Scientists can recover ancient DNA from a decorated cave wall, but the method is not completely non-invasive. In the 2026 Nature Communications study of Iberian rock art, teams removed very small, carefully chosen samples under archaeological permits and conservation controls. Clean-room extraction and sequencing then detected ancient human mitochondrial DNA in five of 120 samples. The approach is designed for minimal impact—not zero material removal—and a positive DNA result does not identify the person who made nearby art.

What “without damaging the site” means in practice

Ancient-DNA sampling and contact-free documentation are different activities. A camera, structured-light scanner or image-processing system can record a decorated surface without touching it. The French Ministry of Culture’s Chauvet documentation program, for example, describes photographic and image-processing surveys that avoid contact with decorated walls. Those methods preserve visual information; they do not collect DNA.

DNA analysis requires contact with a physical trace. At a protected cave, researchers first obtain permission from the authority responsible for the site, define which panels and materials may be sampled, and follow restrictions for each location. They select the smallest useful fragment—such as a flake of calcite crust or a minute portion of an unpigmented wall surface—rather than scraping a broad area. The resulting disturbance is intended to be minimal and documented, but it is still sampling.

Conservation controls also include recording the sample’s exact position, avoiding visually important details, using clean protective clothing and tools, and preventing visitors or equipment from introducing modern DNA. A site manager can reject a proposed sample if the conservation risk outweighs its scientific value.

What the 2026 Iberian study found

The study examined 24 rock-art panels in 11 caves on the Iberian Peninsula. Across 120 collected samples, only five produced authenticated ancient human mitochondrial DNA:

  • One sample was a pigmented calcite crust from Escoural.
  • Four samples came from unpigmented wall surfaces at Escoural and Covarón.

This low recovery rate is important. Finding ancient DNA on one wall does not mean that every painted panel, cave or artifact will yield a readable genetic signal. The Altamira airbrush sample illustrates the limitation: although a bird-bone tool with pigment was a plausible source of saliva-derived DNA, its very small sampled portion produced no ancient human DNA signal. The investigators note that modern contamination also makes such results difficult to interpret.

Alba Bossoms Mesa, the study’s first author, summarized the evidentiary boundary: “Although we cannot directly connect the traces of ancient human DNA we have found to the creation of rock art, this is the first evidence for human DNA preservation on cave walls for thousands of years.”

How a cave-wall DNA investigation is carried out

1. Define the scientific question and conservation limits

Archaeologists, conservators and geneticists decide what information is needed before anyone enters the cave with sampling equipment. A plan may distinguish pigment-bearing crust from nearby unpigmented stone, include associated sediment or tools for comparison, and exclude fragile or visually significant areas. Permit conditions establish the allowable locations, sample sizes and documentation requirements.

2. Document the panel before contact

Researchers photograph and map the panel, noting pigment boundaries, surface texture, calcite deposits, cracks and signs of water movement. This record serves two purposes: it preserves a visual baseline and lets the team relate a laboratory result to a precise wall context. Contact-free imaging can supplement this record, but it cannot replace the physical sample needed for DNA extraction.

3. Collect a deliberately tiny sample

Sampling is adapted to the material. A pigmented calcite crust may contain mineral growth over or around pigment; an unpigmented area can provide a control for DNA that reached the wall independently of paint. The team removes only the permitted amount, labels it immediately and records its location. “Minimal impact” describes this restricted intervention, not an untouched surface.

4. Control modern contamination

Ancient DNA is usually present in very small quantities and can be overwhelmed by DNA from people, clothing, tools or laboratory surfaces. Staff use protective equipment, dedicated clean areas and procedures that separate ancient-DNA work from modern genetic material. Blank controls and other contamination checks help reveal whether a sequence could have entered during collection or processing.

5. Extract DNA in a clean room

The Iberian work used clean-room ancient-DNA extraction with silica-based chemistry. In simplified terms, the sample is chemically opened, DNA molecules are captured on silica under binding conditions, and inhibitors and debris are washed away before the DNA is recovered. Ancient molecules are often short and chemically damaged, so extraction conditions are optimized for degraded material rather than for the long, intact DNA targeted in many modern tests.

6. Sequence and enrich informative molecules

Extracts are converted into sequencing libraries and read with high-throughput instruments. Because most molecules in a wall sample may be environmental or microbial, targeted enrichment can increase the proportion of reads from human mitochondrial DNA or other mammalian targets. The laboratory then checks sequence quality, damage patterns, duplication and contamination before accepting a result as authentic ancient DNA.

7. Compare the result with the wall context

A genetic signal is interpreted alongside pigment, mineral crust, sediment, tool and water-flow observations. Human DNA alone, human DNA mixed with animal DNA, and a complete negative result each carry different implications. The context prevents a sequence from being treated as proof that an artist stood at that exact spot.

Why researchers sample both painted and unpainted surfaces

Comparison What it can show What it cannot prove
Pigmented surface or pigment-associated calcite DNA preserved in material physically associated with pigment; at Escoural, one pigmented calcite-crust sample contained ancient human mitochondrial DNA. That the DNA came from the person who applied the pigment or was deposited at the same time.
Unpigmented wall A comparison for DNA left by touching, sediment, droplets or other environmental processes; four positive samples came from unpigmented walls at Escoural and Covarón. That a nearby painting or engraving caused the signal.
Human DNA without detected faunal DNA At two Escoural samples, the pattern is consistent with relatively direct human contact. A unique event, a named individual or a direct connection to art-making.
Human and faunal DNA together Mixed signals support possible indirect movement through sediment, water or other environmental pathways. A precise route, date or identity for either source.

These categories are probabilistic clues, not labels that turn a wall into a time-stamped record. DNA may have been deposited when someone touched the stone, when sediment moved across it, or when water carried biological material. The study’s authors emphasize that DNA and pigment could have arrived thousands of years apart.

What the recovered DNA can—and cannot—tell us

It demonstrates preservation on cave walls

The five positive samples show that ancient human mitochondrial DNA can survive in cave-wall material for millennia. Matthias Meyer, the senior author, said the finding “fundamentally changes how we think about where ancient DNA can be found.” It expands the list of possible archaeological reservoirs beyond bones, teeth and sediments.

It may indicate contact or environmental transfer

Human DNA without detectable faunal DNA is compatible with direct human contact, while mixed human-and-animal DNA can be consistent with sediment transfer or water movement. Those interpretations remain compatible with several histories, so the surrounding geology and sampling pattern matter as much as the sequence itself.

It does not identify the artist

A wall sequence is not a genetic signature of the person who painted, blew or rubbed pigment there. Someone could have touched the wall long after the artwork was made, and the same person’s DNA could have arrived from another surface. Mitochondrial DNA is inherited through the maternal line and is less individually distinctive than a complete nuclear genome in any case. The Iberian authors do not claim to name artists, assign a painting to a population, or establish that a particular individual created a panel.

It does not automatically date the artwork

The paper estimates minimum DNA ages of about 1,000 years for one Covarón sample and about 2,000 years for the other samples from molecular damage patterns. These are estimates of how old the DNA molecules may be, not direct calendar dates for deposition and not dates for the art. The authors note that substantially older ages remain possible. A direct connection to the painting would require independent archaeological and chronological evidence.

How scientists reduce risk to decorated caves

  • Permission first: Sampling follows permits and site-specific restrictions set by the responsible authorities.
  • Targeted locations: Teams choose tiny, scientifically informative areas and avoid irreplaceable visual features whenever possible.
  • Baseline records: High-resolution photographs, maps and condition notes document the panel before removal.
  • Contamination barriers: Protective clothing, clean tools, controlled handling and laboratory blanks limit modern DNA.
  • Material accounting: Each fragment is labeled, tracked and retained for repeat analysis or independent verification.
  • Conservative interpretation: Negative results and ambiguous mixtures are reported rather than converted into claims about artists.

These safeguards manage risk; they do not make the process universally harmless. A permit may allow sampling at one cave while prohibiting it at another, and a method suitable for a calcite crust may be inappropriate for friable pigment or a flaking wall.

Why recovery is uncommon

DNA degrades through time, heat, moisture, chemical reactions and microbial activity. Cave walls also vary widely in mineral composition and water exposure. A molecule can be present but too fragmented, too scarce or too contaminated to pass authentication. Sampling may be deliberately tiny to protect the site, reducing the amount available to the laboratory. Sequencing can therefore produce a genuine negative even when people once touched the wall.

The five positives among 120 samples are a measured success rate for this particular set of caves, materials and protocols—not a universal probability for future projects. The unsuccessful Altamira airbrush result reinforces that a plausible DNA source does not guarantee recovery.

How to read a future cave-DNA claim

  1. Check whether the material was actually sampled or only photographed. Imaging is non-contact documentation, not DNA recovery.
  2. Ask how many panels and samples were examined, not just how many produced DNA.
  3. Look for authentication evidence such as ancient-DNA damage patterns, contamination estimates and laboratory controls.
  4. Separate “DNA found near art” from “DNA came from the artist.” The latter requires evidence the cave-wall studies do not establish.
  5. Check whether an age is a direct date, a minimum estimate from molecular damage or an age inferred from associated archaeology.
  6. Read negative results and mixed human-faunal profiles as part of the evidence, because they define how often the method succeeds and how DNA may move.

Where this method fits in cave-art conservation

Ancient-DNA sampling is one tool in a broader, staged investigation. Non-contact photography and image processing can survey an entire decorated surface. Archaeological recording, mineral analysis, pigment chemistry and sediment studies can then provide context. DNA sampling is considered only where the expected information justifies a small, permitted intervention. Keeping those roles separate prevents a conservation-friendly imaging method from being mistaken for a non-invasive genetic test.

The Iberian study’s most defensible conclusion is therefore narrow but significant: carefully controlled micro-sampling can recover authentic ancient human DNA from some cave-wall contexts. It opens a new source of evidence while preserving the distinction between detecting ancient biological traces and proving who made a particular work of art.
