A 2026 study found that mutations in DNA templates encoding self-assembling RNA nanostars can change the architecture of RNA droplets. Some mutant droplets develop long-lived internal vacuoles, move over micrometer distances for more than 50 hours, and partition tested cargo into dilute internal regions. They are engineered, cell-like laboratory structures—not living cells, autonomous swimmers, or medical products.
What the researchers built
The team started with custom DNA templates that encode RNA nanostars. As the templates are transcribed, the RNA folds and nanostars assemble into droplets through complementary “kissing-loop” interactions. The DNA template is encapsulated along with the RNA, physically linking a sequence to the structure it produces.
That genetic link makes the droplets experimentally evolvable: researchers can identify mutations in the DNA, observe the resulting phenotype, and test whether the altered sequence changes droplet architecture or behavior. The group used next-generation sequencing to identify mutant templates, reverse-engineered selected nanostars, and mixed mutant and non-mutated variants to examine how composition affects the resulting droplets.
How mutations create internal vacuoles
The notable mutant phenotype is a droplet containing one or more internal vacuoles. These are dilute regions inside an otherwise RNA-rich phase. They are not conventional membrane-bound organelles.

The paper attributes vacuole formation to altered interactions among nanostars and a local instability in the RNA-dense material. Unlike many short-lived vacuolated condensates, the reported vacuoles remained stable for days. In this system, a change in the encoded RNA sequence therefore produced a durable change in internal architecture.
How can mutations make an RNA droplet move?
Confocal time-lapse microscopy showed enhanced micrometer-scale movement in mutant droplets compared with wild-type droplets. The researchers associate that movement with vacuole formation, internal mass redistribution, increased encounters and fusion, and continuing changes in the droplet’s internal organization. Motion persisted for more than 50 hours in the reported experiment.

This is not persistent self-propulsion. Vacuoles appeared at essentially random locations, so the droplets did not maintain a stable front, rear, or propulsion axis. Calling them autonomous swimmers would overstate the result: their movement is an emergent consequence of internal rearrangement rather than directed locomotion.

What is an RNA droplet?
An RNA droplet is a concentrated, liquid-like compartment formed when RNA molecules interact and separate from the surrounding solution. In this study, the RNA nanostars provide the interaction network that makes the dense phase. The droplets resemble some organizational features of cellular compartments, but they do not contain the complete machinery of a living cell.

Can these synthetic structures store cargo?
The experiments showed selective, size- and interaction-dependent partitioning rather than unrestricted cargo loading.

Heidelberg University summarized the finding through Prof. Dr. Kerstin Göpfrich: “They take up small molecules and can store this cargo inside them.” In context, that statement refers to the synthetic RNA droplets and the specific probes tested, not to a general delivery platform. The results do not show that arbitrary cargo, therapeutic drugs, or whole vesicles can be loaded into the vacuoles.

Are these artificial cells alive?
No. The structures are cell-like experimental models. They reproduce selected properties—genetic encoding, compartmentalization, internal organization, movement and cargo partitioning—but the study does not demonstrate metabolism, reproduction, homeostasis, autonomous swimming or other criteria normally associated with living cells.

Why the genetic link matters
- Genetic control: The DNA template and RNA nanostar sequence are tied to the compartment phenotype.
- Architecture: Mutant sequences can produce long-lived vacuoles, while comparison droplets lack that internal structure.
- Motion: Movement emerges from vacuole dynamics and redistribution, not a persistent propulsion mechanism.
- Cargo partitioning: Size, solubility and molecular interactions determine whether material enters the dense phase, occupies a vacuole or associates with the interface.
- Evolutionary testing: Sequencing and reconstruction let researchers connect particular mutations with physical outcomes.
How this differs from other programmable RNA droplets
A separate 2026 study used ribozyme sequences in RNA nanostars to program droplet dissolution and segregation. That work illustrates another form of RNA-based control, but it is not the same mechanism as the mutation-driven vacuoles and movement described here. The present study focuses on emergent architectural and dynamic changes arising from sequence mutations and altered nanostar interactions.
What could come next—and what has not been shown
The platform may help scientists investigate how synthetic compartments evolve new functions and may eventually inform more elaborate synthetic-cell designs. Drug delivery is a prospective direction, not an outcome demonstrated by this work. No treatment, clinical intervention, consumer product or ready-to-use delivery system resulted from the experiments.
The findings are also experiment-specific. The reported persistence, cargo behavior and movement apply to the constructs, mixtures, probes and imaging conditions used by the researchers; they are not general performance guarantees for every RNA droplet.

The work was reported in a 2026 Nature Chemical Engineering paper titled “Genetic encoding and mutagenesis of RNA droplet phenotypes.” Heidelberg University’s announcement, dated 13 October 2026, identifies Prof. Dr. Kerstin Göpfrich’s group at the university’s Center for Molecular Biology, with collaborators at the German Cancer Research Center and the University of Augsburg.
