A retrospective study of preserved frogs from Brazil’s Boracéia Biological Station identifies amphibian chytridiomycosis—the disease caused by Batrachochytrium dendrobatidis (Bd)—as the most directly supported explanation for a die-off that began in 1979. Researchers found no detectable Bd evidence in specimens collected before 1979, while more than half of the 1980s specimens tested positive. The result is compelling for Boracéia, but it does not prove that Bd caused every amphibian decline across Brazil’s Atlantic Forest.
What happened at Boracéia
Amphibian numbers fell sharply in Brazil’s Atlantic Forest during the 1970s. At Boracéia Biological Station, a series of die-offs between 1979 and 1982 removed frogs that had previously been common. Earlier explanations included climate change and extreme weather, but the historical record did not identify a specific biological cause.
The new work, by researchers at the Smithsonian National Zoo and Conservation Biology Institute (NZCBI) and Pennsylvania State University, was published in Molecular Ecology on October 14, 2026. Its central finding is temporal: the first evidence of Bd appears after the disappearances began, and infection is widespread in specimens collected during the 1980s. That timing makes chytrid the explanation most directly supported by the surviving evidence.
“After almost 50 years, we still aren’t confident what caused many historical amphibian die-offs, making these declines some of the most enduring cold cases in wildlife epidemiology,” said Jack Boyette, an NZCBI graduate fellow and Pennsylvania State University doctoral candidate.

How museum specimens reconstructed the outbreak
A collection with before-and-after samples
William Ronald Heyer’s Smithsonian National Museum of Natural History collections preserve material from five Boracéia field campaigns: two in the 1970s, before the sudden declines, and three in the 1980s, after them. Researchers examined more than 370 specimens representing 17 species. Because the animals were collected at known times, the series functions like a historical timeline that living surveys alone cannot recreate.
Testing the historical material
NZCBI scientists tested the preserved specimens for evidence of Bd and compared the results with the collection dates. They also returned to Boracéia six times between 2022 and 2024 with researchers from the University of São Paulo. The modern visits provided a way to ask whether species represented in the old collection still occur at the site and whether the fungus remains part of the local environment.

The evidence does not require collectors in the 1970s to have recognized a new disease. Their specimens were preserved for ordinary natural-history purposes; decades later, those same samples could be examined for a pathogen that was not being tracked at the time. As Michael Campana, an NZCBI genomicist, put it, museum collections make it possible to detect pathogens that “went unnoticed for decades” and reconstruct outbreaks that would otherwise remain unresolved.

Why the timing points to chytrid
Bd attacks amphibian skin, an organ frogs need for water and electrolyte balance. The Boracéia data do not by themselves show every step of transmission, but the sequence is consistent with a pathogen invasion: no detectable Bd in the pre-1979 samples, a die-off from 1979 to 1982, and frequent positive specimens in the 1980s.
Boyette said that environmental factors could have contributed, while adding that the findings “most directly implicate the chytrid fungus” because regional pathogen arrival coincided with the unexplained disappearances. That wording matters. The study identifies the likeliest culprit at this site; it does not eliminate weather, habitat conditions or other stresses as contributing factors.
The counterintuitive species pattern
The specimens also contain an apparent contradiction. Species reported to have declined most dramatically had no detectable infections in the sampled material, while some species that did not decline were heavily infected.

The researchers propose a sampling explanation. Frogs that were highly susceptible may have died before collectors arrived, leaving few or no infected bodies in the museum series. More tolerant species could survive infection, remain common enough to collect and therefore appear frequently among positive specimens. This is a hypothesis about survival and sampling, not proof that every species responded in the same way.

Did the frog populations recover?
Modern surveys found 12 of the 17 species represented in the historical collection. Five were not found, but their current local status remains uncertain: absence during the modern visits does not establish local extinction.
The surveys suggest that most species have returned or persisted despite Bd still being present. One possible explanation is that tolerance to the fungus became widespread among species that suffered the historical losses. The study does not establish whether that tolerance arose through genetic change, ecological filtering, or another process; researchers are now using DNA from museum samples to investigate the genomic mechanisms behind historical responses.

What this adds to amphibian-disease research
Museum-based pathogen studies can reveal when an infection first appeared, whether it followed or preceded a population crash, and which hosts were present during an outbreak. A separate peer-reviewed analysis of Cascades frogs detected Bd in 13 of 364 museum specimens collected from 1907 to 2003 (3.5%); the earliest positive sample was from 1978. That precedent shows how archived material can extend disease records far beyond the period of routine pathogen surveillance.
At Boracéia, the value comes from the unusually close match between collection dates and the documented decline. The approach can also expose survivorship bias: the animals most vulnerable to a pathogen may be missing from the post-outbreak record, while tolerant animals remain overrepresented.
What the study cannot establish
- It is site-specific. Boracéia is one location, and the findings cannot automatically be generalized to the entire Atlantic Forest.
- It is not a universal explanation. Other historical amphibian declines may have had different causes or multiple interacting causes.
- Five species remain unresolved locally. Not finding them in six modern visits leaves their status uncertain rather than proving extinction.
- Persistence is not immunity. Current frogs may tolerate Bd better, but the fungus remains present and can still pose a conservation risk.
What researchers are doing next
Scientists are analyzing DNA preserved in the museum material to look for genomic features associated with survival or susceptibility during the outbreak. Ancient environmental DNA may also help fill periods when collectors took few specimens or missed the most vulnerable species.

The work has a wider warning beyond frogs. A related chytrid pathogen, Batrachochytrium salamandrivorans, threatens European salamanders and could threaten North American populations. Historical collections therefore serve not only as records of past losses but also as reference points for recognizing and responding to future pathogen invasions.
Bottom line
For Boracéia’s 1979–1982 die-off, preserved specimens provide the strongest evidence yet for Bd as the cause: pre-1979 samples were negative, while more than half of 1980s samples carried detectable infection. Most historically represented species were found again in surveys from 2022–2024, suggesting recovery or persistence with greater tolerance, but the result remains a carefully bounded conclusion about one Brazilian site—not a blanket explanation for every frog disappearance.
