When repeated chemistry experiments disagree, do not immediately repeat the run or blame “human error.” First preserve the raw observations and the actual conditions for every run. Then classify the pattern (random scatter, a consistent offset, or a qualitatively different outcome), verify the measurement and calculation chain, compare reaction conditions one variable at a time, and use replicates plus a suitable reference to distinguish random variation from systematic bias. Keep every change within your method’s safety instructions and your laboratory’s supervision requirements.

Start by describing exactly how the results are inconsistent

“Different results” can describe several fundamentally different problems. Diagnosis is faster when you name the pattern before changing the procedure.

Widely scattered numerical results

If nominally identical runs produce values spread over a broad range, suspect random variation first. The spread may come from reading uncertainty, inconsistent liquid handling, temperature fluctuations, timing differences, mixing, or other run-to-run changes. Scatter does not prove that the average is accurate.

Values that cluster but are all too high or too low

Tightly grouped values that disagree with an accepted value or reference indicate a possible systematic error. A miscalibrated instrument, an incorrect blank or zero, a conversion error, or a consistently biased preparation step can produce precise-looking but inaccurate data. Repeating the same biased procedure and averaging the results will not remove that offset.

A qualitative change, such as no reaction or a lower yield

A failed reaction, unexpected color, precipitate, gas evolution, or markedly lower yield points to a changed condition rather than ordinary measurement scatter. Compare scale, reagent and solvent identity or quality, temperature, time, mixing, and the order and rate of additions with the successful run.

Do not discard an inconvenient result. Preserve the original value, observation, and context; the outlier may identify the condition that changed.

Freeze the evidence before troubleshooting

Make a run-by-run record before recalibrating equipment, remaking solutions, or altering the protocol. A useful comparison sheet contains the following fields.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.
Record for every run What to compare
Sample and reagent identity Name, concentration, preparation date, and lot or batch where available
Amounts and scale Masses, volumes, concentrations, vessel size, and total reaction scale
Equipment Instrument or glassware used, identification if available, settings, range, and resolution
Temperature Set point, measured temperature, equilibration time, and any excursions
Timing Start point, reaction or incubation duration, sampling time, and delays
Mixing and additions Stirring or shaking method, speed or description, addition order, and addition rate
Observations Color, clarity, precipitate, gas, phase separation, endpoint appearance, and anything unusual
Calculations and data handling Raw readings, units, conversions, significant figures, spreadsheet formulas, exclusions, and rounding

Compare the successful and unsuccessful runs side by side. Mark each condition as identical, different, or unknown. “Unknown” is a useful finding: an omitted detail in a published or recorded procedure can be the source of irreproducibility.

Check the entire measurement and calculation chain

Match the instrument to the measurement

Confirm that the instrument is suitable for the magnitude and precision required. Instruments differ in sensitivity and capacity; using equipment outside its useful range can create either large scatter or a consistent offset. The University of Iowa chemistry lab manual emphasizes that these properties affect results. Check the laboratory’s existing balance, pipette, buret, thermometer, spectrometer, or other device before assuming new equipment is needed.

Verify zeroing, calibration, and setup

Follow the applicable procedure for zeroing and calibration, including the correct standards, range, warm-up or equilibration period, and environmental conditions. Record the calibration result rather than simply writing “calibrated.” If the instrument fails its check, stop using data collected since the last acceptable check and follow your laboratory’s corrective procedure.

Audit units and arithmetic

Recalculate at least one run independently from the raw readings. Check mass-versus-volume substitutions, dilution factors, molar units, prefixes, significant figures, and conversions. Inspect spreadsheet formulas cell by cell, including copied references and hidden rounding. Confirm that any computer program or equation behaves as expected with a simple known input; Cornell’s virtual-lab guidance recommends this type of check and comparison with known reference values.

Separate transcription from experimental variation

Compare the instrument display, notebook entry, electronic file, and final report. A misplaced decimal or swapped sample identifier can mimic a chemical failure while leaving the physical experiment unchanged. Keep the original entry visible and annotate corrections rather than overwriting it.

Measurements are rarely exact; each has associated uncertainty or error, as stated in the University of Iowa CHEM:1110 Lab Manual (2020). The practical question is whether the observed uncertainty is consistent with the method and whether a bias is present.

Decide whether you are seeing random error or systematic error

Use replicates to quantify scatter

Repeat measurements under consistent conditions and calculate the mean with an appropriate spread statistic, such as standard deviation, while retaining every raw value. The number of observations matters: report n and explain what was repeated. Five readings of one prepared sample are not equivalent to five independently prepared reaction runs.

The Iowa manual gives an illustrative titration report of 12.04 ± 0.02 mL (n=5), with the mean rounded to match the reported standard deviation. This is a worked example from that manual, not a universal precision target for titrations.

Test accuracy separately from repeatability

Question Evidence What it tells you
Are repeats close to one another? Spread among replicate values Repeatability or precision
Are repeats close to an accepted value? Comparison with a suitable reference or known quantity Accuracy and possible bias
Are repeats independent? Separate preparations, runs, or samples documented explicitly Whether the result represents independent experimental variation
Could both methods share the same error? Comparison with an independent instrument or technique Whether an apparent agreement is falsely reassuring

If values cluster tightly but remain offset, investigate systematic error. Cornell notes that “Systematic error can be difficult to identify and correct” and that averaging repeated measurements leaves a systematic error unchanged. Calibration, an independent instrument or technique, and a test of the whole procedure against a known quantity are more informative than simply collecting more repeats.

For reaction work, compare the conditions that control the outcome

For a reaction that worked once but not again, make a condition-by-condition comparison with the original notebook entry or procedure. Change one factor at a time whenever practical so that a successful correction can be identified.

Reaction scale

Check whether every amount was scaled proportionally and whether vessel geometry, headspace, heat transfer, or mixing changed with scale. A procedure that behaves on a small scale may not behave identically at a larger one.

Reagent and solvent identity or quality

Confirm names, concentrations, preparation dates, storage, and lot information. Check for substitutions, degradation, contamination, evaporation, or an unrecorded change in solvent grade. If the original procedure does not state a detail that affects quality, record it explicitly for the next run.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Temperature

Compare the actual temperature, not only the nominal setting. Note equilibration time, heating or cooling rate, thermometer location, and any excursion during addition or reaction.

Time and sampling

Define when the clock starts and ends. Delays before addition, inconsistent incubation duration, or sampling at different stages can change conversion and yield even when the written duration appears identical.

Mixing

Describe how the mixture was stirred, shaken, or otherwise mixed and for how long. Inadequate or inconsistent mixing can create local concentration and temperature differences.

Order and rate of addition

Record which reagent was added first, the vessel position, and whether additions were fast, dropwise, or staged. The University of Rochester Department of Chemistry identifies addition order and rate as reproducibility factors. Do not assume that a brief phrase such as “add reagent” captures a chemically important rate.

Published or recorded procedures can omit operational details, and a reported result may not reproduce exactly as written. Treat missing details as variables to document, not as permission to improvise outside the approved method.

Design repeats that answer the right question

Repeated readings of one sample

Multiple readings from one preparation test instrument or reading repeatability. They cannot reveal variation introduced while weighing, dissolving, diluting, or running the reaction itself.

Independent preparations or reaction runs

Separate sample preparations and runs capture more of the actual method variation. Label these as independent runs in the report and keep their raw records separate.

Describe the source of every replicate

Assay-reporting guidance from the NIH Assay Guidance Manual recommends stating whether replicates came from a sample stock, plate, run, or separate run. Adapt that principle to your experiment. Also record changes in temperature, instrument limits, liquid handling, and other fluctuations that could contribute to random variation.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Choose a meaningful number and spread statistic

Use enough consistent repeats to characterize the variation relevant to your decision, and state why that number was chosen when the method or laboratory procedure specifies a minimum. Report the mean, the spread statistic, and n together. Do not report a mean alone: it can hide a failed run, a bimodal pattern, or a steadily drifting instrument.

Use a reference or independent method to test for bias

A reference check asks whether the complete procedure measures a known quantity correctly. Use a suitable accepted value, reference material, or other known quantity when feasible and permitted by the method. Compare the result, not just the instrument reading, because preparation and calculation errors can occur before measurement.

An independent instrument or technique provides a stronger test when the two methods do not share the same likely failure mode. Agreement between two instruments that were calibrated against the same faulty standard may not expose a common bias; document what is genuinely independent about the comparison.

If the reference comparison fails, do not “correct” the data by subtracting the observed difference unless your validated procedure explicitly authorizes that correction. Identify and fix the cause, then repeat the required checks.

A practical decision tree for the next run

  1. Preserve: Save raw readings, observations, calculations, instrument status, and the full condition record for every run.
  2. Classify: Decide whether the problem is scatter, a consistent offset, or a qualitative reaction change.
  3. Verify: Check instrument suitability, zeroing, calibration, units, formulas, data entry, and reference values.
  4. Compare: Build a side-by-side table of scale, reagents and solvents, temperature, time, mixing, and addition sequence.
  5. Test one explanation: Make the smallest approved change that distinguishes between plausible causes; avoid changing several conditions at once.
  6. Replicate appropriately: Use repeated readings for measurement repeatability and independent preparations or runs for method reproducibility.
  7. Check bias: Compare with a known quantity or an independent method when feasible.
  8. Document the outcome: Record what changed, what stayed constant, the new raw results, and whether the suspected cause was supported.

Match symptoms to the most useful check

Symptom First checks Next action
Replicates are widely spread Timing, temperature excursions, mixing, liquid handling, reading technique, and instrument limits Repeat under tightly documented conditions and report the spread
Replicates are tight but miss the accepted value Calibration, zero, standard or reference, units, formula, and sample preparation Test the full procedure against a known quantity or independent method
Only one run is extreme Raw observation, transcription, sample identity, and any unusual event Retain it, investigate it, and do not delete it without a documented rule
Reaction yield falls while measurements look normal Reagent or solvent lot and quality, scale, temperature, time, mixing, and addition order or rate Reproduce the successful condition set one variable at a time
Results change after a software or spreadsheet update Formula references, units, rounding, and test inputs with known outputs Restore or correct the calculation workflow and recalculate from raw data

Worked examples

A precise titration with the wrong concentration

Suppose five titres are close together, similar to the Iowa example of 12.04 ± 0.02 mL (n=5), but the calculated concentration disagrees with a suitable reference. The narrow spread supports repeatability, not accuracy. Check buret zeroing and calibration, standard concentration, endpoint interpretation, dilution and unit conversions, and the calculation formula. More titres using the same standard will not remove a common bias.

A reaction that succeeds once and then gives little product

Keep the successful notebook entry beside the failed one. Compare reagent and solvent identity or lot, scale, actual temperature profile, elapsed time, mixing, and addition order and rate. If one detail is missing from the original procedure, treat it as unknown and obtain an approved instruction rather than guessing. Test the most plausible difference in an otherwise unchanged run.

Stable instrument readings but changing results after preparation

If repeated readings of one solution agree but independently prepared solutions disagree, the instrument may be repeatable while preparation is not. Compare weighing, dissolution, dilution, reagent condition, and timing between preparations. Report the replicate source so readers do not mistake repeated readings for independent experiments.

Report results so another person can judge the inconsistency

A defensible report includes the raw data or a traceable record, the mean, a suitable spread statistic, and n. State whether repeats were readings of one sample, separate preparations, or independent runs. Identify the instrument and relevant settings, calibration or reference checks, reagent and solvent identities or lots where available, scale, temperature, timing, mixing, addition sequence, observations, and every calculation or data-processing step that affects the result.

State what the evidence supports: for example, “repeatability was good but accuracy was not established,” or “variation decreased after temperature and addition rate were controlled.” Avoid calling a result “accurate” solely because replicates agree.

Safety and stopping rules

Troubleshooting is not a reason to bypass a method’s safety controls. Follow the experiment-specific procedure, safety data, institutional rules, required supervision, and equipment limits. Stop and consult the responsible supervisor when a reagent identity or concentration is uncertain, an instrument fails a required check, an unexpected reaction occurs, or a proposed change would alter hazard, scale, containment, or waste handling. The corrective experiment must be both scientifically informative and approved for the laboratory in which it is performed.
