A 2026 study used synchronized two-photon fluorescence lifetime imaging (FLIM) and phosphorescence lifetime imaging (PLIM) to measure metabolism and vascular oxygenation at the same time in the living cortex of an Alzheimer’s disease mouse model. The model showed a higher protein-bound NAD(P)H fraction and lower vascular oxygen partial pressure than wild-type mice—findings the researchers interpreted as altered mitochondrial activity alongside greater oxygen use.

What the study measured

The work, published 11 September 2026 in Neurophotonics, combined two lifetime-imaging measurements that are usually performed separately. V. K. Sagar and colleagues examined anesthetized mice at rest, comparing an Alzheimer’s disease model with wild-type controls. Measurements were made in the living cerebral cortex rather than in fixed tissue or a dish.

FLIM supplied a readout of cellular metabolism from endogenous NAD(P)H, while PLIM estimated oxygen in cortical blood vessels with an oxygen-sensitive ruthenium probe. Collecting both signals in the same preparation allowed the investigators to examine metabolic state and oxygen availability together.

How FLIM turns NAD(P)H fluorescence into a metabolic readout

Lifetime, not brightness, is the key signal

NAD(P)H is naturally fluorescent. After excitation, it remains in an excited state for a measurable lifetime before emitting light. The lifetime differs when NAD(P)H is free in the cell versus bound to proteins, including enzymes involved in mitochondrial energy production. FLIM records those lifetimes across the imaged cortex and estimates the protein-bound fraction.

What changed in the Alzheimer’s model

The Alzheimer’s-model cortex had a higher protein-bound NAD(P)H fraction than the wild-type cortex. The researchers treated that directional difference as evidence of altered mitochondrial metabolic activity. It does not identify a single biochemical defect, nor does it by itself establish whether total energy production increased or decreased; it indicates that the balance of free and protein-associated NAD(P)H shifted.

How PLIM estimates oxygen in brain vessels

Why a ruthenium probe is needed

Unlike FLIM’s endogenous NAD(P)H signal, PLIM in this experiment used a ruthenium-based oxygen-sensitive probe. The probe emits phosphorescence whose lifetime changes with oxygen concentration. After calibration, that lifetime can be converted into an estimate of vascular oxygen partial pressure.

The oxygen result

Vascular oxygen partial pressure was lower in the Alzheimer’s-model cortex than in wild-type animals. The authors interpreted lower vessel oxygen as consistent with greater oxygen consumption by the surrounding tissue: more oxygen extracted from blood would leave less oxygen in the vasculature. That interpretation is physiologically plausible, but the measurement does not prove the cause. Lower oxygen could also reflect changes in blood flow, delivery, vascular regulation or other disease-related processes.

Why combining FLIM and PLIM matters

Metabolic fluorescence and tissue oxygenation are often studied with different instruments or in separate experiments. Simultaneous FLIM-PLIM places the two readouts in the same living brain, at the same time and under the same physiological conditions. That makes it possible to ask whether a metabolic shift coincides with a change in oxygen availability instead of trying to align results collected on different days or from different animals.

The synchronization is technically demanding. According to investigator Periasamy, simultaneous measurement requires precise timing between a Zeiss two-photon microscope and Becker & Hickl lifetime-imaging electronics. The optical signals also have different physical origins and calibration requirements: FLIM separates NAD(P)H lifetime components, whereas PLIM requires an oxygen-sensitive probe and a lifetime-to-oxygen calibration.

What the findings do—and do not—show

  • They show a linked pattern in mice: the Alzheimer’s model had a higher protein-bound NAD(P)H fraction and lower vascular oxygen partial pressure than wild-type controls.
  • They support a mechanistic interpretation: the combination is consistent with altered mitochondrial metabolism and increased oxygen use in the model cortex.
  • They do not establish human disease physiology: the experiment was preclinical and used an Alzheimer’s mouse model, not people with Alzheimer’s disease.
  • They do not provide a clinical diagnostic test: the report supplies directional group differences but no numeric effect sizes, sample sizes, confidence intervals or p-values.

Because the animals were anesthetized and measured at rest, the observations describe that controlled state. Anesthesia, baseline activity and the specific mouse model can all influence metabolism, blood flow and oxygenation. The study therefore presents an imaging framework and a biological association, not a measurement that can yet be transferred directly to patients.

PLIM alone No direct metabolic measurement Yes, through probe lifetime Ruthenium oxygen-sensitive phosphorescence probe Probe calibration and delivery Used to estimate vascular oxygen partial pressure
Simultaneous FLIM-PLIM Yes Yes Endogenous NAD(P)H plus an exogenous ruthenium probe Optical and electronic synchronization, lifetime calibration Demonstrated in an Alzheimer’s mouse model and wild-type controls
Human clinical imaging Not measured by this experiment Not measured by this experiment Not applicable Would require a validated, noninvasive clinical protocol Not established by the report

Two-photon imaging is well suited to experiments in exposed living cortex because it can resolve signals from cells and nearby vessels while restricting excitation to the focal plane. The study does not state numerical spatial or temporal resolution, probe dose, animal numbers or calibration ranges, so those specifications should not be inferred from the publication summary.

What researchers need to reproduce the measurement

  1. Prepare a living cortical window: the experiment requires optical access to the cortex and physiological control of the animal.
  2. Acquire the FLIM signal: excite endogenous NAD(P)H with the two-photon microscope and record fluorescence lifetimes suitable for separating free and protein-bound components.
  3. Introduce and calibrate the oxygen probe: use the ruthenium probe’s phosphorescence lifetime response to establish the relationship between lifetime and oxygen partial pressure.
  4. Synchronize acquisition: coordinate the microscope timing with the lifetime electronics so FLIM and PLIM data correspond to the same moment and location.
  5. Analyze paired readouts: compare the protein-bound NAD(P)H fraction with vascular oxygen estimates while accounting for anesthesia, rest state and the chosen disease model.

The practical advance is the paired measurement itself. It gives preclinical researchers a way to test whether interventions change metabolism, oxygen use or both in the same tissue. Establishing whether the pattern occurs in human Alzheimer’s disease—and whether it tracks symptoms, progression or treatment response—will require separate clinical studies.
