A high valuation can reflect financing conditions, platform enthusiasm, scarce intellectual property or expectations about a large market. It does not show that a particular molecule reaches its target, helps patients or can be manufactured profitably. Evaluate the candidate in a separate chain of evidence: define exactly what the AI did, test the model for that task, examine the biological mechanism, verify prospective experiments, read the human trial record and then assess development and commercial execution.
The distinction matters because “AI-designed” is not a standardized level of contribution. A model may have helped find a target, generate or optimize a molecule, predict properties, design a trial or run operations. Those uses answer different questions and require different validation.
Start by separating three objects of analysis
Before reviewing a pitch deck or valuation, label the unit you are judging:
Do these 3 things before closing this tab:
1Fix the driver behind crashes, sound loss and screen glitches2Clear out junk files and repair common Windows errors3Scan for outdated or missing drivers - takes under a minute- The company: its valuation, cash, financing terms, ownership, runway, partnerships and portfolio.
- The platform: the models, data, laboratories and workflow claimed to produce or prioritize candidates.
- The candidate: one defined molecule, its target, mechanism, formulation, development stage and clinical evidence.
A company can have a valuable platform and weak individual assets, or a promising candidate inside a company whose valuation assumes unrealistic future sales. Never treat a platform benchmark or a funding round as a clinical result.

1. Specify what “AI-designed” actually means
Ask management to describe the model’s contribution in operational terms, not slogans. The U.S. Food and Drug Administration (FDA) describes AI use across nonclinical, clinical, postmarketing and manufacturing activities. “AI-powered” therefore does not establish that the candidate’s chemical structure was generated by AI.
Map the claimed contribution to a development stage
- Target discovery: the system identifies or ranks disease-relevant proteins, pathways or patient subgroups.
- Molecule generation: a generative model proposes chemical structures that satisfy specified constraints.
- Lead optimization: algorithms suggest changes to potency, selectivity, exposure, solubility, stability or other properties.
- Property prediction: models estimate activity, toxicity, pharmacokinetics, manufacturability or formulation behavior.
- Trial design and operations: AI helps select sites, identify participants, model enrollment or analyze data.
- Manufacturing and postmarketing: systems support process control, quality testing, safety surveillance or label management.
Require a timeline showing where human scientists made decisions and where the model generated, filtered or merely scored options. A molecule that was designed conventionally and later prioritized by a model should not be described as wholly AI-designed. Conversely, a generated structure still needs ordinary chemical synthesis, assays, toxicology and clinical testing.
Questions that expose vague claims
- What exact input did the model receive, and what output did it produce?
- Was the model used prospectively to choose this candidate, or was it applied retrospectively to explain a decision already made?
- How many model-generated compounds were synthesized and tested, including failures?
- Which decisions remained human-controlled, and what acceptance criteria were used?
- Can an independent party reproduce the result with the described data and model version?
2. Define the model’s context of use before judging performance
Model credibility is task-specific. FDA’s January 2025 draft guidance, Considerations for the Use of Artificial Intelligence To Support Regulatory Decision-Making for Drug and Biological Products, proposes evaluating credibility for a defined context of use (COU): how the model is used to answer a particular question. The guidance is a draft Level 1 document, nonbinding and marked “Not for implementation”; it is not a final rule. Its risk-based structure is nevertheless a useful diligence framework.
Write the COU as a testable statement
A useful COU identifies the decision, input data, output, users and consequences of error. For example: “The model ranks synthesized compounds for biochemical potency against target X using assay data available before the experiment, and the ranking determines which compounds enter the next test.” That is materially different from “the model discovers drugs.”

Check five elements of credibility
- Question and decision: What regulatory, scientific or operational decision will the output support?
- Inputs: Which assays, structures, patient records or manufacturing measurements are permitted? Are data from the same disease, laboratory and instrument conditions as the intended use?
- Validation design: Were test sets separated in time, geography, chemistry or patient population from training data? Could information leakage make the result look better than it is?
- Performance measure: Is the metric appropriate to the decision—such as calibration, sensitivity, precision, ranking quality or prospective hit rate—rather than a convenient aggregate score?
- Consequence of failure: What happens if the model is wrong? A false-positive ranking may waste experiments; a false-negative safety prediction could expose people to serious harm.
Ask for confidence intervals, predefined success thresholds, external validation and monitoring plans. A high accuracy number without the class balance, test-set construction and error costs is not enough to assess reliability.
3. Test the biological rationale independently of the model
Computational attractiveness cannot rescue an invalid target or a weak disease hypothesis. The central biological questions are ordinary drug-development questions, even when AI selected the target or molecule.
Target and mechanism
- What protein, pathway or biological state is being targeted?
- What evidence links that target to the disease and to the intended patient population?
- Does the molecule bind the proposed target at clinically plausible concentrations?
- Is target engagement measured directly, through a validated biomarker or a well-supported surrogate?
- Does engagement produce the predicted functional effect in relevant cellular systems?
Patient and disease fit
Mechanistic plausibility depends on who receives the drug. A target may matter only in a molecularly defined subgroup, at a particular disease stage or alongside a companion treatment. Check whether the proposed trial population reflects the biology used to justify the program. Broad enrollment can dilute a real effect; narrow enrollment can make recruitment and commercial scale difficult.
Recommended Free Tools
Look for disconfirming evidence
Request data on inactive analogues, target knockdown or knockout, rescue experiments, off-target activity and contradictory disease models. Ask whether negative experiments were reported or whether only successful examples appear in presentations. Independent replication by a laboratory that did not develop the model is especially valuable.

4. Move from computational claims to prospective experiments
Separate four evidence types that are often blended in investor materials:
| Evidence type | What it can show | What it cannot establish alone |
|---|---|---|
| Retrospective model benchmark | Performance on a defined historical or held-out dataset | That a new molecule will work in a living system or outperform standard discovery |
| Prospective biochemical assay | Whether a synthesized compound affects the target under stated laboratory conditions | Human exposure, safety or patient benefit |
| Cellular and animal studies | Functional activity, pharmacology and selected toxicity signals in the tested systems | Reliable efficacy or safety in people |
| Human clinical trial | Safety, pharmacology and, depending on design and phase, evidence of efficacy in a defined population | Success in other diseases, doses or patient groups |
Prospective design matters
A credible claim should identify the model and decision criteria before experimental results are known. Ask how many candidates were predicted, synthesized and tested; how many failed; and whether the company changed the model after seeing results. Reported “hit rates” are meaningful only with a denominator and a prespecified definition of a hit.
Replication and controls
- Use orthogonal assays that measure the same mechanism in different ways.
- Include negative controls, inactive compounds and known reference compounds.
- Repeat key findings across laboratories, instruments or biological systems.
- Compare model-selected compounds with a conventional baseline selected under the same conditions.
- Track potency, selectivity, exposure, stability, solubility and toxicity together; optimizing one property can damage another.
5. Read the clinical record, not the announcement
For every trial, obtain the protocol, statistical analysis plan where available, registry entry, results posting and publication. A press release can omit a failed endpoint, a changed analysis population or a short follow-up period.
Minimum fields to extract
- Phase and objective: Phase I usually informs safety, tolerability, dose and pharmacology; it does not establish patient benefit.
- Population: inclusion criteria, biomarker status, disease severity, prior treatment and geographic mix.
- Dose and schedule: dose levels, exposure, dose interruptions and whether the intended therapeutic range was reached.
- Endpoint: primary and secondary endpoints, measurement method, timing and clinical relevance.
- Comparator: placebo, standard of care, active control or single-arm design, with the limitations of that choice.
- Sample size and follow-up: enrollment, attrition, analysis population and duration.
- Safety: serious adverse events, discontinuations, laboratory abnormalities, dose-limiting toxicities and exposure-response patterns.
- Reporting completeness: distinguish registered, completed, announced and fully reported studies.
Later-stage evidence is more decisive for efficacy because it tests a more defined population and outcome, but design quality still controls how persuasive the result is. A small uncontrolled signal can justify another study; it cannot prove a medicine works.

What the current evidence says about AI-discovered success
Jayatunga, Ayers, Bruens, Jayanth and Meier reported an estimated 80–90% Phase I success rate for AI-discovered molecules in a 2024 analysis published in Drug Discovery Today. Treat “80–90% in Phase I — Jayatunga et al., 2024” as an early estimate, not a mature end-to-end success rate or proof that AI is superior to conventional discovery.
The methods and full denominator details of that estimate were not available in the reviewed full-text material. Before using the percentage as a rigorous comparison, verify the cohort definition, outcome classifications and denominator. “AI-discovered” can describe very different levels of model involvement, so companies may not be comparing like with like.
Andrew Marshall’s November 2025 report in Nature Biotechnology said definitive data on the success of AI-generated drugs were still awaited. It quoted Boston Consulting Group managing director and partner Chris Meier: “From the limited phase 2 data we’ve seen so far, there’s no indication that AI-discovered targets or drugs are necessarily any more successful than classically discovered drugs.” The statement supports caution about claims of superiority; it does not say that AI programs cannot succeed.
FDA said it had reviewed more than 500 drug and biological-product submissions with AI components since 2016. That figure counts submissions containing AI components, not AI-designed candidates, clinical successes or approved AI-discovered drugs. Keep those categories separate when interpreting regulatory adoption.

6. Value the asset separately from the company
Use valuation as a financing and expectations signal, not as a probability-of-success estimate. A high valuation may price in a large market, a partnership, platform optionality or future fundraising power. It can also leave little room for clinical disappointment.
Build an asset-level probability view
For each candidate, record the current evidence, the next value-creating experiment, the time to that readout, the likely failure modes and the capital required. Then ask how much of the company’s valuation depends on this asset versus earlier programs or an unproven platform.
- Evidence risk: target engagement, reproducibility, translational gap and clinical endpoint uncertainty.
- Execution risk: manufacturing scale-up, formulation, trial enrollment, site performance and regulatory interactions.
- Financial risk: cash runway, financing needs, dilution and milestone obligations.
- Competitive risk: standard of care, rival mechanisms, patent barriers and speed of competing development.
- Commercial risk: diagnosed population, treatment duration, reimbursement, delivery setting and access constraints.
Manufacturing, formulation, intellectual property, regulatory path, enrollment, standard-of-care competition and market access can change the company’s prospects without proving that the candidate benefits patients. Keep those analyses in separate columns from biological and clinical evidence.

Comparison is valid only when the candidates are aligned by disease, target context, development stage and evidence type. Do not compare a platform prediction for one asset with a completed controlled clinical outcome for another as though they were equivalent.
| Comparison axis | Questions to ask |
|---|---|
| Biology | Are the targets, mechanisms, biomarkers and patient hypotheses supported by comparable evidence? |
| AI contribution | Did AI discover the target, generate the molecule, optimize it, predict properties or support trial operations? |
| Model validation | Is there a defined COU, independent test set, prospective validation and an error-management plan? |
| Experimental stage | Are results retrospective, biochemical, cellular, animal or human, and were they independently replicated? |
| Clinical quality | How do phase, population, endpoint, comparator, enrollment, duration and reporting completeness differ? |
| Safety and pharmacology | Does exposure reach the proposed therapeutic range without unacceptable toxicity? |
| Development feasibility | Can the molecule be formulated, manufactured, patented and tested within the available resources? |
| Commercial context | What competition, access, pricing, diagnosis and treatment logistics affect eventual adoption? |
8. A practical diligence workflow
- Define the asset: write the molecule, target, indication, formulation, trial stage and intended patient group in one sentence.
- Map AI’s role: document the model output, human decisions, data sources and whether use was prospective.
- Write the COU: state the decision the model supports, allowed inputs, output, performance metric and consequences of error.
- Audit biological logic: trace target engagement to functional activity and to the proposed patient benefit.
- Reconstruct the experiment: record denominators, controls, failed candidates, replication and any conventional benchmark.
- Extract clinical facts: capture phase, population, dose, endpoint, comparator, sample size, follow-up, adverse events and reporting status.
- Separate feasibility: review manufacturing, formulation, intellectual property, regulatory path, enrollment and competition independently.
- Stress-test the valuation: identify which assumptions require a clinical win, how much cash reaches the next readout and what happens if the lead asset fails.
- Assign a decision: proceed, monitor, request more evidence or reject; record the specific evidence that would change your view.
Red flags that deserve a pause
- The company cannot state what the model did or which version produced the result.
- A benchmark uses familiar compounds or data that may have appeared in training, but no leakage analysis is supplied.
- Only successful predictions are shown; the number of synthesized failures is missing.
- “AI-designed” is used interchangeably for target discovery, molecule generation and trial analytics.
- Animal efficacy is presented without exposure, pharmacodynamic or translational information.
- A Phase I safety result is described as proof of efficacy.
- A clinical announcement omits the prespecified endpoint, comparator, enrollment or adverse-event detail.
- The valuation discussion substitutes partnership headlines for asset-level probability and cash requirements.
- The company claims regulatory endorsement from FDA’s review of AI components; FDA’s more-than-500 figure does not mean those submissions were AI-designed successes.
How to report an evidence-based conclusion
Use calibrated language tied to the evidence. Say that AI contributed to a defined task, that a model was or was not validated for its stated COU, that the molecule showed activity in specified experiments, or that a trial produced a particular result. Avoid saying that AI “proves” a target, guarantees a candidate or makes a company undervalued.
A defensible conclusion should answer four separate questions: What did the AI do? Does the biology support the mechanism? What prospective human evidence exists? What valuation assumptions remain after those facts are recognized? If any answer is unresolved, identify the missing evidence and the event that would resolve it.
