Penicillin is made by growing a production strain of Penicillium in a controlled, oxygenated fermentation, then recovering the antibiotic from the broth. Manufacturers separate fungal biomass, extract and purify the product, and may form a salt, crystallize it, and dry it. The exact strain, operating conditions, and recovery sequence vary by product and manufacturer; there is no single public recipe that describes all current commercial production.

What happens between the mold and the finished antibiotic?

Penicillin production has two broad parts. In the upstream part, a selected production culture is expanded and grown in a large fermentation process so it produces the desired antibiotic. In the downstream part, the antibiotic is recovered from the fermentation broth, separated from fungal cells and other materials, and processed into a form suitable for further pharmaceutical manufacturing.

The fermentation broth is not a finished medicine or a purified active pharmaceutical ingredient (API). It contains the producing organism, components of the growth medium, and other material that must be removed or controlled. Recovery therefore matters as much as production: the process must obtain the antibiotic without compromising its quality, and it must control impurities and contamination throughout the operation.

Public descriptions explain representative process stages, but not a universal industrial recipe. Commercial methods can differ in production strain, nutrients, feeding approach, recovery solvents, and purification sequence. Exact industrial process technology may be confidential, so a figure or procedure published for one example should not be treated as a current standard for every manufacturer.

How the fermentation stage works

1. Establish and expand the production culture

Manufacturing begins with a selected production strain and a seed-culture expansion. The seed train is the sequence used to grow enough culture to inoculate the production fermentation; it is not simply a matter of putting an arbitrary mold into a tank. The identity and handling of the production organism are part of controlled manufacturing.

FDA guidance for active pharmaceutical ingredient (API) good manufacturing practice describes cell-bank establishment and maintenance as upstream controls for APIs made by classical fermentation. The production strain and detailed expansion scheme are manufacturer-specific. Public information does not establish one strain or one seed-train schedule for all penicillin plants.

2. Grow the organism in an oxygenated fermentation

Penicillin production is commonly described as submerged fungal fermentation: the organism grows in a liquid medium while the process supplies oxygen and manages nutrients. Oxygen transfer and nutrient control matter, but the conditions are process-specific. A general description should not be mistaken for a tank recipe, because the public sources do not establish one set of values that applies to every strain and product.

Public process overviews describe media containing carbon sources, nitrogen sources such as corn-steep liquor or other nitrogen-bearing materials, and minerals. The recipe is not just a list of ingredients. The organism, medium, oxygen supply, and process controls operate together, and manufacturers may use different feeding strategies. The available public descriptions do not justify publishing a single nutrient concentration, aeration rate, or fermentation schedule as the commercial norm.

3. Direct which penicillin is produced

Precursor selection can change the penicillin side chain. Phenylacetate is associated with penicillin G, while phenoxyacetic acid is associated with penicillin V. These examples show why “penicillin” does not necessarily mean one identical product or one interchangeable production procedure. The precursor is one part of a product-specific process, not a complete recipe by itself.

Penicillin G and penicillin V are useful examples for understanding the role of precursor choice, but their processes should not be collapsed into one set of instructions. The production strain, medium, feeding strategy, and later recovery steps may also differ. No exact commercial conditions can be inferred from the precursor relationship alone.

How penicillin is recovered and purified

4. Harvest and clarify the broth

After fermentation, the product is recovered from the broth. The first downstream task is to separate fungal biomass and other solids, for example by filtration. This clarification step removes the producing organism and suspended material so the antibiotic can be handled in subsequent recovery operations.

Harvest is a quality-sensitive transition. The process must limit product loss and preserve product quality while separating biomass and solids. It must also control contamination. The specific harvest conditions and equipment are not established as universal by the public process descriptions.

5. Extract the antibiotic and remove unwanted material

A representative classical recovery sequence acidifies the clarified broth and transfers penicillin into an organic solvent. This extraction separates the antibiotic from at least some of the water-based broth components. Further processing removes impurities or pigments, with additional pH adjustment or salt addition used to move or precipitate the antibiotic in a form suitable for isolation.

This is a schematic description, not a direction to carry out an extraction outside a regulated manufacturing setting. The solvent, pH values, number of extraction stages, and purification sequence depend on the process. The public material does not establish one solvent system or operating condition for current commercial plants.

Purification has to remove more than visible solids. FDA’s ICH Q7 API guidance discusses removing the producing organism, cell debris, media components, and other impurities while preserving product quality. A process cannot be judged solely by whether it yields a solid at the end: the quality and consistency of the recovered material also matter.

6. Form a salt, crystallize, and dry when required

Depending on the product and manufacturing process, downstream operations may include salt formation, crystallization, and drying. These stages provide ways to isolate and handle the product in an appropriate form. The exact sequence and final form depend on what is being manufactured; public descriptions do not support treating every penicillin product as having the same salt-formation or crystallization procedure.

Physical processing is part of API manufacture, alongside packaging and quality testing. The release requirements and validated controls are product-specific. A public account of one crystallization sequence does not establish the specifications used to release a different product or a different manufacturer’s batch.

Why published conditions are not a universal recipe

The University of Bristol’s educational account gives one historical or illustrative process: aerobic fermentation for 4–5 days at 25–27°C, followed by acidification, solvent extraction, decolorization, potassium acetate precipitation, crystallization, and drying. Those details describe that account. They are not evidence of a universal or confirmed current commercial recipe.

An academic overview notes that commercial producers regard exact industrial penicillin process technology as highly confidential. That distinction is important when reading numerical conditions online. A temperature and duration tied to an educational or historical example should remain attributed to that example; it should not be repeated as an industry-wide operating specification. The same caution applies to the choice of solvent, nutrient recipe, fermentation controls, and purification sequence.

Public descriptions are still useful for understanding the process at a high level: culture expansion, aerobic fermentation, biomass separation, extraction, impurity removal, and possible salt formation and crystallization. They explain what the stages accomplish without disclosing a single standardized process that all manufacturers follow.

How quality and containment shape manufacture

Quality controls span the whole process

Quality is controlled throughout manufacturing, not only by testing the final powder. FDA’s ICH Q7 guidance addresses cell banks, inoculation and culture expansion, critical fermentation parameters, harvest, impurity removal, bioburden monitoring where appropriate, and consistent recovery quality. In practice, these controls connect the identity and condition of the starting culture with the quality of the recovered API.

Impurity specifications are also a regulatory concern. FDA’s draft guidance issued in April 2026 addresses organic impurities in antibiotics made by fermentation and semisynthesis. It is explicitly nonbinding and marked “Not for implementation,” so it should not be described as an effective manufacturing rule. EMA’s adopted guideline covers related-impurity specifications for human and veterinary antibacterial substances that are fermentation products or semisynthetic derivatives. The EMA page lists its current effective version as first published and last updated on 13 July 2012.

These FDA and EMA documents concern quality specifications; they do not prescribe one universal fermentation recipe. Their scope should not be confused with a detailed plant operating procedure or with a claim that all manufacturers use the same impurity limits.

Penicillin requires special attention to cross-contamination

Penicillins are highly sensitizing materials, so containment and separation are important manufacturing concerns. FDA’s ICH Q7 guidance recommends dedicated production areas for highly sensitizing materials such as penicillins. Separately, FDA explains that U.S. CGMP regulations require adequate separation of penicillin-manufacturing facilities and air-handling systems from facilities used for other drugs.

This separation addresses the risk that penicillin could contaminate other products or manufacturing areas. It is a facility and process-control issue, not a step that can be replaced by testing only the final penicillin API. Requirements should be described in their regulatory context: the cited separation statement concerns U.S. CGMP, while the broader guidance discusses quality practices for APIs.

What changed during penicillin’s historical scale-up

The FDA’s historical account says that 21 penicillin production plants came online in the United States during the wartime scale-up period. It also reports historical improvements of 100-fold in yield and 85-fold in purity from improvements to the mold and its cultivation. These are figures about historical development, not benchmarks for present-day plant performance or a prediction of yield from any particular fermentation.

The historical figures illustrate why strain and cultivation development mattered: production depended on more than discovering a mold that made penicillin. But they do not reveal present commercial yields, current plant capacity, or the conditions under which a specific modern product is made. Those details are not established by the historical account.

Putting the process together

  1. Prepare the production culture: maintain a controlled production strain and expand it through seed culture.
  2. Ferment: grow the organism in a submerged, oxygenated process with a managed medium and product-specific precursor selection.
  3. Harvest: separate fungal biomass and other solids from the broth while protecting product quality.
  4. Recover: use pH-controlled extraction and purification to separate the antibiotic from broth components and impurities.
  5. Finish and test: where applicable, form a salt, crystallize, and dry the product, then carry out product-specific quality controls and packaging.

The sequence captures the logic of penicillin manufacture without implying that every plant uses identical equipment or operations. The essential distinction is between biological production in fermentation and pharmaceutical recovery after harvest: both stages must be controlled to obtain a consistent API.
