Choose the material by the holder’s job, not by filament name. A dry carrier or positioning fixture can often start with FDM PLA or PETG. A part inside the optical path needs a measured fluorescence background. A liquid chamber or glass-bonding surface may need a design-specific MSLA resin process instead of FDM. Any surface touching a specimen, culture medium or cleaning chemistry requires validation of the exact printed material, geometry and workflow.

No polymer is universally best for microscopy holders. The practical decision is to separate external mechanical parts from wetted, optical and sample-contact parts, then test the finished print under the conditions in which it will be used.

Start by defining what the holder actually does

One assembly may contain several printed parts, and each part can have a different material requirement. Write down the holder’s function before choosing a spool or resin.

External carrier or positioning fixture

An external carrier keeps a slide, grid, dish or chamber aligned and provides stiffness, mounting holes or clearance for the microscope stage. It is usually dry and outside the imaging field. Dimensional accuracy, flatness, rigidity, print time and ease of replacement matter more than optical purity. FDM PLA is a sensible prototype choice; PETG is another common candidate when the part needs greater durability or more resistance to handling.

Optical-path component

A printed wall, window surround, spacer or mask can sit close enough to the specimen that its color or autofluorescence enters the camera. The relevant questions are the excitation and emission bands, the material’s color, the distance to the specimen and whether the print is inside the field of view. A part that never appears in the illumination cone has a different risk profile from a black-looking part that is directly illuminated.

Liquid-containing chamber

A chamber must resist leakage, maintain its dimensions, present a smooth sealing surface and, in some designs, bond reliably to glass. Layer gaps, stair-stepping, warping and resin-processing defects can all become leak paths. In a 2022 Mod3D study, the authors found FDM chambers unreliable for their required watertightness, resolution and glass bonding, then used MSLA resin for the chambers while retaining FDM for the carrier. That is a design precedent, not a blanket rule that every resin chamber is suitable.

Sample-contact or culture-medium surface

Contact with cells, tissue, media or a specimen introduces questions that a generic “biocompatible” label cannot answer. You need the exact polymer or resin grade, colorants, additives, print settings, post-processing, cleaning agent, sterilization method, exposure time and biological model. Published holder experiments support only the tested design and protocol.

Material choices at a glance

Material or process Most defensible starting use Advantages Risks requiring validation
FDM PLA Dry prototypes, external carriers and fixtures Simple printing and relatively low printer requirements; easy to iterate Autofluorescence, layer porosity, heat and chemical limitations, and uncertain suitability for direct sample contact
FDM PETG Durable external parts and selected chemical-resistant holders Commonly accessible and generally tougher than PLA in routine handling Fluorescence background, stringing and dimensional effects, plus protocol-specific contact and cleaning compatibility
MSLA resin High-detail chambers or surfaces that need a repeatable glass interface in a tested design Fine features and a study-specific improvement in watertightness and glass bonding over FDM Formulation-dependent chemistry, post-cure requirements, uncured-resin residue, brittleness and biological/cleaning uncertainty
ABS or polypropylene Possible chemical-resistant options where the printer and process support them The NIH 3D TH GridPlate resource lists chemical-resistant materials such as PETG, ABS and polypropylene for its tray context That guidance is specific to a TEM grid-staining tray; it does not establish optical, biological or dimensional performance for your holder

Del Rosario and co-authors described PLA and PETG as the best starting materials for 3D printing because of their simplicity and relatively low printing requirements. “Starting material” does not mean “validated material” for every microscope, wavelength or specimen.

How PLA and PETG differ in a microscopy workflow

PLA: the fast, inexpensive geometry test

PLA is useful when the immediate question is whether a holder fits the stage, slide, grid or objective. It is straightforward to print and economical for multiple geometry revisions. Use it for dry carriers, alignment jigs and proof-of-concept parts that stay outside the optical and wetted regions.

Do not infer optical or biological suitability from printability. PLA can emit fluorescence under imaging conditions, and its layers can leave microscopic gaps or ridges. A PLA prototype that fits perfectly may still need to be replaced for a final chamber, optical mask or sample-contact surface.

PETG: a tougher candidate, not an optical guarantee

PETG is often selected when a carrier must tolerate more handling or when a project wants a commonly available chemical-resistant candidate. It can be a reasonable next step after a PLA fit check. PETG still needs the same fluorescence, leakage, contact and cleaning checks. In a biomedical-imaging study, PLA and PETG emissions could be similar in intensity to tissue signals under the tested optical conditions; PETG background made skin and bone samples indistinguishable in one reported comparison. Those findings cannot be extended to every PETG grade, color, geometry or microscope, but they rule out assuming that clear or translucent PETG is optically neutral.

Check fluorescence before trusting an optical measurement

Material background is a measurement variable, not merely a cosmetic issue. A bright emission from a holder can raise the baseline, obscure weak structures, create apparent signal around the specimen or alter threshold-based segmentation.

What changes the background

  • Excitation and emission bands used by the microscope.
  • Filament or resin color and any pigments, fillers or additives.
  • Distance between the printed surface and the specimen.
  • Whether the surface lies in the illuminated field or only near the edge.
  • Layer orientation, roughness and post-processing, which can scatter light.
  • Exposure, detector gain, filters and the optical path of the specific instrument.

A practical blank-control test

  1. Print the exact material, color, wall thickness, infill and surface finish planned for use. Test a representative coupon as well as the complete holder, because edges and cavities can put material in different parts of the field.
  2. Place the print in the microscope using the same objective, illumination, filters, exposure and detector settings used for the specimen.
  3. Record images with no specimen and with the holder loaded but empty. Measure the mean and peak background in the region where the sample will appear.
  4. Repeat with the intended specimen or a non-fluorescent surrogate. Compare the holder signal with the weakest biological or chemical signal you need to detect.
  5. If the background is unacceptable, move the material outside the field, add an opaque barrier only after checking that the barrier itself is quiet, or redesign the holder. Do not assume that a different color of the same product has identical fluorescence.

The published fluorescence result is a warning to test the finished part under the actual imaging setup. It is not a universal ranking of PLA versus PETG.

Match mechanical and thermal requirements to the printed design

Microscope holders fail mechanically through warping, flexing, creep, poor layer orientation, loose interfaces and insufficient objective clearance. Evaluate the part as a design, not just as a material coupon.

Flatness and stiffness

A carrier that bows can change focus across a slide or shift a grid relative to the objective. Orient critical load paths so that forces do not depend on weak inter-layer adhesion. Increase wall thickness or add ribs before increasing infill indiscriminately; a stiff, well-supported frame can be lighter and more stable than a solid block.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Temperature and dimensional stability

One cited live-cell system used a 4 mm-thick FDM holder with 20% grid infill and reported dimensional stability up to 37 °C. Treat that as a result for that holder geometry, print setup and material combination, not as a universal temperature rating for PLA or PETG. If your microscope enclosure, incubator or illumination raises the part above room temperature, measure the assembled holder at operating temperature.

Clearances and repeatability

  • Check objective nosepiece, condenser, stage insert and cable clearances in every axis.
  • Measure the sample seat, locating pins and screw holes on several prints rather than relying only on nominal CAD dimensions.
  • Check whether repeated clamping permanently deforms the part.
  • Record the print orientation and settings that produced the accepted fit so replacement parts remain interchangeable.

When liquid containment or glass bonding decides the process

FDM deposits material in roads and layers. Even a visually sealed wall can contain microscopic channels, and a nominally flat top surface can have enough texture to prevent a reliable glass bond. Sealants may alter optical background, introduce extractables or complicate cleaning.

The Mod3D authors summarized their FDM chamber result as: “We found this method unreliable, with the resolution not up to the standards required, a poor ability of FDM prints to be watertight, and a glass bonding surface with too many potential imperfections to allow reliable bonding.” Their switch to MSLA resin addressed the requirements of that project. It does not validate every resin, printer, wash sequence, post-cure schedule or adhesive.

Leak test the finished chamber

  1. Inspect the chamber under magnification for layer gaps, pinholes, uncured resin, warping and damage around ports.
  2. Fill it with the intended liquid, or a safe surrogate with similar surface tension, before placing a specimen inside.
  3. Hold the chamber for the longest expected dwell time and inspect the underside, seams and bonded glass for weeping.
  4. Repeat after the planned cleaning and sterilization cycle. A part that is leak-free when new may crack, swell or lose its bond after treatment.
  5. Use a sacrificial chamber for early testing; do not expose valuable samples until the assembly passes repeated tests.

For a glass interface, measure bond width and continuity on multiple parts. If a seal depends on a commercial adhesive, validate the adhesive and printed substrate together rather than treating the resin alone as the seal.

Evaluate sample contact, cleaning and reuse as one workflow

“Biocompatible” is not a sufficient acceptance criterion for a microscopy holder. A material may be acceptable for brief contact in one cell assay and unsuitable after solvent exposure, repeated autoclaving or prolonged immersion.

Document the contact conditions

  • Specimen type: fixed tissue, live cells, protein solution, culture medium or another sample.
  • Contact duration and temperature.
  • Whether the sample touches the printed surface directly or through glass, a membrane or a coating.
  • Cleaning chemicals, concentration, contact time and rinsing method.
  • Sterilization method, including alcohol, ultraviolet exposure, heat or another process.
  • Number of planned reuse cycles and the visual or dimensional checks required before reuse.

A cryo-EM grid-holder study reported reuse up to 15 times without observed adverse effects on sample preparation, but that result belongs to its particular designs and procedures. It is evidence that a reuse protocol can work, not a certification for another polymer, specimen or cleaning cycle.

Inspect after each treatment

Look for swelling, whitening, cracks, delamination, sticky residue, loss of locating accuracy and changes in fluorescence. If a part contacts live cells, include an appropriate negative control and compare cell attachment, viability or morphology with the established holder or vessel used by your laboratory. Do not transfer a result from a fixed-sample workflow to live-cell culture without testing.

Choose FDM or MSLA according to the part

FDM is usually the efficient first pass

FDM supports rapid, low-cost iteration with PLA or PETG and is well suited to carriers, lids, brackets and positioning frames. It is also easier to repair or reproduce in many laboratories. The trade-offs are visible layer texture, anisotropic strength, possible porosity, limited small-feature resolution and the need to control warping.

MSLA can solve a detail or sealing problem, with added process obligations

MSLA can produce finer features and, in the cited chamber project, a more usable surface for watertightness and glass bonding than the tested FDM approach. Resin selection, washing, post-curing and handling are part of the material choice. Residual uncured resin or an unsuitable formulation can compromise a sample-contact workflow, and a fully cured part may still fluoresce or become brittle. Validate the exact resin and post-processing sequence rather than treating “resin” as one material category.

A step-by-step selection workflow

  1. Map the interfaces. Draw the holder and mark every region that touches the stage, objective, glass, liquid, specimen or cleaning agent.
  2. Classify each region. Label it external carrier, optical-path component, liquid chamber or sample-contact surface. Split the design into separate parts when one material cannot satisfy all four roles.
  3. Set the mechanical envelope. Record load, flatness, locating accuracy, maximum temperature, objective clearance and acceptable dimensional drift.
  4. Choose a process candidate. Start dry external parts with FDM PLA or PETG. Consider MSLA for a chamber or glass interface only when its detail and sealing advantages justify resin-specific validation.
  5. Print representative features. Include the thinnest wall, smallest hole, sealing land, locating feature and optical-facing surface in the test piece.
  6. Measure fit and stability. Check dimensions at room temperature and at operating temperature, then repeat after clamping and cleaning.
  7. Run an optical blank. Image the empty holder with the intended settings and quantify background where the specimen will be measured.
  8. Run a containment test. Fill the chamber, inspect for leaks, and repeat after the planned treatment cycle.
  9. Run a contact and cleaning test. Use the real specimen class or a justified surrogate, then inspect the part and compare sample behavior with a known control.
  10. Record acceptance criteria. Save material lot or formulation, color, printer, nozzle or layer settings, orientation, post-processing and the tests the part passed. Approve a material only for the documented use, not for every future holder.

Common failure modes and fixes

Symptom Likely cause Useful response
Unexpected bright signal around the specimen Material autofluorescence, pigment emission or light scattering Run an empty-holder blank under the actual settings; move the print outside the field or change the validated material and color
Slow leak from a chamber wall FDM inter-layer porosity, pinhole, warped seal or damaged bond Inspect under magnification, repeat the leak test and redesign the wall or process; do not rely on an unvalidated coating
Glass bond fails between prints Rough or uneven bonding land, contamination or incompatible adhesive/substrate pair Improve surface control, clean consistently and test bond width and repeatability on multiple parts; consider a design-specific MSLA chamber
Sample shifts during imaging Flexible carrier, loose locating feature, creep or thermal expansion Add ribs or a better locating interface, verify orientation and measure at operating temperature
Part becomes cloudy, cracked or sticky after cleaning Chemical attack, residual uncured resin or incompatible sterilization Stop reuse, identify the treatment that caused the change and validate a different material or cleaning protocol
Cells or tissue behave differently on the holder Surface chemistry, residue, roughness or extractables Compare with a control surface using the intended biological model and test the complete print-and-clean workflow

What published examples actually demonstrate

Mod3D optical chamber and carrier

The Mod3D study is useful because it separates functions. Its FDM approach was not reliable enough for the chamber’s watertightness, resolution and glass-bonding requirements, so the authors used MSLA resin for chambers and FDM for the stiff carrier. The same work reported a 4 mm FDM holder with 20% grid infill that remained dimensionally stable up to 37 °C. Both findings apply to the reported design and setup, not to all FDM parts.

Biomedical-imaging fluorescence study

The reported PLA and PETG emissions demonstrate why a material that looks harmless in room light can interfere with fluorescence. Under that study’s optical conditions, emissions could be similar in intensity to tissue signals, and PETG background made skin and bone samples indistinguishable in one comparison. Use the result to justify a blank-control measurement, not to declare one filament universally unusable.

SEM holder fabrication

A 2017 PLOS ONE study by Meloni and Bertotti reported conductive PLA holders fabricated in under 30 minutes at an average cost below $0.50 per piece. These are study-era fabrication figures for SEM holders; they do not establish current costs, fluorescence behavior or optical-microscopy performance.

Cryo-EM grid holders

Faessler and co-authors reported reuse up to 15 times without observed adverse effects on sample preparation for their cryo-EM grid-holder designs and procedures. The number should be treated as a protocol-specific reuse result, with inspection and cleaning conditions attached.

NIH 3D TH GridPlate guidance

The NIH 3D resource recommends printing its tray in a chemical-resistant material such as PETG, ABS or polypropylene and spraying a water-resistant sealant over the tray wells. That advice is for the resource’s TEM grid-staining tray context. It does not prove that a sealant is optically quiet, biologically suitable or durable for a different microscopy holder.

Decision guide for typical holder parts

Part in your assembly Reasonable first candidate Approval test before routine use
Dry stage carrier or alignment frame FDM PLA; PETG if greater durability is needed Fit, stiffness, thermal stability and repeated clamping
Lid or protective cover outside the field FDM PLA or PETG Clearance, warping, cleaning and mechanical retention
Part near a fluorescence specimen Any candidate only after optical screening Empty-holder background with the exact microscope settings
Watertight chamber with a glass interface Design-specific MSLA resin candidate; FDM only after leak and bond testing Repeated leak, bond, dimensional and post-cleaning tests
Direct specimen-contact insert No default material; select from a validated workflow Contact, residue, sterilization and biological comparison tests

Bottom-line selection rule

Use PLA to prove geometry, PETG when a dry carrier needs more durability, and MSLA resin when a particular chamber design needs detail, watertightness or a repeatable glass interface that FDM cannot provide. For any part near fluorescence, measure the finished print’s background. For any part exposed to liquid, specimens or cleaning, validate the complete material-and-process workflow. The best material is the one that passes those tests for the specific holder—not the one with the most attractive generic label.
