Use an optical dissolved-oxygen sensor for rapid profiles, autonomous deployments, and continuous seawater records. Use Winkler iodometric titration when you need a chemically traceable reference or a calibration check. Neither method is universally more accurate. Results depend on calibration, sampling integrity, salinity and temperature corrections, pressure and depth metadata, chemical interferents, and the quality-assurance plan. The strongest seawater workflow commonly pairs them: log the sensor continuously, collect bubble-free Winkler samples at representative depths, and quantify any sensor bias.

What the two methods actually measure

Optical oxygen sensors: fluorescence quenching

ISO 17289:2014 defines optical dissolved-oxygen measurement as a sensor method based on fluorescence quenching. A dye or sensing layer is excited by light; dissolved oxygen reduces (quenches) the emitted fluorescence. The instrument converts that change into oxygen concentration, percent saturation, or both.

Because the probe produces a digital reading immediately, it can be mounted on a CTD package, lowered through a water column, installed on a mooring, or carried on an autonomous platform. ISO 17289 covers field, laboratory, and continuous monitoring applications. The instrument still needs valid temperature, pressure, and salinity inputs, and its calibration must match the conditions in which it is deployed.

Winkler titration: an iodometric reference procedure

Winkler analysis fixes the oxygen chemically in a completely filled, bubble-free bottle. Reagents form an iodine equivalent to the oxygen originally dissolved in the sample. The iodine is then titrated, and the endpoint gives the oxygen concentration.

ISO 5813:1983 identifies the modified Winkler procedure as the reference method for water above 0.2 mg/L and up to twice saturation when interfering substances are absent. USGS guidance describes Winkler as accurate and precise under controlled laboratory conditions. NOAA’s seawater protocol uses Carpenter’s modification of Winkler chemistry and reports dissolved oxygen in micromoles of O2 per kilogram of seawater.

Is one method more accurate?

There is no universal winner. A carefully collected and titrated Winkler sample is the reference against which many sensors are checked, but a damaged probe, a mismatched calibration, or incorrect salinity entry can make an optical result wrong. Conversely, a Winkler result can be biased by a bubble introduced during sampling, delayed fixation, contaminated or poorly standardized reagents, an indistinct endpoint, or chemical interferents.

Think of accuracy as a measurement-system property rather than a brand or technology label. For a shipboard survey, the sensor may provide the most representative picture because it records every depth and avoids the gaps between discrete bottles. For a laboratory reference value, Winkler can provide stronger chemical traceability if the sample and titration are controlled. A paired design reveals which limitation dominates at your site.

Side-by-side comparison

Measurement consideration Optical sensor Winkler titration
Principle Fluorescence quenching measured by an oxygen-sensitive sensor. Iodine formed during chemical fixation is titrated to an endpoint.
Deployment Immediate readings, vertical profiles, autonomous operation, and continuous logging. Discrete bottles followed by fixation and titration; no continuous record.
Primary role Operational measurement, profiling, and long-term monitoring; verify against discrete samples. Reference or calibration check when controlled chemical handling is possible.
Outputs Concentration in mg/L, percent saturation, or both, depending on instrument configuration. Concentration calculated from the titration; seawater protocols commonly report µmol O2/kg.
Routine needs Calibration, battery and logger care, correct temperature/pressure/salinity inputs, and periodic sensor-cap replacement. Oxygen bottles, reagents, titration equipment, waste handling, fresh standardized solutions, and trained endpoint work.
Important interferences Reactive gases, salinity differences, calibration mismatch, and degraded membrane or sensing components. Iodine-fixing substances, hydrogen sulfide, bubbles, delayed fixation, and endpoint or reagent errors.
Low-end performance cited in standards Detection limits of 0.1 or 0.2 mg/L are reported as achievable, depending on the optical instrument manual. The ISO 5813 reference range starts above 0.2 mg/L when interferents are absent.
High-oxygen conditions ISO 17289 discusses supersaturation of 200% and more; handling must prevent oxygen loss from supersaturated samples. Applicable up to twice saturation under the ISO 5813 conditions.

Seawater corrections and handling that determine the result

Enter salinity before reporting concentration

Oxygen solubility changes with salinity. An optical instrument can display a plausible number while still reporting the wrong concentration if it is configured for freshwater or the wrong salinity. Enter measured salinity or a validated conductivity-derived value before converting the sensor signal to concentration. Keep the salinity record with every profile and bottle result.

Percent saturation is also condition-dependent. Record the temperature, salinity, atmospheric pressure or hydrostatic pressure used by the calculation, and the convention used by the instrument or data-processing software. Do not compare a sensor’s uncorrected mg/L value with a Winkler result reported in µmol/kg without documenting the conversion and assumptions.

Preserve the sample’s oxygen state

ISO 17289 warns that supersaturated water can lose oxygen during handling. Water above 100% saturation must not be shaken, splashed, or left with a headspace. Undersaturated water can instead gain oxygen from air. Fill bottles without bubbles, cap them promptly, and keep the handling sequence consistent between sensor observations and discrete samples.

Watch for hydrogen sulfide and other chemical interferents

NOAA’s seawater Winkler protocol states that its method is unsuitable for seawater containing hydrogen sulfide. Winkler chemistry can also be affected by substances that fix iodine. If sulfide or another reactive constituent is expected, document it and do not treat an apparently precise titration as a valid reference without a method-specific interference assessment. Optical probes can also respond incorrectly in unusual reactive-gas environments or when their sensing components are fouled or damaged.

Which method fits your project?

Shipboard surveys and estuary transects

Use an optical probe on the CTD or profiling package so oxygen is recorded continuously with depth. Collect Winkler bottles at selected depths, especially near the surface, the oxycline, the deepest point, and any location where the sensor trace changes abruptly. The bottles provide an independent check without sacrificing the spatial coverage of the profile.

Autonomous platforms and moorings

An optical sensor is the practical choice for an autonomous deployment because it can log repeatedly without a person present. Before launch, verify the sensor’s calibration, clock, battery, memory, pressure rating, and salinity input. Include a deployment and recovery plan for sensor-cap inspection and post-deployment comparison with Winkler samples or another validated reference.

Laboratory reference work

Choose Winkler when the project requires a documented iodometric procedure and controlled handling. Prepare fresh, standardized reagents; use clean oxygen bottles; fix immediately; and perform duplicate or replicate titrations. A laboratory that needs many time-resolved observations may still use an optical instrument, but it should establish agreement with Winkler over the relevant oxygen and salinity range first.

Routine field screening

A Winkler field kit is useful when the sampling team needs an inexpensive, self-contained chemical check and can perform careful fixation and titration. EPA guidance gives a typical price of $35–$200 for commercial Winkler dissolved-oxygen field kits, with approximately 50–100 tests per kit. Those figures describe the kit category and its stated capacity, not a guaranteed total project cost; replacement reagents, waste disposal, training, and repeat samples add to the budget.

How to calibrate and validate an optical sensor with Winkler samples

Use paired observations rather than treating a single bottle as a universal calibration. EPA’s CTD quality-assurance approach compares sensor readings with Winkler values at representative depths and oxygen conditions.

  1. Define the comparison range. Select stations and depths that cover the salinity, temperature, pressure, and oxygen range the sensor will encounter. Include low-oxygen water and, where relevant, supersaturated surface water.
  2. Prepare and document the sensor. Complete the manufacturer’s calibration procedure, inspect the sensing cap or membrane, check batteries and logging settings, and enter the salinity and pressure conventions used by your project. Record calibration date, standards, and any replacement parts.
  3. Stabilize the profile measurement. Allow the probe to equilibrate at each target depth according to the instrument’s operating procedure. Record the raw reading, temperature, pressure, salinity, depth, time, and instrument identifier rather than retaining only a converted final value.
  4. Collect the Winkler bottle at the same water mass. Trigger the sampler at the sensor depth or collect immediately adjacent to the probe. Fill the bottle completely, allow sufficient overflow, and inspect for bubbles before capping.
  5. Fix the sample immediately. Add the prescribed fixation reagents without introducing air, mix as specified by the method, and protect the fixed sample from conditions that could alter the iodine chemistry. Note any delay or visible bubble.
  6. Perform duplicate or replicate titrations. Use fresh standardized reagents, a clean burette or titrator, and a defined endpoint procedure. Record titrant concentration, volume, endpoint observations, replicate spread, and any blank correction required by the laboratory method.
  7. Compare on the same basis. Convert units only after documenting the seawater density, salinity, temperature, pressure, and oxygen convention used. Compare the sensor and Winkler result from the same depth and time, not a bottle collected hours later.
  8. Diagnose before adjusting. Plot sensor-minus-Winkler difference against depth, oxygen level, temperature, and salinity. A constant offset suggests calibration bias; a depth- or pressure-dependent difference suggests pressure compensation, equilibration, or metadata problems; scattered differences point to sampling or titration variability.
  9. Apply and verify any correction. Adjust the sensor only within the range supported by the paired data and the instrument’s calibration model. Recheck with independent Winkler samples after adjustment, and preserve the original readings and correction history.

An IOCCP oxygen-measurement handbook reports representative Winkler-based calibration precision of 2 µmol/L in a specific context. Treat that figure as an example of achieved precision under stated conditions, not as a universal accuracy specification for every sensor, bottle, or seawater sample.

How to run a defensible Winkler measurement

  1. Use the correct bottle. Select a clean, calibrated oxygen bottle with a stopper that seals without trapping a headspace.
  2. Fill from the sampling device. Let water overflow enough to replace the bottle volume, keep the outlet below the surface of the sample, and avoid turbulence, splashing, and bubbles.
  3. Inspect before fixation. Reject or flag any bottle with a visible bubble, leak, incomplete overflow, or unexplained delay.
  4. Add fixation reagents in the prescribed order. Insert the reagent delivery tip below the surface where possible, cap immediately, and mix without creating foam. Follow the chosen standard’s reagent concentrations and reaction times.
  5. Complete the iodine reaction. Allow the precipitate and acidification steps to proceed as specified. Keep samples protected from unnecessary heat and light while waiting for titration.
  6. Titrate to the defined endpoint. Use a standardized thiosulfate solution and the endpoint technique specified by your method. Record all volumes and perform duplicates when the result will be used to assess sensor bias.
  7. Check chemistry suitability. Flag samples containing hydrogen sulfide or suspected iodine-fixing substances; the NOAA seawater protocol is not suitable for hydrogen-sulfide-containing seawater.

Optical-sensor setup and maintenance

  • Calibration: Follow the instrument’s zero and air- or water-based calibration procedure, and record the standard, temperature, salinity, and date.
  • Salinity and pressure: Confirm that the probe is receiving the measured salinity and the correct pressure or depth input, not a default freshwater or surface setting.
  • Temperature response: Let the probe equilibrate and verify that the temperature channel is functioning; oxygen solubility and percent saturation calculations depend on it.
  • Sensing surface: Inspect for scratches, biofouling, coating damage, or a depleted cap. Replace the sensor cap at the interval specified for that instrument and deployment conditions.
  • Logging: Synchronize clocks, select a sampling interval appropriate to the profile speed or mooring dynamics, and retain raw data alongside processed concentration and saturation channels.
  • Post-deployment check: Compare recovered readings with Winkler samples or another validated reference before accepting a long deployment as a continuous record.

Common failure modes and fixes

The probe and Winkler result disagree by a nearly constant amount

Check calibration date, salinity entry, unit conversion, and pressure compensation first. Then inspect the sensor cap and compare several paired samples. Do not erase the offset by recalibrating against one bottle until sampling and titration quality are confirmed.

Differences increase with depth

Review pressure input, equilibration time, profiling speed, and CTD-to-bottle depth alignment. A bottle triggered above or below the probe’s logged position is not a valid point-for-point comparison.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Replicate Winkler titrations are widely scattered

Look for bubbles, incomplete fixation, inconsistent endpoint recognition, contaminated glassware, or unstandardized reagent. Resolve the titration repeatability problem before using those values to correct a sensor.

Surface readings exceed 100% saturation

Supersaturation can be real, particularly during strong algal growth; ISO 17289 discusses values of 200% and more. Prevent outgassing during bottle collection and verify that temperature, salinity, atmospheric pressure, and sensor calibration are correct before dismissing the result as an error.

The Winkler result looks normal in sulfide-containing water

Do not assume a normal-looking endpoint proves validity. NOAA identifies hydrogen sulfide as a condition that makes its seawater Winkler protocol unsuitable. Flag the sample and select a method validated for the interference.

What to report with seawater oxygen data

  • Method and standard: optical fluorescence-quenching measurement under ISO 17289:2014, modified Winkler under ISO 5813:1983, or the specific seawater protocol used.
  • Instrument identifier, sensor-cap or membrane condition, calibration date, calibration standards, and any applied correction.
  • Sampling date and time, latitude and longitude or station, depth, pressure, temperature, and salinity.
  • Result units: mg/L, percent saturation, or µmol O2/kg, including the conversion convention when values are transformed.
  • For Winkler samples, bottle type, fixation timing, reagent standardization, titrant information, replicate results, endpoint method, and blank treatment.
  • Quality flags for bubbles, delays, sulfide, suspected iodine-fixing substances, sensor fouling, out-of-range values, or failed replicates.
  • The paired sensor-versus-Winkler difference and the rule used to accept, reject, or correct sensor data.

Practical decision

Choose optical sensing when coverage, immediacy, and unattended logging matter. Choose Winkler when you need a chemically traceable reference and can control sampling and titration. For high-consequence seawater measurements, deploy the optical probe and collect Winkler checks at representative depths and oxygen levels. That combination gives you continuous observations plus an independent test of calibration, while making salinity, temperature, pressure, unit, and interference assumptions visible.
