Scientists identify an unknown grain or metallic inclusion in glass by combining methods, not by relying on one instrument. Microscopy shows where the feature is and what it looks like; elemental analysis indicates which elements are present; and methods such as X-ray diffraction (XRD) or Raman spectroscopy can help determine the material’s phase or structure. Those findings can characterize an inclusion or support a comparison, but they do not automatically prove a unique mineral identity or source.

Why one measurement may not identify the material

“What is this inclusion?” can mean several different things. A scientist may need to establish whether a speck is inside the glass or on its surface, describe its texture, determine which elements it contains, identify a crystalline mineral phase, or compare the glass with another sample. Those are related questions, but they are not interchangeable.

An elemental result is a list or pattern of chemical elements detected in the analyzed area. It does not, by itself, show how those elements are arranged in a crystal or establish which mineral phase is present. Likewise, an image can reveal shape and texture without providing a definitive chemical identity. The laboratory chooses methods according to the question, the size and condition of the sample, the spatial scale needed, and the sensitivity required.

This distinction matters especially when the target is tiny. A measurement may combine information from the inclusion and nearby glass, depending on the analyzed volume and instrument. A result should therefore be interpreted in light of where the instrument measured and what it can resolve—not treated as a label produced automatically by a machine.

How a laboratory investigates an unknown inclusion

1. Examine the glass and locate the feature

The first task is to document the sample and find the material of interest. Optical or petrographic examination can record visible properties such as color, shape, and texture, and help assess whether a feature appears to be embedded in the glass or is surface contamination. The appropriate preparation and examination depend on the sample’s condition, its size, and the question being asked; there is no single preparation that fits every case.

Establishing location is important because a surface particle and an internal inclusion can have different implications. It also helps the laboratory decide where to focus later measurements. NIST’s description of forensic glass examination includes physical and optical properties among the criteria used for comparison. That forensic context concerns comparing glass fragments and is related to, but distinct from, identifying one geological grain in glass.

2. Image the feature and examine local chemistry

A scanning electron microscope (SEM) can provide images of a selected area. In particular, backscattered-electron imaging can help characterize texture and distinguish parts of a sample by how they appear in the image. The image guides interpretation, but it is not, by itself, a mineral identification.

Energy-dispersive X-ray spectrometry (EDS), often used alongside SEM, detects characteristic X-rays from the analyzed volume. Those signals indicate which elements are present there. This makes SEM-EDS useful for investigating the composition of a small inclusion in context: the analyst can relate a local elemental result to the imaged feature rather than treating the whole glass sample as a uniform material.

A 2024 USGS study of Meteor Crater impact melts used backscattered-electron imaging and EDS to characterize samples texturally and compositionally, including glass, mineral phases, and metallic inclusions. That is an example of a geological application, not a universal protocol for every kind of glass. Its broader lesson is that image and composition can complement each other while answering different parts of the identification question.

EDS has an important limit: it is not suitable for every trace-element comparison. NIST’s Materials Trace Interpretation Document, surfaced as draft guidance in 2025, cautions that SEM-EDS does not have suitable sensitivity for some trace-element questions and points to more sensitive elemental techniques when enough sample is available. That guidance should be understood as draft material, not presented as a current standard.

3. Determine whether a mineral phase needs to be identified

If the question is not merely “which elements are here?” but “which crystalline phase is this?”, elemental analysis alone may not settle it. X-ray diffraction (XRD) is used to identify crystalline phases. Raman spectroscopy provides molecular or vibrational information that can help distinguish materials whose elemental compositions alone do not resolve the question.

The 2024 USGS impact-melt report illustrates why an additional phase-sensitive method can matter: it notes that distinguishing silica polymorphs is difficult without Raman spectroscopy or another suitable technique. Silica polymorphs share the same basic chemical composition but differ in structure. The example shows why an elemental result can be consistent with more than one phase; it does not mean Raman is required or sufficient in every investigation.

USGS’s 2011 overview of mineralogical analyses describes combined use of methods including XRD, petrography, SEM, and microprobe analysis. The relevant choice depends on what remains unresolved after the initial examination. A lab investigating an apparent metal-rich speck, for example, may first need to establish its location and elemental character; a question about a crystalline mineral phase may call for diffraction or vibrational information as well.

4. Measure glass composition or trace elements when comparison requires it

Sometimes the target is the composition of the glass itself, or a comparison between glass samples, rather than the identity of one inclusion. X-ray fluorescence (XRF), including micro-XRF, and inductively coupled plasma mass spectrometry (ICP-MS), including laser-ablation ICP-MS (LA-ICP-MS), are among the methods used to characterize or compare glass elemental composition.

NIST’s Glass Evidence Analysis program describes XRF and LA-ICP-MS as techniques commonly used by forensic glass analysts. Its work concerns elemental analysis for forensic glass comparisons, not a guarantee that either method will identify an individual mineral grain. NIST also notes that more sensitive elemental methods may be needed for trace-element comparisons when sample quantity permits. The method should match both the scale of the question and the sensitivity it requires.

What each method can—and cannot—tell you

Method Primary contribution Important limit
Optical or petrographic examination Documents physical and optical properties and helps locate and describe a feature. Appearance alone does not establish elemental composition or a unique mineral phase.
SEM imaging, including backscattered-electron imaging Shows the selected area’s texture and provides context for local analysis. An image alone does not establish a definitive chemical or phase identity.
EDS Detects elements in the analyzed volume, often alongside SEM imaging. Elemental detection does not by itself identify crystal structure or settle every trace-element question.
XRD Identifies crystalline phases. It answers a phase-identification question, not every question about glass composition or source.
Raman spectroscopy Adds molecular or vibrational information useful for distinguishing materials and phases. It is one possible phase-sensitive method; the suitable approach depends on the material and question.
XRF, including micro-XRF Characterizes elemental composition and is used in forensic glass analysis. An elemental result is not, by itself, a mineral-phase identification or proof of source.
ICP-MS, including LA-ICP-MS Supports elemental characterization and trace-element comparison; LA-ICP-MS is used in forensic glass analysis. Its use and suitability depend on the analytical question and available sample.

The table describes the roles established in the cited USGS and NIST materials; it is not a universal ranking or a promise that each instrument can analyze every specimen in the same way. Exact spatial scale, preparation, sample consumption, and sensitivity depend on the particular instrument and laboratory protocol. Ask the laboratory about those specifics before deciding how much material to submit or whether a measurement may alter it.

How laboratories choose a method sequence

A useful sequence moves from locating and describing the feature toward whichever unresolved question matters most. A lab may combine methods or change course when an early result shows that a different kind of evidence is needed. The order below is a way to understand the logic, not a prescribed protocol for every sample.

  1. Clarify the objective. Decide whether the priority is to locate an inclusion, characterize its texture, measure elements, identify a crystalline phase, compare glass composition, or assess whether two samples are consistent with one another.
  2. Document the sample and feature. Use appropriate optical or petrographic examination to record visible properties and evaluate whether the target appears internal or at the surface.
  3. Choose a local imaging and chemistry approach if the feature is small. SEM imaging with EDS can connect local elemental information with the appearance and texture of the analyzed area.
  4. Add phase-sensitive analysis when chemistry leaves a structural question open. XRD or Raman spectroscopy may be relevant when identifying a crystalline phase or distinguishing materials that elemental composition alone cannot separate.
  5. Use an elemental method suited to comparison sensitivity. XRF or an ICP-MS-family method may be appropriate when the question concerns glass composition or trace-element comparison. A lab should assess whether the available sample and requested sensitivity support that choice.
  6. Interpret results in combination and state their limits. The conclusion should distinguish what was observed, what was identified, and what comparison—if any—the evidence supports. Similarity in one measurement should not be reported as unique-source proof.

Why a “match” does not automatically prove a common source

Two samples can appear similar in one measurement without that similarity establishing that they came from the same object or location. The evidential meaning depends on what was measured, how discriminating that measurement is, and the comparison framework used. A local EDS result, a phase identification, and a broader glass elemental comparison support different kinds of conclusions.

NIST has noted that standardized interpretation of glass evidence remains an issue. A 2021 NIST interlaboratory study evaluated refractive-index, micro-XRF, and laser-induced breakdown spectroscopy (LIBS) data for forensic comparison of glass samples. Seventeen laboratories participated. That figure is the number of participating laboratories, not an accuracy rate and not evidence that a method identifies every unknown inclusion correctly.

The study recommends using verbal scales and/or background databases where available to express the significance of a comparison. In practical terms, a laboratory report should communicate how strong a comparison is under its stated framework rather than converting a similarity into an unsupported claim of a unique origin. An identification of a phase and an attribution of a source are separate conclusions.

What to ask the laboratory before submitting glass

If you are arranging an analysis, a clear question helps the laboratory choose a useful method and avoid spending sample on a measurement that cannot answer it. Share what is known about the specimen and ask how the proposed analysis relates to the decision you need to make.

  • Is the target thought to be a surface particle, an internal inclusion, or part of the glass itself?
  • Do you need an elemental composition, a crystalline-phase identification, a glass-to-glass comparison, or some combination?
  • Can the selected method analyze the feature at the required spatial scale, or might the result include signal from surrounding glass?
  • Is the method sensitive enough for the elements or trace-element differences of interest?
  • What sample amount and condition are needed, and what preparation will be performed?
  • Could the measurement consume, alter, or limit later use of the sample? The answer is instrument- and protocol-specific.
  • How will the laboratory describe uncertainty and the significance of any similarity to a comparison sample?

These questions are particularly useful when the specimen is small or irreplaceable. The reviewed official sources establish that sample condition and the required sensitivity influence method choice, but they do not provide one set of preparation or destructiveness rules that applies to every laboratory and instrument.

Limits of what current evidence supports

There is no general accuracy percentage for identifying all unknown minerals or metals embedded in every type of glass. Instrument performance and interpretation depend on the material, feature size, method, sample condition, and the definition of a correct result. A laboratory’s validated procedure and the evidence it reports are more meaningful than a single accuracy figure applied across unrelated cases.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

The source examples also have different scopes. The USGS 2024 impact-melt study concerns a geological application; the NIST glass program and its interlaboratory study concern forensic comparisons of glass; and the NIST trace-interpretation document surfaced as draft guidance in 2025. None should be treated as a universal protocol for all unknown glass inclusions. USGS’s 1996 analytical methods manual provides background on method validation and quality assurance, not a current instrument-specific recommendation.
