Gene-synthesis screening reduces the chance that a designed DNA or RNA order will be used for harmful purposes by checking both the sequence being ordered and the legitimacy of the customer and stated use. The process can flag sequences of concern (SOCs), pause or escalate questionable orders, and create an auditable record. It makes misuse harder; it does not guarantee that every dangerous order or actor will be detected.

In the United States, the best-known requirements are procurement conditions attached to federally funded life-sciences work, not a universal law governing every laboratory or supplier. The 2024 federal framework is also subject to revision under Executive Order 14292, so organizations should verify the current framework and award terms before placing orders.

What gene-synthesis screening checks

Screening happens when a customer orders synthetic genetic material or, in some cases, purchases equipment capable of making it. Two complementary questions are asked:

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.
  • Sequence screening: Does the requested DNA or RNA contain a sequence, or a translated amino-acid sequence, associated with a sequence of concern?
  • Customer screening: Is the purchaser identifiable and legitimate, and does the stated research use make sense for the order?

A sequence match alone does not prove malicious intent. Conversely, a customer who appears legitimate does not make a concerning sequence harmless. Combining both checks gives a supplier more context for deciding whether to fulfill, seek clarification, escalate, or report an order.

Why screening occurs at the order point

The supplier is the last common checkpoint before a designed sequence enters a laboratory. At that point the provider has the sequence, customer information, shipping details, and declared purpose together. A review can therefore stop or delay a transaction before synthesis, rather than relying only on downstream inspections after material has been delivered.

The 2023 guidance from the U.S. Department of Health and Human Services (HHS) treats screening as a risk-control measure. It is intended to deter misuse, identify suspicious combinations of sequence and customer information, and support follow-up when an order raises concerns.

How a screened order moves through the process

1. The provider receives the sequence and order information

The supplier collects the requested construct or fragments, customer identity, institutional affiliation, contact and shipping information, and the declared purpose. Orders for synthesis equipment may trigger a similar legitimacy review even when no sequence is submitted.

2. Software and reference databases examine the sequence

Screening tools compare the order with curated sets of sequences of concern. Depending on the provider’s method, the comparison can include nucleotide sequence and a translation into amino-acid sequence. The Gene Synthesis Screening Information Hub FAQ describes the 2024 framework as covering synthetic DNA or RNA at least 200 nucleotides long; organizations should confirm the length rule in any replacement framework.

3. A potential match is reviewed in context

A flag is a prompt for human or policy review, not an automatic finding of wrongdoing. Reviewers may examine whether the match is meaningful, whether the order is split across multiple submissions, whether the customer has the facilities and expertise claimed, and whether the proposed work is consistent with the institution’s mission and approvals.

4. The provider decides whether and how to proceed

Possible outcomes include requesting additional documentation, seeking confirmation from an institutional biosafety or compliance official, declining the order, or escalating it to the appropriate authority. The federal framework calls for reporting potentially illegitimate orders, while applicable privacy, contractual, and legal requirements govern what information can be shared.

5. Records and security controls preserve an audit trail

Providers are expected to retain records that show what was screened, how a concern was handled, and which customer information supported the decision. Cybersecurity and information-security measures protect sequence data, customer identities, screening results, and any reports from unauthorized access.

What the 2024 U.S. federal framework required

The 2024 Office of Science and Technology Policy (OSTP) framework set six actions for providers of synthetic nucleic acids and manufacturers of benchtop synthesis equipment that wanted to serve covered federally funded research.

Framework action What it means in practice
Attest to implementation Provider or manufacturer confirms that it has implemented the framework’s controls.
Screen synthetic nucleic-acid purchase orders for SOCs Orders are checked against an appropriate sequence-of-concern process before fulfillment.
Screen customers placing SOC-containing orders and purchasers of benchtop equipment The organization verifies identity, affiliation, intended use, and other legitimacy information when risk indicators appear.
Report potentially illegitimate orders Suspicious transactions are escalated or reported through the channels required by the applicable policy.
Retain records Screening decisions, customer checks, and related documentation are kept for later review.
Apply cybersecurity and information-security measures Systems and procedures protect sensitive sequence, customer, and incident information.

The framework is a procurement standard: it tells a covered federal buyer to use providers and equipment manufacturers that adhere to these measures. It is not, by itself, a blanket operating license for every synthesis company or a comprehensive criminal statute.

Does the Framework apply to all labs, regardless of funding sources?

No. The question comes from the Gene Synthesis Screening Information Hub FAQ, which describes the 2024 U.S. framework in the context of federal procurement. Its direct force is tied to federally funded life-sciences research and the award or contract terms that incorporate the requirement. A privately funded laboratory is not automatically subject to the federal procurement condition solely because it orders synthetic DNA.

Other obligations can still apply. A private institution may adopt the framework voluntarily, a state or institutional policy may impose its own screening rule, and a project can receive federal money through an award with specific terms. Laboratories should therefore read the terms of their own grant, cooperative agreement, contract, and institutional compliance policies rather than infer coverage from funding labels alone.

NIH implementation and documentation

The National Institutes of Health (NIH) stated that, for covered NIH awards, agreements, and contracts, its funds could be used only to procure synthetic nucleic acids or benchtop synthesis equipment from providers or manufacturers adhering to the framework. NIH identified April 26, 2025 as the start date for those procurement expectations.

Recipients were also told to retain documentation showing provider or manufacturer adherence. A practical record may include the supplier’s attestation, the framework version referenced, the date the status was checked, and procurement records linking the purchase to the covered award. The exact records required can vary with award terms, so the notice and current contracting language control.

What changed after Executive Order 14292

Executive Order 14292, issued May 5, 2025, directed OSTP to revise or replace the 2024 screening framework. It also directed life-sciences funding agencies to ensure that procurement uses providers and manufacturers adhering to the updated framework. The order included a 90-day direction to act; that deadline does not, by itself, prove that a replacement document was published.

ASPR’s framework status page says it will be updated when a revised or replacement framework is available. Treat the 2024 OSTP document as the existing published framework described above, but check ASPR, OSTP, NIH, and the relevant agency notice immediately before relying on it. A new framework could change sequence-length thresholds, covered equipment, attestations, reporting channels, or effective dates.

What industry screening can look like

The International Gene Synthesis Consortium (IGSC) describes its members’ practice as screening the complete DNA and translated amino-acid sequences of every double-stranded gene order against its curated Regulated Pathogen Database, while also vetting customers. This is a description of consortium members, not evidence that every supplier uses the same database, coverage, or review process.

IGSC defines biosecurity as “measures taken to prevent or stop misuse or exposure of harmful biological agents.” Sequence screening is one biosecurity layer. Physical security, inventory controls, personnel reliability, biosafety procedures, information security, and responsible research oversight address risks that an order-level check cannot see.

How NIST testing relates to screening quality

The National Institute of Standards and Technology (NIST) provides test resources for evaluating screening systems. Its reported resources include:

Resource Reported specification What it does not establish
Benchmark dataset 200-base-pair sequences with known performance metrics. It is not a certification of a supplier or a universal performance guarantee.
Monthly test set, beginning August 2025 1,000 sequences: 200 true positives, 200 true negatives, and 600 ungraded sequences. The 600 ungraded sequences are not false positives or false negatives; the set does not validate every provider or every possible sequence.

These resources let participating providers test detection and reporting behavior against common material. They should be read as measurement tools, not as proof that a system catches all harmful designs or that two systems are equivalent.

How to evaluate a provider’s screening program

A laboratory choosing a supplier, or a procurement office documenting compliance, can ask for evidence against the following criteria:

Best Value
3D Molecular Designs Flow of Genetic Information Kit 1-Group Set: Educational Biology Kit - DNA Replication, Transcription, Translation - High School & Undergraduate Students
  • Identify essential enzymes like helicase and polymerase
  • Model replication of the leading and lagging strands of DNA
  • Explore transcription as they copy one strand of DNA into mRNA using an RNA polymerase
  • Engage in translation/protein synthesis as they decode the mRNA into protein on the ribosome placemat
  • Reenact the different results of the Meselson and Stahl experiments
Evaluation axis Questions to ask
Sequence coverage Are complete orders screened? Are both DNA and translated amino-acid sequences considered? What minimum length and order types are covered?
Customer legitimacy What identity, affiliation, intended-use, and equipment-purchaser checks are performed? Who handles unresolved discrepancies?
Flagged-order follow-up Is there a documented review and escalation path? Can an institutional biosafety or compliance contact respond securely?
Reporting and records What events are reported, to whom, and how long are screening and decision records retained?
Cybersecurity and information security How are sequence files, customer data, reviewer notes, and reports protected, accessed, and deleted?
Framework attestation Does the provider publicly attest to the applicable federal framework, and is the attestation current for the award’s effective period?

No single public comparison ranks providers on all of these dimensions with a common, current methodology. A procurement team should request the supplier’s written policy and attestation instead of treating a marketing statement or a database name as proof of equivalent protection.

Implementation checklist for laboratories

  1. Identify the governing requirement. Determine whether the purchase is charged to a covered U.S. federal award, contract, or agreement, and read its current procurement terms.
  2. Confirm the supplier’s status. Obtain an attestation or other documentation showing adherence to the framework version required by the award.
  3. Provide accurate customer information. Use the legal institutional name, verifiable contact details, shipping location, and a clear description of the intended research.
  4. Prepare for a legitimate flag. Designate a biosafety, compliance, or principal-investigator contact who can answer questions without sending sensitive material through an insecure channel.
  5. Keep procurement evidence. Store the attestation, order records, correspondence about any review, and the date on which the framework status was checked.
  6. Recheck policy status. Before a new project, renewal, or unusual order, review current ASPR, OSTP, NIH, and award-specific notices because the 2024 framework may be replaced.

What screening cannot prevent

  • Undetected sequences: A novel design, fragmented order, or sequence outside a database may not produce a match.
  • False positives and delays: Benign research can resemble a SOC and require extra documentation or manual review.
  • Non-synthesis routes: Screening does not cover every way genetic material can be obtained, copied, or transferred.
  • Insider and operational risks: A legitimate customer can misuse material after delivery, and poor laboratory security can undermine a careful procurement check.
  • Jurisdictional gaps: Federal procurement conditions do not automatically impose the same controls on private, state, or overseas transactions.

For these reasons, screening works best as one layer in a broader biosafety and biosecurity program. Institutions still need responsible-research review, access controls, secure data handling, inventory management, incident response, and training.

Distinguishing guidance, procurement rules, and law

HHS guidance explains why sequence and customer screening are recommended. The OSTP framework specifies actions for participating providers and equipment manufacturers. NIH notices translate those actions into conditions for covered NIH-funded procurement. An executive order directs executive-branch policy work but does not itself supply every operational detail a laboratory needs.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Those categories matter when a supplier or researcher asks, “Is screening legally required?” The accurate answer depends on the transaction: the funding source and award terms, the agency involved, the equipment or material being purchased, institutional policy, and any applicable state or national law. A laboratory should not assume that a federal framework automatically governs every order, nor that an order outside federal funding has no compliance obligations.

Why the approach remains useful despite its limits

Order-level screening creates a practical interruption point where sequence information and customer information can be assessed together. It can expose implausible identities, unexplained purchasing patterns, or sequences that merit expert review before synthesis. The framework’s requirements for escalation, records, and information security also make decisions more consistent and auditable than an informal, ad hoc check.

Its value is therefore risk reduction, not certainty. The strongest program is one that uses up-to-date sequence references, qualified reviewers, documented customer checks, secure escalation, and continual policy review as the U.S. framework evolves.
