Nanoparticle drug delivery is not one competing platform. “Nanoparticle” is an umbrella term that includes lipid nanoparticles, polymeric nanoparticles, micelles, inorganic particles and hybrid systems. Liposomes are lipid-membrane vesicles within the broader nanoscale delivery landscape, while polymer carriers use engineered polymers in particles, micelles or related structures. No architecture is universally best: the appropriate choice depends on the drug or genetic cargo, where it must go, how it should be released, its stability and storage requirements, manufacturing constraints, safety profile and the clinical evidence for the exact formulation.

What “nanoparticle drug delivery” means

A nanoparticle drug-delivery system is a nanoscale carrier designed to transport a therapeutic payload through the body and, in some cases, into a particular cell compartment. The label describes size and function more than a single material. A formulation may be made from lipids, polymers, inorganic substances or combinations of these materials. Two products can both be called nanoparticles yet behave very differently because their composition, size distribution, surface coating, electrical charge, shape, drug-loading method and release chemistry differ.

That broad category is why a simple “nanoparticles versus liposomes” comparison can mislead. Liposomes and many lipid nanoparticles are themselves nanoscale particles. A meaningful comparison identifies the architecture first, then asks how it handles the intended cargo and biological destination.

Main architectures covered by the term

  • Liposomes: spherical vesicles bounded by one or more phospholipid bilayers.
  • Lipid nanoparticles (LNPs): lipid-based particles with an internal organization that is not necessarily a classic aqueous-core liposome; ionizable LNPs are widely used for nucleic acids.
  • Polymeric nanoparticles: particles made from biodegradable or non-biodegradable polymers that can entrap, dissolve or chemically bind a drug.
  • Polymeric micelles: self-assembled structures with a hydrophobic core and a hydrophilic shell, often used for poorly water-soluble molecules.
  • Inorganic and hybrid particles: systems using materials such as metals, silica or combinations of lipid and polymer components; their suitability depends heavily on material-specific toxicity and clearance.

How liposomes differ from polymer carriers

Liposomes: a lipid bilayer around an aqueous space

A liposome has an aqueous interior surrounded by a lipid bilayer. Hydrophilic drugs can be held in the aqueous core, while hydrophobic drugs can partition into the bilayer. This dual compartment is the central reason liposomes can accommodate drugs with different chemical properties. Surface lipids can also be modified with polymers or ligands to alter circulation time, interactions with proteins and uptake by cells.

The flexibility is not automatic performance. Entrapment efficiency, leakage, particle size, lamellarity, oxidation of lipids and the exact surface composition all influence the finished medicine. A liposome that works for one molecule may be unsuitable for another, even when both products use the same broad architecture.

Polymer carriers: a tunable material framework

Polymeric nanoparticles and micelles use polymers to create a drug-containing matrix, core or shell. A drug may be dissolved in the polymer, physically entrapped, adsorbed to the surface or linked through a cleavable chemical bond. Polymer selection and molecular weight can change degradation rate, mechanical properties, drug diffusion and interactions with immune cells.

Polymer carriers can be engineered for slow release, environmental responsiveness or protection of unstable molecules. They are not categorically safer, more stable or more effective than liposomes. Those claims require a matched comparison using the same payload, dose, route, particle characteristics and clinical objective.

Liposomes, lipid nanoparticles and polymer nanoparticles are not synonyms

Liposomes and LNPs are related because both use lipids, but they are distinct design families. A classical liposome is defined by its bilayer vesicle structure. An ionizable LNP typically contains an ionizable lipid blended with helper lipids, cholesterol and a lipid-polymer conjugate. Its internal organization and behavior are optimized for transporting nucleic acids, especially through uptake by cells and release from endosomes.

The distinction matters when interpreting examples. Onpattro (patisiran) is an ionizable LNP formulation for small-interfering RNA in hereditary transthyretin amyloidosis; the cited review lists its U.S. approval in 2018. That approval validates an LNP approach for a particular siRNA product, not every lipid nanoparticle or every genetic medicine.

mRNA vaccines are another prominent clinical use of LNP technology. Their success demonstrates that an LNP can protect fragile RNA, support cellular delivery and be manufactured at scale under demanding quality controls. It does not establish that an LNP is the best vehicle for a hydrophobic small molecule, a protein or a long-acting injectable.

Payload or requirement Liposomes Polymer carriers Lipid nanoparticles
Hydrophilic small molecules Can be retained in the aqueous core; leakage and osmotic effects must be controlled. Possible through matrix entrapment or chemical conjugation, but loading depends on polymer–drug compatibility. Possible, although formulation is usually selected for a specific lipid composition and release profile.
Hydrophobic small molecules Can partition into the lipid bilayer. Often well suited to hydrophobic cores or polymer matrices. Possible, but not automatically advantageous compared with a liposome or polymeric system.
Nucleic acids Can be associated with charged lipids or encapsulated, but formulation-specific barriers remain. Can be complexed or conjugated with polymers; endosomal escape and toxicity must be demonstrated. Ionizable LNPs are a clinically validated architecture for siRNA and mRNA delivery.
Proteins and peptides May protect a protein in an aqueous compartment, subject to leakage and denaturation risks. Can provide protection and sustained release, but processing stresses and polymer degradation products require evaluation. Suitability is highly formulation-specific and is not implied by the success of RNA products.

The table describes design possibilities, not guaranteed performance. Loading percentage, dose capacity and release kinetics must be measured for the actual formulation.

How loading and release are engineered

Loading a liposome

Manufacturers may load a drug during vesicle formation or use an active-loading process driven by a transmembrane pH or ion gradient. Passive loading is simpler but can produce lower and more variable encapsulation for some drugs. Active loading can improve retention when the molecule’s ionization behavior supports precipitation or trapping inside the vesicle. The method affects free-drug content, batch consistency and the risk of premature leakage.

Loading a polymer carrier

Polymeric systems can be produced by nanoprecipitation, emulsion-based methods, solvent evaporation, ionic gelation or chemical conjugation. A hydrophobic drug may dissolve in the polymer phase; a hydrophilic drug may require a water-in-oil-in-water process or a covalent linker. Residual solvent, particle aggregation and uneven drug distribution are practical manufacturing concerns.

Loading an LNP

For nucleic acids, rapid mixing of an acidic aqueous nucleic-acid solution with an ethanolic lipid solution promotes encapsulation as particles form. The ionizable lipid is positively charged under acidic manufacturing conditions, helping associate with negatively charged RNA, and becomes less charged near physiological pH. That charge transition is a design feature, not a universal property of all lipid particles.

Controlling release

Release can be governed by diffusion, membrane permeability, polymer degradation, erosion, pH sensitivity, enzymatic cleavage or a combination of mechanisms. A long circulation time is not inherently beneficial if the payload must reach a rapidly dividing tumor or if delayed release reduces the active concentration at the target. Conversely, a rapid burst may be useful for a vaccine but undesirable for a toxic cytotoxic drug.

Stability, storage and physical properties

Particle stability includes more than whether the drug remains chemically intact. A product must maintain size, polydispersity, morphology, surface composition, encapsulation and potency during its labeled storage period. Liposomes can suffer from oxidation, hydrolysis, leakage, fusion and changes in membrane rigidity. Polymers can hydrolyze, crystallize, swell or release acidic degradation products. LNPs can be sensitive to lipid oxidation, hydrolysis, aggregation and repeated freeze–thaw cycles.

Surface chemistry changes the protein corona that forms after injection. Polyethylene glycol (PEG) or another hydrophilic coating can reduce aggregation and slow uptake by some clearance cells, but it may also reduce cellular uptake or contribute to anti-PEG immune reactions in susceptible settings. Particle size and charge influence filtration by the kidneys, uptake by the liver and spleen, vascular passage, and interactions with cell membranes. These relationships are continuous rather than fixed cutoffs; a size that works for one route or organ may be unsuitable for another.

Storage requirements must be taken from the formulation’s validated product information. A platform-level statement such as “liposomes are more stable” or “polymers are shelf-stable” is not reliable because excipients, concentration, container, sterilization method and temperature all matter.

Circulation, biodistribution and “targeting”

After administration, a carrier encounters proteins, immune cells, blood flow, vascular barriers and clearance organs. The liver and spleen commonly contribute to uptake of circulating particles, while renal elimination is more important for very small species or released drug. Surface charge can promote cellular association but also increase nonspecific protein binding and toxicity. Neutral or shielded surfaces may circulate longer but enter target cells less efficiently.

Passive accumulation and the EPR effect

Nanomedicine discussions often describe passive tumor targeting through the enhanced permeability and retention (EPR) effect: abnormal tumor vessels may allow some particles to leave the bloodstream, while poor lymphatic drainage can retain them. Human tumors vary substantially in vascular permeability, stromal structure, blood flow and immune infiltration. The review literature emphasizes that EPR is inconsistent and has important translational limits, so it should not be presented as guaranteed tumor targeting in patients.

Active targeting

An antibody, peptide, sugar or other ligand can be attached to a carrier to recognize a cell-surface feature. Binding does not ensure delivery to the desired intracellular compartment. The ligand must remain accessible in blood, the target must be expressed at adequate density, the particle must reach the tissue and the internalization route must release the payload. A ligand can also increase off-target uptake or accelerate clearance.

Safety and immune response

Safety is determined by the complete product, including carrier, payload, impurities, degradation products and route of administration. Potential concerns include complement activation, infusion reactions, cytokine release, liver or spleen accumulation, local inflammation, genotoxicity from the payload and toxicity from persistent materials. A biodegradable polymer is not automatically harmless, and a naturally occurring lipid is not automatically non-immunogenic.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Dose, repeat administration and disease state matter. A carrier tolerated once may trigger antibodies or altered clearance after repeated dosing. For nucleic-acid products, endosomal escape is necessary for activity but can disturb membranes and activate innate immune sensors. For cytotoxic drugs, reducing free-drug exposure may protect healthy tissue while still causing carrier-related effects or delayed toxicity.

Clinical examples: different jobs, not a league table

Product Architecture and payload What it demonstrates What it does not prove
Doxil PEGylated liposomal doxorubicin; the cited reviews list U.S. FDA approval in 1995 and use in ovarian cancer and Kaposi’s sarcoma. A liposomal formulation can become an established medicine for a conventional cytotoxic drug. That all liposomes improve outcomes or have the same safety profile.
Onpattro (patisiran) Ionizable LNP carrying siRNA for hereditary transthyretin amyloidosis; the cited review lists approval in 2018. An LNP can protect and deliver siRNA in a clinical product. That an LNP is the best carrier for every RNA, small molecule or protein.
Vyxeos (CPX-351) Liposomes carrying daunorubicin and cytarabine at a fixed 5:1 molar ratio. A liposome can preserve a defined combination-drug ratio, a key clinical proof of concept for coordinated exposure. That maintaining a ratio is useful for every combination or that the same ratio applies outside this product.

One 2022 review survey counted 14 types of liposomal products authorized by the FDA and EMA within its stated scope. That count excluded generics, lipid complexes and some nationally authorized European products, so it should not be treated as a complete current global total.

Manufacturing and scale-up considerations

Clinical feasibility depends on reproducible manufacturing as much as on elegant laboratory design. Critical quality attributes may include particle size and distribution, morphology, drug or RNA content, encapsulation efficiency, residual solvent, sterility, endotoxin, potency and release rate.

Liposome manufacturing challenges

  • Maintaining consistent size and lamellarity across batches.
  • Removing unencapsulated drug without losing vesicles.
  • Controlling lipid oxidation, hydrolysis and PEG-lipid distribution.
  • Choosing sterilization and filling methods that do not rupture or aggregate vesicles.

Polymer-carrier challenges

  • Controlling polymer molecular weight, composition and residual monomer.
  • Keeping solvent removal, particle formation and drug distribution consistent at larger scale.
  • Managing degradation products and changes in release during storage.
  • Demonstrating that a formulation process remains equivalent when mixing and heat-transfer conditions change.

LNP manufacturing challenges

  • Controlling rapid-mixing conditions, lipid ratios and RNA-to-lipid ratio.
  • Removing ethanol and unencapsulated nucleic acid while preserving particle quality.
  • Maintaining potency through filtration, filling, freezing and thawing.
  • Establishing assays that measure encapsulation, integrity and biological activity rather than size alone.

Which delivery system is best for a particular drug?

There is no platform-independent winner. Use a formulation decision process that starts with the therapeutic objective and eliminates architectures that cannot meet it.

  1. Define the cargo. Record molecular weight, solubility, pKa, chemical liabilities, dose, required concentration and whether the payload is a small molecule, protein, peptide, DNA, mRNA or siRNA.
  2. Define the destination. Specify the tissue, cell type and intracellular compartment that must receive the active payload. A carrier for liver hepatocytes has a different problem from one for a solid tumor or a central-nervous-system target.
  3. Choose the required exposure profile. Decide whether the medicine needs rapid release, sustained release, a fixed combination ratio, repeat dosing or protection from degradation during circulation.
  4. Screen compatible architectures. Compare a liposome, LNP, polymeric nanoparticle, micelle or hybrid design against loading capacity, leakage, endosomal escape, degradation and route-specific constraints.
  5. Measure the full particle profile. Characterize size distribution, surface charge, morphology, encapsulation, free drug, release kinetics, stability, potency and impurities under relevant conditions.
  6. Test biological behavior in stages. Examine serum interactions, cell uptake, intracellular release, toxicity and immune activation before moving to disease models. Do not infer human targeting from a single cell line or mouse tumor.
  7. Evaluate manufacturability early. A formulation that works only in a small laboratory batch may fail on scale-up, sterilization, filling or storage. Include process capability and analytical methods in the selection decision.
  8. Match the evidence to the claim. Use clinical data for the exact product and indication. Evidence that one liposomal or LNP medicine works cannot be transferred automatically to another carrier or payload.

Common comparison mistakes

Treating “nanoparticle” as a material

Ask what the particle is made of and how it is organized. “Nanoparticle” alone cannot predict loading, degradation or biodistribution.

Assuming smaller is always better

Reducing size can alter circulation and tissue penetration, but it may also reduce loading, change clearance or make the particle less stable. The useful size range depends on route, cargo and target.

Calling a product “targeted” without qualification

Separate passive accumulation, ligand-mediated binding, cell uptake and intracellular delivery. Each is a different claim requiring different evidence.

Using a platform’s success as a universal proof

Doxil, Onpattro and Vyxeos demonstrate feasibility for defined formulations and clinical jobs. They do not rank all liposomes, LNPs or polymer carriers.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Ignoring the free-drug fraction

Unencapsulated payload can dominate early exposure and toxicity even when the nominal formulation is highly loaded. Measure free and carrier-associated drug separately.

Comparing only laboratory potency

A higher cell-culture signal may reflect nonspecific membrane damage, different exposure time or a concentration that cannot be achieved safely in patients. Compare matched dose, exposure, controls and clinically relevant models.

What evidence is needed before a clinical claim?

For a product-specific decision, verify the current regulator-approved label in the relevant jurisdiction for indication, contraindications, warnings, dosing, storage and administration. The examples above are educational illustrations of architecture and clinical use; they are not prescribing advice. Regulatory status and labels can change, and a high-level review does not replace the current primary label.

A robust comparison should report the exact formulation, route, dose, particle attributes, comparator, disease model, endpoint and follow-up period. It should also distinguish surrogate measures such as tissue concentration from clinical outcomes such as survival, symptom control or reduced adverse events.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Bottom line: choose the formulation, not the label

Liposomes offer a mature vesicle architecture with clinical precedents for small-molecule and combination drugs. Ionizable LNPs are validated for nucleic-acid medicines, including siRNA and mRNA applications. Polymeric nanoparticles and micelles provide a separate, highly tunable family for loading and release design. Their advantages and liabilities overlap in some areas and diverge in others.

The scientifically defensible question is not “Which carrier wins?” It is “Which exact formulation can load this cargo, reach this biological destination, release it at the required time, remain stable, be manufactured reproducibly and show acceptable safety and clinical evidence?”
