Norman Dovichi’s contribution to human genome sequencing was part of a collaborative effort to turn analytical chemistry into practical sequencing instruments. In a 2013 interview, he described how his group and a Hitachi group independently developed capillary-array electrophoresis instruments using a sheath-flow cuvette; the instruments were patented, licensed to Applied Biosystems, and commercialized as the Model 3700 DNA sequencer.
Why analytical chemistry mattered to genome sequencing
Sequencing depends not only on identifying DNA, but also on separating and detecting the labeled fragments used to read it. Analytical chemists developed methods that could handle large numbers of fragments at high throughput, helping make sequencing instrumentation practical for the human genome project.
Dovichi placed this work within a broader community effort. As he put it in Neil Withers’s 2013 Chemistry World interview: “The analytical chemistry community played a fundamental role in the genome project, perhaps starting with Lloyd Smith and Leroy Hood’s development of fluorescent-based DNA sequencing.”

How the capillary-array instruments worked
In the approach Dovichi described, many fluorescently labeled DNA fragments were separated by capillary-array electrophoresis. Spectrally resolved laser-induced fluorescence detected the separated fragments. The separation and detection methods worked together: electrophoresis sorted fragments, while fluorescence provided a way to observe their labels.

Dovichi said his group and a Hitachi group independently developed instruments based on a sheath-flow cuvette. The instruments were patented and licensed to Applied Biosystems, which commercialized the technology as the Model 3700 DNA sequencer. Dovichi called it “the workhorse instrument used in the human genome project.” This account describes his group’s role alongside other contributors; it does not make him the sole inventor of DNA sequencing or the genome project.
What Dovichi’s other research illustrates
The sequencing work was one application of a wider interest in sensitive separations and detection. Dovichi described combining capillary electrophoresis with laser-induced fluorescence, reasoning that fluorescence detection in a small volume could be highly sensitive. He reported that, for a fairly wide range of analytes, the method could detect “a few hundred molecules.” That is his statement in the 2013 interview, not an independently verified performance specification.

Single-cell analysis
Dovichi also discussed using sensitive analytical methods to study individual cells and cell-to-cell heterogeneity. He said this work proved more challenging than developing technology for the genome project. The distinction is important: sequencing involved separating many labeled DNA fragments, whereas single-cell analysis asks researchers to measure scarce material in individual cells and characterize differences between them.

Peptide analysis and proteomics
In another research direction, his group combined capillary electrophoresis with mass spectrometry for peptide analysis. Dovichi reported detection limits in the high zeptomole range for many peptides. He framed capillary electrophoresis-based bottom-up proteomics as a challenge to the chromatography-based paradigm, but described it as ongoing research—not as a field-wide replacement for chromatography.

How the methods differ by research goal
| Research context | Separation or detection approach | Sample or target | Purpose |
|---|---|---|---|
| Genome sequencing instrumentation | Capillary-array electrophoresis with spectrally resolved laser-induced fluorescence | Many fluorescently labeled DNA fragments | Separate and detect fragments for high-throughput sequencing |
| Single-cell analysis | Sensitive analytical methods; the interview does not specify one single method for all of this work | Individual cells | Investigate cell-to-cell heterogeneity |
| Peptide analysis and proteomics | Capillary electrophoresis coupled with mass spectrometry | Peptides | Analyze peptide samples in bottom-up proteomics research |
These are research contexts, not competing products. The appropriate separation and detection strategy depends on what is being measured and the scale of the sample.

Neil Withers’s Chemistry World interview, published 21 March 2013, reports that Dovichi received the 2012 Royal Society of Chemistry Robert Boyle Prize for Analytical Science at that year’s Pittcon. It describes the prize rationale as including pioneering ultrasensitive separations, first separations at zeptomole and yoctomole levels, and capillary-electrophoresis-based DNA sequencing for the human genome project. Those descriptions are the interview’s account of the recognition, not independently assessed measurement claims.
