Choose a TFF membrane by defining what must remain in the retentate, screening a suitably tighter MWCO, and verifying retention, recovery, flux, and product quality with the real feed. Then scout transmembrane pressure (TMP) and crossflow together: increase TMP only while it adds useful flux, and raise crossflow only when the reduction in concentration polarization justifies its shear, yield, fouling, and cost effects. Manufacturer ratios and example setpoints are starting points, not universal recipes.

What you are selecting in a TFF process

Tangential flow filtration (TFF) pumps feed parallel to a membrane surface. Pressure drives part of the liquid through the membrane as permeate while retained material continues through the retentate path. In ultrafiltration (UF), the target molecule is normally retained and smaller solutes pass. In diafiltration (DF), fresh buffer replaces permeate to exchange the solution around the retained product. TFF can also be used for clarification, concentration, or fractionation, but the correct membrane and operating window depend on which of those jobs you need to perform.

A practical selection has two linked decisions. First, select a membrane that gives the required sieving without sacrificing recovery or quality. Second, establish a TMP and crossflow combination that delivers acceptable flux over the entire run, not just during the first minutes. The Cytiva technical guide and Cytiva cross-flow handbook both emphasize that membrane behavior and operating conditions must be evaluated as an interacting system.

  • Acceptance criteria: minimum target recovery, allowable impurity carryover, concentration endpoint, product-quality limits, and maximum process time.
  • Feed conditions: composition, pH, ionic strength, temperature, solids content, viscosity, and any components likely to foul or bind the membrane.
  • Equipment constraints: available membrane area, cassette or hollow-fiber holder, pump range, pressure sensors, tubing, and module pressure limits.
  • Lifecycle policy: single-use or reuse, cleaning agents, sanitization method, storage, and the number of batches expected from a module.

This definition prevents a common error: choosing an MWCO from the target molecular weight alone while ignoring adsorption, aggregation, viscosity, pressure drop, or the impurities that must actually be removed.

What MWCO should I use?

MWCO is a screening parameter, not a guaranteed molecular sieve boundary. The observed retention of a protein, virus, polymer, or other molecule changes with molecular shape, aggregation, concentration, solution chemistry, membrane structure, and hydrodynamic conditions. Start with a tighter cutoff than the target and confirm performance using your actual feed.

Device format Manufacturer starting heuristic How to use it
Flat-sheet cassette Choose a cutoff 2–3 times tighter than the target molecular weight. Use as an initial screen, then measure target in retentate and permeate.
Hollow fiber Choose a cutoff 3–5 times smaller than the target molecular weight. Treat as hollow-fiber-specific guidance and verify retention experimentally.

The cassette ratio is described in Sartorius cassette guidance; the hollow-fiber ratio appears in its hollow-fiber guidance. These recommendations differ because module geometry and membrane construction affect sieving and mass transfer. Do not combine them into one universal ratio.

Screening procedure for candidate cutoffs

  1. Prepare a representative feed, including the concentration and buffer conditions expected at production scale.
  2. Run at least two candidate MWCOs around the initial heuristic rather than committing to one catalog value.
  3. Measure target concentration in feed, retentate, and permeate. Calculate recovery and apparent retention from measured quantities, not nominal MWCO.
  4. Record permeate flux, TMP, crossflow, temperature, and run time at comparable feed concentrations.
  5. Check product quality after processing for aggregation, activity, structural changes, or other assays relevant to the molecule.
  6. Repeat near the final concentration and during DF, because retention and flux can change as viscosity and contaminant loading change.

Select the loosest membrane that still meets the retention and quality requirement only if the resulting recovery and fouling behavior are acceptable. A tighter membrane may improve retention but can reduce flux and increase processing time; a looser membrane may improve throughput while allowing unacceptable target loss.

How do I choose membrane chemistry?

Material choice should follow product binding, solution chemistry, cleanability, and lifecycle requirements rather than a generic claim that one polymer is best. The Sartorius cassette documentation describes two common families:

Material family Manufacturer-described attributes Questions to verify with your feed
Hydrosart, a stabilized cellulose-based membrane Hydrophilic and described as non-protein-binding. Does the actual product show lower adsorption, adequate recovery, and acceptable cleaning or storage behavior?
Polyethersulfone (PES) Common in biotechnology and pharmaceutical processing; described as offering broad pH and temperature tolerance and high flux. Are the pH, temperature, solvent, sanitizer, and product-compatibility limits suitable for this module and run?

Those are manufacturer attributes, not guarantees for every molecule or process fluid. Run adsorption and recovery studies with the selected membrane, and confirm chemical compatibility in the current module manual. A membrane that is excellent for a dilute protein may perform differently with a viscous, cold, high-solids, or detergent-containing feed.

Cassette or hollow fiber: which format fits?

Compare the physical flow path as carefully as the MWCO. Flat-sheet cassettes provide parallel channels over a defined membrane area and are often convenient when a system already supports cassette holders. Hollow fibers provide many small lumens and can be advantageous when the available pump, scale, or shear environment favors that geometry. Neither format is automatically superior.

Use viscosity and temperature to size the hollow-fiber path

The Sartorius hollow-fiber guidance gives 0.5 mm lumen fibers as an example for standard protein solutions and 1.0 mm lumen fibers for viscous or cold processing. Treat those dimensions as product-family guidance: verify the pressure-drop and flow limits for the exact cartridge. For higher-viscosity upstream feeds, the same guidance advises shorter cartridges to control pressure drop; longer cartridges are suggested for lower-viscosity downstream streams.

Account for shear sensitivity and cleanability

Higher crossflow can reduce concentration polarization but also increases pump work and may expose shear-sensitive products to more stress. Channel dimensions, recirculation time, dead volume, and the ability to flush or clean the device affect recovery. Document whether the module is intended for single use or validated reuse, and do not assume that a chemistry-compatible membrane is automatically compatible with your cleaning cycle.

What must I record before setting TMP and crossflow?

Make the operating study reproducible. Record feed composition, temperature, viscosity when available, starting and final volume, target concentration factor, membrane area, module format, flow-path dimensions, and the assays used for recovery and quality. During every run, capture:

  • Feed or inlet pressure and retentate or outlet pressure.
  • Permeate pressure, if measured by the system.
  • Permeate flow and calculated flux, with membrane area and units documented.
  • Crossflow rate or the pump setting used to establish it.
  • Temperature, concentration, retentate volume, and time.
  • Any module pressure, temperature, or flow limits stated in the current manual.

TMP is the pressure driving permeation through the membrane; crossflow sweeps along the membrane surface. A TMP value without the associated crossflow, fluid, geometry, temperature, and module limits is not a transferable recipe. The Cytiva guide explains why both variables must be considered together.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

What TMP should I run?

Use a controlled pressure sweep rather than copying a number from another process. At a fixed crossflow, increase TMP in small steps and measure the stabilized flux at each step. Plot flux against TMP. At first, flux may increase as pressure rises. Once concentration polarization or a gel layer limits mass transfer, additional pressure produces little extra flux and can worsen fouling or product stress.

The useful region is generally just before that diminishing-return behavior. Cytiva’s handbook describes the objective as examining the interaction between crossflow and TMP and operating before substantial gel-layer accumulation. Stop increasing pressure when the incremental flux gain is small, when pressure approaches the module limit, or when recovery or quality begins to deteriorate.

How do I set the crossflow rate?

Start within the flow range allowed by the selected module, then compare more than one crossflow at controlled TMP values. Increasing crossflow can thin the concentration-gradient layer at the membrane surface. Cytiva states in its Handbook: Cross flow filtration method: “At a given TMP, increasing the cross flow rate helps to reduce the concentration gradient layer and increase flux.” The effect is process-specific: higher flow can increase shear, pump energy, pressure drop, recirculation time, and operating cost.

Crossflow scouting sequence

  1. Choose a conservative crossflow that stays below the module’s pressure and shear limits.
  2. At that flow, perform the stepped-TMP test and identify the point where flux begins to level off.
  3. Repeat the test at a higher crossflow that the product and module can tolerate.
  4. Compare stabilized flux, flux decline, pressure drop, recovery, quality, and energy or pump burden.
  5. Choose the lowest crossflow that controls concentration polarization sufficiently at the selected TMP.

Do not optimize for the highest initial LMH. A slightly lower starting flux that remains stable during concentration and DF may deliver a shorter and more reliable complete batch than a high initial value that rapidly declines.

Step-by-step operating-condition study

  1. Condition the membrane. Follow the current module instructions for wetting, flushing, sanitization, and any storage-agent removal. Record the conditioning buffer and volume.
  2. Establish the flow path. Confirm tubing orientation, pressure-sensor locations, permeate routing, vessel volume, and that no air remains in the module.
  3. Set a baseline crossflow. Use a flow that is inside the module’s stated range and appropriate for viscosity and shear sensitivity.
  4. Load the representative feed. Record starting volume, concentration, temperature, and composition before recirculation changes the material.
  5. Step TMP upward. Allow flux and pressures to stabilize at each step. Record permeate flow, crossflow, inlet and outlet pressure, temperature, and product samples.
  6. Repeat at another crossflow. Keep other variables as constant as practical so the TMP/crossflow interaction is interpretable.
  7. Map the full run. Continue measurements as volume decreases and concentration rises. Note when viscosity, pressure drop, or flux changes.
  8. Run DF separately. Record buffer addition, permeate removal, volume control, and DF flux independently from the concentration phase.
  9. Assay the endpoints. Test retentate and permeate for recovery, purity or impurity removal, aggregation, activity, and any molecule-specific quality attributes.
  10. Define the control window. Set a normal TMP and crossflow range, alarm limits, sampling intervals, and a stop rule for pressure, flux decline, or quality loss.

Why concentration and diafiltration need separate performance records

During concentration, retained material and viscosity rise while available volume falls. During DF, buffer addition changes composition and the permeate is used to exchange small solutes. These phases can have different flux, pressure, and quality behavior even with the same membrane and pump settings.

Track flux against concentration or DF volume, not only against elapsed time. Recheck product recovery at the final concentration and after DF. If flux declines, distinguish reversible concentration polarization from irreversible fouling by examining whether the flux responds to a controlled crossflow change or to the approved cleaning and flush procedure. Do not infer DF throughput from a UF result.

A named application example: useful as a reference, not a recipe

A Sartorius application note describes a Sartocon Slice 200 Hydrosart 300 kDa cassette with 0.2 m² effective filtration area, a 0.8 bar pressure difference (ΔP), target inlet pressure of 1.0 bar, and TMP of 0.5–0.6 bar. In that particular clarified-feed setup, the reported average flux was 131 LMH for UF and 58 LMH for DF, with 16 minutes for UF and 20 minutes for DF. The source is the Sartorius application note.

131 LMH UF and 58 LMH DF average flux — Sartorius, 2026 (application-note publication date shown approximately six months before 2026-10-13; exact date not present in search result). Manufacturer guidance says conditions should be evaluated for each application and cassette type. Confirm exact limits in the current manual for the selected device rather than transferring values from a different product family. The Sartorius TFF overview and Cytiva guide provide background for this comparison.

Concrete laboratory cassette specification

For product discovery, the Vivaflow 200 product page lists a 200 cm² PES membrane cassette with 50 kDa MWCO, maximum operating pressure of 4 bar, recommended feed volume of 1,000–2,500 mL, and recommended flow rate of 200–400 mL/min. These specifications describe that laboratory cassette, not every 50 kDa membrane or every TFF system. Check the current regional specification, compatibility, and instructions before use.

Troubleshooting common selection and operation failures

Target appears in permeate

  • Check whether the MWCO is too open for the molecule’s actual shape or aggregation state.
  • Confirm that the target assay distinguishes intact product from fragments or aggregates.
  • Test a tighter candidate and compare recovery, flux, and product quality on the real feed.

Flux is high initially and then collapses

  • Check concentration polarization or gel-layer formation before raising TMP further.
  • Test a controlled crossflow increase within module and shear limits.
  • Track viscosity, concentration, and foulant accumulation; a change in feed conditions can move the process outside its original window.

Pressure rises faster than expected

  • Verify temperature and viscosity against the conditions used to select cartridge length and lumen or channel size.
  • Inspect tubing, valves, sensors, and the module for blockage or trapped air.
  • Reduce flow or stop the run if the current manual’s pressure limit is approached; investigate the cause before resuming.

Recovery is below the mass balance expectation

  • Measure product in retentate, permeate, hold-up volumes, and flushes.
  • Investigate membrane or tubing adsorption and product instability during recirculation.
  • Compare cassette and hollow-fiber materials only after matching MWCO, area, temperature, and operating history.

UF works but DF does not meet the time target

  • Do not use UF flux as a DF promise; the cited application example reports different average fluxes for the two phases.
  • Check buffer composition, viscosity, temperature, volume-control accuracy, and the number of DF volumes required.
  • Re-scout TMP and crossflow during DF while protecting product quality.

Scale-up and documentation rules

Scale by preserving the variables that control mass transfer and product exposure, not by copying a pressure number. Document membrane area, channel or lumen geometry, crossflow per area or equivalent pump condition, TMP, temperature, concentration profile, residence time, and endpoint assays. Verify that the larger system’s sensors and pump can control the same operating window and that pressure drop remains acceptable at the production viscosity.

Before approving a process, archive the membrane lot or identifier, MWCO, material, module format, conditioning procedure, feed and buffer composition, TMP and crossflow ranges, flux trend, recovery, quality results, cleaning or disposal decision, and deviations. Product specifications and availability can change, so use the selected device’s current documentation when converting a laboratory study into a controlled process.
