence that its results distinguish the relevant condition in the intended population and setting, along with appropriate clinical validation and authorization. The cited studies do not establish those points for a consumer saliva-based heart-disease test.
What this means if you are worried about heart disease
Do not use a saliva CRP result—or the absence of one—to decide whether you are having a heart attack or to rule out heart disease. The research described here is not a substitute for medical evaluation. If you have symptoms that could be a heart attack, seek emergency medical care rather than waiting for or relying on a saliva test.

How the idea began
A 2005 Sandia Lab News report described a portable diagnostic device as still under development. Project lead Anup Singh said the team had tested healthy saliva samples for gum disease and expected to begin testing diseased samples. That account documents early device development, not a finished cardiac diagnostic. Read Sandia’s January 7, 2005 report.
