Embryonic stem cells (ESCs) come from the inner cell mass of a blastocyst-stage embryo. Induced pluripotent stem cells (iPSCs) are created by reprogramming differentiated cells, such as skin cells, in the laboratory. Both can be pluripotent, meaning they may produce cell types representing all three embryonic germ layers, and both support research into development, disease, medicines and potential cell therapies. Their origins create different consent and oversight questions, while the most useful scientific comparison depends on the specific cell line, its demonstrated differentiation behavior and the experiment’s purpose.

Neither category is a universal winner, and the terms alone do not prove that a line will perform well. A sound comparison examines provenance, functional evidence of pluripotency, controls, intended use and the legal or funding rules that apply where the work is performed.

iPSCs and ESCs at a glance

Comparison point Embryonic stem cells (ESCs) Induced pluripotent stem cells (iPSCs)
Starting material Cells from the inner cell mass of a blastocyst-stage embryo Differentiated, tissue-specific cells reprogrammed in a laboratory
How pluripotent properties arise They are present in the early embryonic cell population from which the line is derived They are acquired through laboratory reprogramming
Developmental potential Can be pluripotent when demonstrated experimentally Can be pluripotent when demonstrated experimentally
Main provenance question How the embryo and its cells were obtained, consented and approved for research How the donor tissue was obtained, consented and handled, plus how reprogramming was performed
Typical uses Developmental studies, disease research, drug testing and work informing cellular therapies Developmental studies, disease research, drug testing and work informing cellular therapies
Best basis for comparing lines Experimental differentiation results, quality controls, genetic and biological characterization, and suitability for the question being studied

“Pluripotent” describes developmental potential, not a guarantee that every line will make every target cell efficiently or safely. A line’s label, age or origin cannot substitute for experiments showing what it actually does.

The table also shows why a simple “which is better?” ranking is misleading. Two lines in the same category may differ more from each other than an ESC and an iPSC tested under the same conditions. The relevant comparison is usually between named lines, protocols and controls, not between acronyms.

What embryonic stem cells are

Origin in the blastocyst

Human ESCs are derived from the inner cell mass of a blastocyst-stage embryo. The inner cell mass is an early population capable of giving rise to the body’s somatic lineages. Establishing an ESC line therefore involves embryo-related provenance, consent and review in addition to the laboratory work needed to maintain and characterize the cells.

That provenance should remain traceable throughout a project. Investigators need to know how the embryo and its cells were procured, what the consent authorized, and which review process covered the work. A line can be scientifically useful yet unavailable for a proposed study if its consent terms or institutional approvals do not cover that use.

Why hESC research remains scientifically important

Human ESC research has provided foundational knowledge about early development, cell identity and differentiation. The International Society for Stem Cell Research (ISSCR) describes human embryonic stem-cell work as contributing to disease modeling, drug screening and development research, and notes that this research also helped inform the development of iPSC approaches. The existence of iPSCs has not eliminated the scientific value of ESCs or made the two categories interchangeable.

What induced pluripotent stem cells are

Reprogramming differentiated cells

iPSCs begin with differentiated cells from a tissue such as skin. Researchers reprogram those cells in the laboratory so that they acquire properties associated with embryonic stem cells, including the capacity for pluripotent differentiation when that capacity is demonstrated experimentally.

The starting tissue gives an iPSC project a different chain of custody from an ESC project. Donor permission, privacy protections, collection procedures and later handling all matter. The record should also describe the reprogramming history and the conditions under which the resulting line was maintained, because those details help others interpret later differentiation results.

What the “induced” label tells you—and what it does not

“Induced” identifies the route used to obtain the pluripotent state; it does not certify a particular level of quality. Different iPSC lines can respond differently to the same differentiation protocol. Reprogramming history, donor-tissue provenance, culture conditions and the tests used to establish identity all matter when deciding whether a line is suitable for an experiment.

An iPSC line should therefore be treated as a characterized biological material, not merely as a donor sample with a new name. Comparisons should record the exact line, passage or culture history available to the study, the differentiation method and the controls used. Those records make it possible to distinguish a biological difference from a difference in handling.

Pluripotency requires functional evidence

Differentiate into all three germ-layer representatives

ISSCR’s research standard says a human pluripotent cell line should be designated pluripotent only after it has been experimentally shown to differentiate into cells representing all three embryonic germ layers: ectoderm, mesoderm and endoderm. In the society’s wording: "A cell line should be designated as pluripotent only if it has been experimentally shown to be capable of differentiating into cells representing all three embryonic germ layers that give rise to the somatic lineages of a developing organism."

For a comparison, ask to see the actual differentiation evidence and the assay context. A panel of marker-positive cells is useful characterization, but marker expression by itself does not demonstrate pluripotency. Results should identify the protocol, the target populations, the controls and how the differentiated cells were confirmed.

Functional evidence is broader than checking whether a few cells carry familiar markers. A convincing record connects the marker or identity test to a differentiation experiment and explains what populations were produced. It should also make clear whether the evidence applies to the specific line being considered, rather than to a related line or a general platform.

Teratoma assays are not mandatory under the ISSCR standard

Older discussions often treat a xenograft teratoma assay as the definitive test. ISSCR’s current research standard states: "Recommendation 2 .1 .2: Xenograft (teratoma) assays are not required to indicate pluripotency." That does not remove the need for rigorous functional testing; it means a teratoma assay is not a required indicator when appropriate differentiation evidence and other characterization are available.

Researchers should not use the absence of a teratoma assay as a reason to skip characterization, nor assume that performing one automatically answers every question about a line. The relevant evidence still has to cover the three germ-layer potential and the intended downstream experiment.

Key differences that affect real research decisions

1. Origin and derivation

The defining difference is origin. ESCs are embryo-derived; iPSCs are generated from differentiated cells by reprogramming. This distinction remains true even when both lines can produce similar differentiated cell types in a particular protocol.

2. Donor, consent and provenance

ESC work requires documentation of how the embryo and inner-cell-mass material were procured, what consent covered, and which review process authorized the work. iPSC work requires equivalent attention to the source tissue and donor consent, together with documentation of reprogramming and subsequent cell handling. The applicable requirements depend on the country, institution, funding source and type of material.

ISSCR guidance calls for rigorous scientific and ethical oversight, including review of procurement and research. U.S. National Institutes of Health guidance addresses informed consent and protections for human tissues. A line that is scientifically attractive can still be unsuitable for a project if its provenance or consent terms do not permit the planned use.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Consent is not a one-time administrative detail. The planned use, sharing arrangements and funding context should be checked against the permission attached to the material. Institutions may also require separate scientific, ethics, biosafety or privacy reviews. Because requirements vary, a decision made for one jurisdiction or funder should not be presented as a universal rule.

3. Functional behavior is line-specific

Neither “ESC” nor “iPSC” predicts a universal differentiation ranking. A useful comparison records the behavior of the particular lines under the particular protocols being considered. Researchers should compare reproducibility, target-cell identity, unwanted-cell populations and other quality measures rather than assuming that one category always performs better.

When a head-to-head result matters, use the same or deliberately matched conditions and document them. Otherwise, a difference in medium, timing, handling or endpoint measurement can be mistaken for an intrinsic difference between ESCs and iPSCs. The conclusion should be limited to the tested lines and protocol, not generalized to every line in the category.

4. Experimental purpose

Both cell types can support development research, disease models, drug testing and the development of cellular therapies. The right choice depends on the biological question and the controls needed. A study focused on early developmental biology may need a well-characterized ESC reference, while a project centered on a particular donor-derived condition may be designed around an iPSC line. Those are examples of experimental fit, not universal rules.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

For disease work, the ability to connect a line to a donor or disease-associated mutation may be important, but the line still needs suitable comparison groups and functional characterization. For drug studies, the differentiated cells must be validated for the question being asked; a pluripotency label alone does not establish predictive performance.

5. Oversight and funding context

Review requirements vary by jurisdiction and by whether a project receives public funding. Embryo provenance is often a central review issue for ESC work, while iPSC projects must address donor-tissue consent and privacy alongside laboratory and clinical requirements. The relevant policy should always be identified by location and date; a rule in one funding system is not a worldwide prohibition or permission.

How the two cell types are used

Developmental research

Pluripotent cells provide models for asking how cells acquire identities and organize into tissues. ESCs offer an embryo-derived reference for studying early developmental states. iPSCs allow researchers to investigate reprogrammed cells and compare differentiation across donor-derived lines. In either case, interpretation depends on the quality of the differentiation model and the controls used.

Disease modeling

Researchers can differentiate pluripotent cells toward disease-relevant cell types and examine cellular mechanisms. The choice between an ESC and an iPSC line is governed by the disease question, the available controls, the provenance permitted by the study and the behavior of the selected line. The fact that the NIH registry includes lines with disease-associated mutations does not establish that every registry line, or every pluripotent line, is an appropriate model for a particular disorder.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Drug discovery and safety testing

Pluripotent-cell-derived models can be used to test candidate medicines and investigate effects in human cell types that are difficult to obtain directly. A credible result requires validated differentiated cells, appropriate comparison groups and assays that distinguish a drug effect from variability in the cell model. The cell type’s label alone cannot establish predictive performance.

Potential cellular therapies

Both ESC-derived and iPSC-derived cells are being studied as sources for cellular therapies. Translation requires evidence that the intended differentiated population is correctly identified, sufficiently controlled and safe for the proposed use. Research capability should not be confused with an approved treatment.

How to compare an ESC or iPSC line before using it

  1. Define the biological question. State whether the project concerns development, a disease mechanism, drug response or a potential therapeutic cell product. The purpose determines which controls and differentiation endpoints are relevant.
  2. Document provenance. Record whether the line came from an embryo-derived inner cell mass or from reprogrammed differentiated tissue. Obtain the consent, procurement and use documentation required by the institution and jurisdiction.
  3. Check identity and quality records. Review the line’s authentication, culture history and available characterization. Do not infer quality from a catalogue label or a single marker panel.
  4. Inspect functional pluripotency evidence. Look for experimental differentiation representing ectoderm, mesoderm and endoderm, with methods and controls described clearly.
  5. Evaluate the intended differentiation protocol. Confirm that the line has been tested, or can be responsibly tested, for the target cell type needed by the project. Compare yield and identity under the same conditions when a head-to-head result is important.
  6. Plan controls. Include suitable reference lines, technical controls and disease-relevant comparisons. A control strategy is part of the experiment, not an optional add-on after unexpected results.
  7. Review oversight before starting. Determine which institutional, ethics, biosafety and funding reviews apply. Embryo-related and donor-tissue requirements should be settled before material is procured or used.
  8. Separate research from clinical claims. If the work concerns a therapy, require evidence and safety review appropriate to that clinical context. Do not treat an undifferentiated pluripotent culture as a ready-made injectable product.

For a formal comparison, preserve the line identifiers, consent status, assay methods and decision criteria in the study record. This prevents a favorable result from one experiment being detached from the conditions that produced it and helps reviewers assess whether the conclusion is transferable.

Safety and clinical-translational limits

ISSCR warns that injecting undifferentiated pluripotent cells is unsafe because they could form tumors. A proposed cell therapy therefore needs a controlled differentiation process, testing for unwanted undifferentiated cells and a broader safety assessment appropriate to the product and route of administration. The warning applies to pluripotent cells generally; switching from ESCs to iPSCs does not remove the risk.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Claims made by clinics or commercial providers should be judged against published evidence, regulatory status and documented safety. The fact that a cell line can differentiate in a laboratory does not establish that a treatment is effective or safe in people. A research-grade line, a differentiated cell preparation and an approved clinical product are different categories and should not be conflated.

U.S. NIH policy update for human embryonic stem-cell lines

On January 23, 2026, NIH announced a pause on reviewing and approving applications for new human embryonic stem-cell lines to be added to its Human Embryonic Stem Cell Registry while it assessed their scientific utility. NIH stated that lines already approved for the registry could continue to be used in NIH-funded research.

NIH reported 503 approved hESC lines, including more than 200 with disease-associated mutations, and said the last approved line was added in December 2023. Those figures describe the NIH registry, not every hESC line worldwide. The notice is a dated U.S. policy development and should not be described as a general ban on ESC research, a worldwide rule or a finding that iPSCs have replaced ESCs.

ISSCR’s guidelines page identifies an August 2025 targeted update limited to sections on stem-cell-based embryo models; it says the other sections remain as published in 2021. Researchers should therefore check the current guidance and the rules of their own institution and jurisdiction rather than generalizing from one policy notice.

Common misconceptions

  • “iPSCs and ESCs are the same thing.” They can share pluripotent properties, but their origins and derivation methods differ, which affects provenance, consent and oversight.
  • “A pluripotency marker proves pluripotency.” Marker expression alone is insufficient. ISSCR calls for experimental differentiation into representatives of all three germ layers.
  • “ESCs are always better.” No universal head-to-head winner is established. Performance is cell-line- and protocol-specific.
  • “iPSCs have made ESC research unnecessary.” Human ESC research remains foundational and has informed iPSC approaches; the two models can answer overlapping but not identical questions.
  • “Any pluripotent cells can be injected as treatment.” Undifferentiated pluripotent cells can form tumors, so laboratory pluripotency is not a clinical safety clearance.
  • “A U.S. registry decision applies everywhere.” NIH registry policy governs the U.S. NIH funding context. Other jurisdictions and funders may apply different requirements.

Bottom line for choosing between iPSCs and ESCs

Choose based on the question, not the acronym. ESCs provide embryo-derived reference material with specific provenance and oversight obligations; iPSCs provide a laboratory-reprogrammed route from differentiated tissue with its own donor-consent and characterization requirements. For either type, demand functional evidence of pluripotency, evaluate the exact line and differentiation protocol, use appropriate controls, and verify the ethical, regulatory and funding rules that apply. Neither cell category is universally superior or automatically interchangeable with the other.

Frequently Asked Questions

Are iPSCs the same as embryonic stem cells?

No. ESCs are derived from the inner cell mass of a blastocyst-stage embryo, whereas iPSCs are made by reprogramming differentiated tissue cells. They may share pluripotent properties, but their origins, provenance and oversight requirements differ.

Which is better, iPSCs or ESCs?

Neither is universally better. The appropriate choice depends on the biological question, the exact line, its demonstrated differentiation behavior, available controls, provenance and the rules governing the project.

How is pluripotency demonstrated?

ISSCR says a human line should be designated pluripotent only after experimental differentiation into representatives of ectoderm, mesoderm and endoderm. Marker expression alone is not sufficient, and the evidence should identify methods, targets and controls.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Is a teratoma assay required to prove pluripotency?

Not under the current ISSCR research standard. ISSCR states that xenograft, or teratoma, assays are not required to indicate pluripotency. Rigorous functional differentiation evidence is still required.

Are embryonic stem cells legal everywhere?

Rules differ by country, institution and funding source. ESC projects must address embryo procurement, consent and review, while iPSC projects have their own donor-tissue and privacy obligations. A policy in one jurisdiction should not be generalized worldwide.

Can undifferentiated iPSCs or ESCs be injected as a treatment?

No. ISSCR warns that undifferentiated pluripotent cells are unsafe because they could form tumors. A potential therapy requires controlled differentiation, testing for unwanted cells and appropriate clinical safety evidence.

Did NIH ban embryonic stem-cell research in 2026?

No. On January 23, 2026, NIH paused review of applications for new hESC lines for its registry while assessing scientific utility. NIH said already approved lines could continue to be used in NIH-funded research.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Do iPSCs make ESC research unnecessary?

No. ISSCR describes human ESC research as foundational to development, disease modeling and drug screening, and says it helped inform iPSC approaches. The two systems can answer overlapping but not identical questions.

The Bottom Line

Choose based on the question, not the acronym. ESCs are embryo-derived and iPSCs are reprogrammed from differentiated tissue; both require line-specific functional evidence, appropriate controls, documented provenance and compliance with applicable oversight. Neither is universally superior or automatically interchangeable.
