Gene editing and cloning are different biological routes in de-extinction research. Gene editing changes selected DNA sequences in living cells from an extant relative, using reconstructed information from an extinct species. Somatic cell nuclear transfer (SCNT), the mammalian cloning method discussed in conservation guidance, moves a donor cell’s nucleus into an egg whose own nucleus has been removed, then develops the embryo with a suitable surrogate.

The distinction matters because the two methods require different starting material and produce different biological outcomes. SCNT copies the nuclear genome of the donor cell if the transfer succeeds; editing usually creates a living relative with selected extinct-species-informed changes. Neither route, by itself, proves that an extinct species has been restored in its genetic, epigenetic, behavioral and ecological entirety.

How is gene editing different from cloning in de-extinction research?

In practical terms, cloning needs a usable, intact donor-cell nucleus. Ancient-DNA editing can begin with sequence fragments and cells from a living relative, because cells from the extinct animal may no longer be viable. SCNT therefore attempts to copy a particular cell donor, while genome engineering reconstructs selected sequences or traits on the biological scaffold of a living species.

That difference changes the claim a project can honestly make. A successful SCNT birth would be a clone of the donor cell, not automatically a recovered species or a functioning population. A gene-edited animal would normally be a proxy: an organism intended to resemble or perform part of an extinct species’ role without being an intact copy of its genome, development, behavior or ecology.

The two methods at a glance

Axis SCNT cloning Genome editing for a de-extinction proxy
Required starting material A donor somatic-cell nucleus that can be used in an egg Reconstructed sequence information and editable cells from a living relative
Main intervention Transfer the donor nucleus into an enucleated egg and develop an embryo Alter selected DNA sequences in a relative’s cells, using that relative as the genomic scaffold
Relationship to the extinct genome Copies the donor cell’s nuclear genome if successful; extinct-species cloning would require preserved extinct cells Combines selected extinct-species-informed changes with much of a living relative’s genome
Main constraint Intact usable cells plus compatible egg and surrogate biology Gaps in ancient DNA, the choice of edits, their biological effects and reliance on a relative
How to describe the outcome A clone of the cell donor, not automatically a restored population or ecosystem An engineered proxy with selected traits, not an intact extinct genome
Conservation question Can viable individuals form a population that advances a defined conservation goal? Do the selected traits and ecological role justify the intervention with manageable risks?

The comparison follows the International Union for Conservation of Nature (IUCN) Species Survival Commission’s 2016 guiding principles. Individual projects can use different laboratory workflows, so the table describes the biological logic rather than a mandatory protocol.

What gene editing means in a de-extinction proposal

Gene editing, also called genome engineering, makes deliberate changes to DNA in living cells. For a de-extinction proposal, researchers compare DNA recovered from the extinct species with the genome of a living relative. They identify differences that might contribute to a candidate trait and edit some of those sites in cells from the relative.

The living relative supplies the framework

Because ancient DNA is usually fragmented and incomplete, the relative’s genome supplies sequence that cannot be reconstructed confidently. The edited cells therefore retain most of the relative’s genetic background. Even if several extinct-species-associated variants are introduced, the result is genetically mixed rather than a complete extinct genome.

Traits are targets, not a complete identity

A project may target a limited set of characteristics associated with the extinct animal. Changing those sites does not recreate every gene, regulatory sequence, developmental interaction or environmental adaptation. The IUCN guidance describes this approach as creating an organism with selected ancestral traits and cautions that the hybrid’s trait expression can be unpredictable.

What somatic cell nuclear transfer means

SCNT is the mammalian cloning method addressed in the IUCN guidance. A somatic cell is a body cell rather than an egg or sperm cell. Its nucleus is removed and placed into an egg cell whose own nucleus has been removed. The reconstructed cell is induced to begin embryonic development and must then be carried by a suitable surrogate.

Why the donor nucleus is decisive

The nuclear genome copied by SCNT comes from the donor cell. In an extinct-species scenario, that means researchers would need a preserved extinct-animal cell whose nucleus remains usable. DNA fragments extracted from bone, hair or other ancient material are not equivalent to a living cell and cannot simply be placed into an egg as a ready-made SCNT donor.

Cloning does not equal species recovery

Even a viable SCNT birth would copy one donor cell. It would not, by itself, provide the genetic diversity needed for a self-sustaining population, restore lost social learning or recreate the ecological conditions in which the species evolved. Population viability and conservation purpose would still have to be demonstrated.

How the workflows differ

Conceptual genome-editing workflow

  1. Recover and interpret extinct DNA. Researchers work with whatever sequence fragments remain and distinguish reliable information from gaps and uncertainty.
  2. Compare genomes. The extinct sequence is compared with that of a living relative to identify candidate differences.
  3. Select edits. Only some sites are chosen, generally those linked to a proposed trait or function. The selection is an interpretation, not a complete reconstruction of the extinct genome.
  4. Edit relative cells. The chosen DNA changes are introduced into cells from the living relative. Missing extinct sequence is supplied by the relative’s existing genome.
  5. Assess the proposed proxy. Any resulting organism must be evaluated for its traits, health, behavior, reproduction and ecological consequences rather than judged by a small edit list alone.

Conceptual SCNT workflow

  1. Obtain a usable donor cell. The donor must contain a nucleus that can support the transfer. For extinct-species cloning, it would need to come from the extinct species itself.
  2. Enucleate an egg. The egg’s own nucleus is removed, leaving cellular machinery that can support early development.
  3. Insert the donor nucleus. The somatic-cell nucleus is placed into the enucleated egg.
  4. Initiate development. The reconstructed cell is induced to begin dividing as an embryo.
  5. Use a compatible surrogate. The developing embryo requires a surrogate capable of carrying the pregnancy. Compatibility and welfare are part of the biological and ethical assessment.

These workflows are not interchangeable. Editing cannot turn ancient sequence fragments into a donor nucleus, and SCNT cannot infer missing extinct DNA by copying a living relative.

Can you clone an extinct animal from ancient DNA?

Usually, ancient DNA alone is not enough for SCNT. SCNT requires a usable cell nucleus, whereas ancient material generally provides degraded fragments rather than intact, living cells. If a preserved extinct cell with a viable nucleus existed, cloning could be considered in principle, but the egg, embryo and surrogate biology would still have to work.

When only fragments remain, researchers face gaps in the extinct sequence. A living relative can fill those gaps as a scaffold for editing, but doing so changes the nature of the result. The engineered animal carries a chosen set of reconstructed differences on a relative’s genome; it is not a nuclear-genome copy of the extinct species.

Why an edited proxy is not a faithful resurrection

Species identity is not determined by a short list of visible features. DNA regulation, embryonic development, physiology, immune function, social behavior and interactions with other organisms all contribute. Ancient-DNA reconstruction has gaps, and the effects of combining selected extinct variants with a relative’s genome may not be predictable.

Epigenetic effects add another layer. Epigenetics concerns regulation of genes without changing the underlying DNA sequence. The IUCN SSC guidance notes that hybrid trait expression can be unpredictable, including because of epigenetic effects. An animal that resembles an extinct species in a few measured traits could therefore differ in ways that matter for health, behavior or its ecological role.

Its 2016 principles warn that the word “de-extinction” implies a resurrection in genetic, behavioral and physiological entirety. The guidance favors the term proxy where appropriate: a living organism designed to resemble or perform some role of an extinct species without claiming complete restoration.

What the 2025 dire-wolf announcement illustrates

The IUCN SSC Canid Specialist Group’s statement of 18 April 2025 described the dire-wolf announcement as involving isolation of endothelial progenitor cells from a grey wolf followed by gene editing. The group acknowledged the technical achievement but questioned describing the resulting animals as recreated dire wolves and asked what conservation purpose the animals would serve.

This account comes from the specialist group’s response to the announcement; it is not an independent full methods paper documenting every experimental step. The careful description is therefore: the announcement concerned gene-edited grey-wolf cells, according to the IUCN SSC Canid Specialist Group, while the group disputed calling the resulting animals recreated dire wolves.

The example shows why “gene-edited relative with selected traits” and “cloned extinct animal” should not be used as synonyms. The former starts with a living species and changes chosen DNA sites. The latter, in the SCNT sense, would require a donor nucleus from the extinct species and would copy that cell’s nuclear genome.

When could a proxy have conservation value?

Technical success is only one test. The IUCN SSC’s 2016 principles treat proxy creation as a possible conservation tool, not as an objective justified by novelty. A proposal would need to address the original causes of extinction, the availability and suitability of habitat, population viability and the effects on other species.

Questions a responsible assessment must answer

  • What problem is being solved? The project should identify a defined conservation outcome rather than assume that producing an animal is valuable on its own.
  • Is habitat available? Releasing a proxy requires suitable habitat and a plan for long-term management.
  • Can a population persist? A few engineered individuals do not establish a viable population. Reproduction, genetic diversity, health and social behavior must be considered.
  • What could go wrong? Researchers must assess ecological effects, disease, competition, hybridization, welfare and other unwanted impacts.
  • Are the original threats addressed? If hunting, habitat loss, invasive species or another pressure caused the extinction, releasing a proxy without changing that pressure may fail.
  • Who participates? Decisions should include relevant knowledge-holders and rights-holders, along with social, cultural and ethical considerations.

The IUCN policy page on synthetic biology, adopted by IUCN Members at the 2025 World Conservation Congress, calls for science-based, case-by-case decisions, explicit attention to uncertainty and a precautionary approach. It also states that synthetic biology is not a substitute for continued efforts to address biodiversity loss.

How to read claims about de-extinction accurately

Readers can separate a strong technical claim from an inflated species claim by checking the starting material, intervention and stated outcome.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.
  • Ask what cells were used. A preserved extinct-species nucleus points toward SCNT; cells from a living relative point toward genome editing.
  • Ask how much of the genome was changed. A list of selected edits does not establish recovery of the extinct genome.
  • Separate appearance from identity. A visible or measurable trait is not proof of matching physiology, behavior or ecology.
  • Look for uncertainty. Ancient-DNA gaps, untested gene interactions and epigenetic effects should be acknowledged.
  • Look for a conservation plan. Habitat, release conditions, population viability and risk management matter as much as laboratory results.
  • Check the evidence level. An institutional statement about an announcement is different from an independently reviewed methods paper.
