Lensless imaging does not capture a conventional, already-focused microscope picture. In a common holographic approach, light scattered by a cell interferes with unscattered reference light at a nearby digital sensor. The sensor records that interference pattern, called a hologram; software then uses the measurement and the setup’s geometry to calculate an estimate of the cell image at the sample plane. Depending on the method, the result can be a refocused two-dimensional image, a phase estimate, a cell trajectory, or a three-dimensional map of refractive index.
What the sensor records—and what the computer reconstructs
A standard camera photograph is formed by a lens that focuses light from a scene onto a sensor. In lensless holographic microscopy, the sample sits close to the sensor and the system instead records the light pattern formed after illumination interacts with the sample. Some light passes through without being scattered; some is scattered or altered by the cell. At the sensor, those portions of the light interfere, producing a pattern that encodes information about the sample but does not look like a familiar, in-focus cell image.
The recorded pattern is an intensity measurement. It does not directly give the complete optical field—the amplitude and phase of the light—as a focused photograph might suggest. Reconstruction is therefore a computational estimate: software applies a model of how light travels between sample and sensor to infer what field could have produced the measurement. The output is not a hidden photograph simply revealed by sharpening; it depends on the measured data, the optical geometry, and the reconstruction method.

How a hologram becomes a cell image
1. Illuminate the sample and record the interference pattern
A lensless setup illuminates a cell and places a CMOS or CCD sensor nearby. Light that is scattered or otherwise changed by the sample reaches the sensor alongside unscattered light. Their interference creates the hologram: a diffraction pattern whose details reflect the optical effect of the cell and the distance and arrangement of the components.
The sensor measurement is the starting point, not the final image. The compact chip-sized microscope described in the Journal of Imaging study, for example, used a micro-LED display and a CMOS camera. That is one specific implementation, not a universal parts list for lensless imaging.
2. Use the system geometry to numerically propagate the light
For a basic holographic reconstruction, software numerically propagates the recorded optical information back toward the sample plane. In effect, it calculates how the measured diffraction pattern would look if the field were propagated in the opposite direction through the modeled setup. This can numerically refocus the cell image without moving a conventional objective lens.
The geometry is part of the calculation. The sensor-to-sample arrangement and illumination setup affect the propagation model and the resulting reconstruction. A reconstruction algorithm cannot make measurements from one geometry equivalent to measurements from another merely by producing an image that looks similar.

3. Address the missing phase information and artifacts
Because a single intensity measurement does not directly supply the full complex optical field, reconstruction can be ambiguous. Inline holography can produce a twin-image artifact: an unwanted contribution that complicates the estimated image, particularly when interpreting amplitude or phase quantitatively. The chip-sized microscope study describes backpropagation and iterative twin-image suppression, while a separate study investigates sparsity-assisted iterative phase retrieval for biological cells.
Phase retrieval uses computational constraints and repeated refinement to estimate a field consistent with the measurement and the method’s assumptions. It can improve a reconstruction and help reduce artifacts, but the available studies do not establish one universally best correction method. A reconstruction should therefore be judged in light of its algorithm and intended use, not only by its appearance.
4. Acquire additional measurements when the target is more than a refocused plane
Some systems make multiple measurements rather than trying to infer everything from one hologram. The 2024 wavelength-scanning Fourier ptychographic diffraction tomography (wsFPDT) study records diffraction patterns under on-axis illumination at wavelengths from 430 to 1200 nm, then applies iterative ptychographic reconstruction. Its intended output is a three-dimensional refractive-index distribution, not just a refocused two-dimensional image.

What different lensless methods can produce
“Lensless imaging” names a family of methods, not one camera configuration or reconstruction algorithm. The examples below illustrate why measurements from different systems should be reported with their method and output type.
| Approach and study | Acquisition and reconstruction | Reported result | What the figure applies to |
|---|---|---|---|
| Chip-sized holographic microscope, Journal of Imaging study | Micro-LED illumination and CMOS camera; numerical backpropagation with iterative twin-image suppression. | 2.19 μm resolution at a 7 mm light-source-to-camera distance. | The reported instrument; the study demonstrates yeast-cell imaging from a wine-fermentation sample. It is not a general specification for lensless microscopes. |
| Wavelength-scanning Fourier ptychographic diffraction tomography, Wu and colleagues, Light: Science & Applications, 2024 | On-axis illumination scanned across wavelengths from 430 to 1200 nm; iterative ptychographic reconstruction. | 775 nm half-pitch lateral resolution; 5.43 μm axial resolution; 29.85 mm² field of view. | That specific wsFPDT setup, which reconstructs a 3D refractive-index distribution and includes imaging of mouse mononuclear macrophage cells. |
| Pixel-super-resolution lens-free microscopy, Biomedical Optics Express study, 2010 | Uses subpixel shifts to reconstruct detail beyond the sensor pixel-size limitation. | Approximately 0.6 μm spatial resolution over approximately 24 mm². | The reported pixel-super-resolution system. These approximate figures should not be treated as the performance of the other methods in this table. |
| Automated lens-free cell-motility analysis, Scientific Reports study, 2014 | Uses sensor-recorded holograms for living-cell tracking and automated analysis. | A 20-hour experiment; an average of more than 47 cell paths extracted per experiment. | The study’s tracking experiments and analysis. These figures describe cell trajectories, not spatial resolution. |
These values are not a controlled ranking. The systems differ in illumination, reconstruction, acquisition strategy, output, and experimental target. For example, a half-pitch lateral resolution and an axial resolution describe different dimensions of a 3D result; they should not be collapsed into one number or compared as if they measured the same thing.
How lensless imaging has been used with living cells
Yeast cells in a fermentation sample
The chip-sized holographic microscope study demonstrates imaging yeast cells from a wine-fermentation sample. Its reported 2.19 μm resolution is tied to that system’s stated 7 mm light-source-to-camera distance. The example shows how a compact arrangement can capture cell-related optical information and computationally reconstruct it; it does not establish that every yeast cell, sample, or lensless setup will yield the same level of detail.
Cell tracking over time
In the 2014 study, lens-free microscopy was used to track living 3T3 cells over a 20-hour experiment. The authors report an average of more than 47 cell paths extracted per experiment. This is a different use of reconstruction from measuring a cell’s detailed internal structure: the useful output is a record of movement over time. It illustrates why a system’s intended analysis—such as tracking—can matter more than whether it produces the highest resolution figure among unrelated systems.

Three-dimensional cell morphology
The 2024 wsFPDT study demonstrates reconstruction of 3D refractive-index data and includes mouse mononuclear macrophage cells. A refractive-index volume provides quantitative optical information from which morphology can be analyzed; it is not interchangeable with a conventional bright-field image or a 2D amplitude image. The study reports 775 nm half-pitch lateral resolution, 5.43 μm axial resolution, and a 29.85 mm² field of view for its specific setup.
Lens-free on-chip methods have also been discussed for cell and tissue imaging at the point of care and in low-resource settings in a review of computational microscopy. That broader application context does not mean every system is portable, validated for diagnosis, or suitable for every clinical workflow; those conclusions require evidence about a particular device and use.

A resolution number is meaningful only with the method and experimental conditions that produced it. A study might report lateral resolution, axial resolution, or a spatial-resolution measure for a 2D image. Those terms refer to different capabilities. A 3D method needs its lateral and axial performance kept distinct, while a cell-tracking study may report paths and duration rather than a resolution result.

- Method and output: Is the result a 2D refocused image, an amplitude or phase estimate, a trajectory, or a 3D refractive-index volume?
- Illumination and acquisition: Is it a single hologram, a set of subpixel-shifted measurements, or measurements collected across wavelengths?
- Geometry: What sample-to-sensor or light-source-to-camera arrangement is reported? Do not transfer a distance from one instrument to another.
- Resolution definition: Is the figure lateral, axial, half-pitch, or another spatial-resolution measure? Keep the authors’ definition and units.
- Field of view: Is the quoted area measured for the same acquisition and reconstruction as the resolution value?
- Sample and task: Was the system demonstrated on yeast, macrophages, 3T3 cells, or another sample, and was the goal imaging, morphology, or tracking?
- Artifact handling: Does the method address twin-image artifacts or recover phase, and what reconstruction approach does the study actually report?
For example, the approximately 0.6 μm spatial resolution over approximately 24 mm² reported in the 2010 pixel-super-resolution study describes a system using subpixel shifts. It should not be presented as the resolution of the 2024 3D tomography method, nor compared directly with the 2014 study’s cell-path count. Each result answers a different question.
What the method can—and cannot—establish
Lensless reconstruction can turn a sensor pattern into useful cell imagery without forming the image through a conventional objective lens. Depending on the setup, it can support label-free cell imaging, quantitative phase or refractive-index estimation, or automated motion tracking. These are research demonstrations of particular systems and analyses, not proof that every reconstructed feature has the same biological meaning across instruments.
The principal interpretive caution is that reconstruction is an inference from measured light. A hologram can contain twin-image artifacts; algorithms attempt to address ambiguities, but results depend on assumptions and implementation. Likewise, a compact camera-and-illumination arrangement is not automatically a validated microscope for diagnosis or a plug-and-play equipment recipe. The cited compact study identifies a CMOS camera, but does not establish that any generic retail USB camera will be compatible or validated for the same performance.
Recommended Free Tools
When reading a cell image, ask what was directly measured, what the algorithm estimated, and what the authors validated. If the claim concerns a quantitative phase or refractive-index value, look for the reconstruction method and the relevant validation in that study. If it concerns temporal behavior, distinguish a cell trajectory from a resolved structural image. This keeps the interpretation aligned with what the system actually acquired.
The practical takeaway
In holographic lensless imaging, a nearby sensor records interference and diffraction created when light interacts with cells. Computational propagation reconstructs an estimated image at the sample plane; iterative phase retrieval or more extensive measurements can address ambiguities or recover additional information. The term covers multiple approaches, so a resolution figure is meaningful only when tied to its instrument, acquisition, reconstruction, sample, and output.
