Horseshoe crabs matter to pharmaceutical manufacturing because their blood supplied the biological basis for a sensitive test for bacterial endotoxins—harmful substances that can cause serious reactions in patients. The traditional test, Limulus amebocyte lysate (LAL), is no longer the only option: recombinant Factor C (rFC) can test for endotoxins without crab-derived material, when the method is shown to suit the product and its use.
What the test detects—and why it matters
Endotoxins are substances released by certain bacteria. If they contaminate a medicine, they can trigger serious adverse reactions. The U.S. Food and Drug Administration (FDA) describes endotoxin testing as a critical safety and quality step in manufacturing biological products. FDA guidance on pyrogen and endotoxin testing
The test does not use horseshoe-crab blood as an ingredient in the medicine. Instead, it uses a reaction in the crab’s blood cells as a signal that endotoxin may be present in a sample.

How horseshoe-crab biology became a laboratory test
Horseshoe crabs have amebocytes, blood cells involved in their innate defense. When exposed to bacterial material such as endotoxin from gram-negative bacteria, these cells trigger a clotting response. Researchers adapted that response into Limulus amebocyte lysate, or LAL, an assay used to detect endotoxin contamination. FDA’s explanation of horseshoe-crab blood and bacterial testing

- Compact & Travel-Friendly: The ultra-compact design allows the adapter to stay plugged into your device without blocking adjacent ports or adding bulk, reducing wear and tear on your original USB ports.
- 12-Month Warranty: Backed by a 12-month manufacturer warranty for peace of mind. Designed to meet strict quality control standards for reliable everyday performance.
LAL tests can use different readouts. In a gel-clot test, the laboratory checks whether a clot forms; in a chromogenic test, a reaction produces a color change. These are different ways of reading an assay built around the same general biological detection system, not interchangeable descriptions of every test’s procedure or performance. FDA’s overview of LAL assay formats
LAL and rFC: what is different?
Recombinant Factor C (rFC) is produced using recombinant technology rather than extracted from horseshoe-crab blood. It reproduces Factor C, the endotoxin-sensitive component that starts the relevant detection pathway. The difference matters both for reagent origin and for what the test detects.

| Comparison | LAL | rFC |
|---|---|---|
| Reagent source | Lysate derived from horseshoe crabs | Recombinantly produced Factor C; does not require crab-derived material |
| Detection pathway | Lysate reagents can respond through Factor C and the separate Factor G pathway | Detects through Factor C and does not recognize Factor G |
A 2020 review reported high correlation in comparative testing between traditional lysate tests and recombinant reagents. That does not establish that every rFC method performs identically to every LAL assay for every product. The same review notes that rFC does not detect Factor G, a separate pathway present in lysate reagents. Bolden et al., PDA Journal of Pharmaceutical Science and Technology (2020)

Neither method is established as universally superior for all applications. The practical question is whether the chosen assay has been shown suitable for the particular material and intended use.

What the 90% reduction figure does—and does not—mean
Researchers reported that using rFC to test water and other common manufacturing materials could reduce the use of crab-derived reagents by 90% in the study’s application. This is a bounded study finding, not a claim that pharmaceutical testing overall can eliminate 90% of horseshoe-crab use or that the same reduction applies to every medicine or test. Maloney et al., PLOS Biology (2018)
FDA’s current approach to recombinant testing
FDA’s revised Edition 2 guidance accommodates a broader range of recombinant reagents for bacterial endotoxin testing. It says sponsors should verify that a recombinant assay is suitable for its intended purpose. FDA also points to USP Chapter <86>, “Bacterial Endotoxins Test Using Recombinant Reagents,” and advises reviewing the supplier’s validation package and conducting experiments appropriate to the complexity of the material. On August 31, 2026, FDA reported that an international collaborative assessment had concluded with approval in June 2026 of a post-approval change protocol. It allowed a manufacturer to transition several biological products—covering cancer, cardiovascular, respiratory, and rare-disease therapies—from a traditional crab-derived lysate test to recombinant endotoxin testing. This demonstrates a transition route for those products, not a blanket approval of every recombinant assay for every product. FDA announcement on international collaboration to advance alternative testing
Why the animal remains relevant
The crab’s continuing pharmaceutical importance is historical and practical: its amebocyte response made LAL possible, and LAL became an established way to detect a serious contamination risk. Recombinant methods now provide a way to test without crab-derived reagents, but manufacturers still have to demonstrate that their selected method works for the specific material and use. The biology remains the origin of the test; it does not mean every current endotoxin test must use horseshoe-crab blood.
