Researchers have produced fertile female mice from male-derived cells after the cells lost their Y chromosome. The result suggests a possible way to reproduce when genetic material from only one sex remains—but it has not been tested in endangered wildlife, and the chromosome loss is not a reliable method for choosing a clone’s sex.
What the mouse experiments found
In a 2009 experiment, researchers used immature Sertoli cells taken from neonatal male mice in somatic cell nuclear transfer, a cloning technique that places a donor cell’s nuclear material into an egg cell. One of 27 clones was female. She grew to adulthood, had a 39,X karyotype, and produced offspring after natural mating with a littermate. The authors concluded that the Y chromosome had been deleted either in the donor cell or early in nuclear transfer. The study appeared in the Journal of Reproduction and Development in 2009.
That female was an accidental outcome, not the product of a controlled sex-selection procedure. The result showed that female offspring could arise from male-derived material under the conditions of that experiment; it did not show that researchers could reliably make female clones on demand.
How the later freeze-dried-cell study differed
A 2022 study used a more involved process beginning with freeze-dried somatic cells. Researchers established embryonic stem-cell lines through nuclear transfer, then used those cells in further nuclear transfer. The study reported 75 cloned mice overall. In one male-derived cell line, the Y chromosome was lost; clones produced from that line were female and fertile. The researchers could not determine exactly when the chromosome loss occurred. The findings were published in Nature Communications in 2022.

| Research approach | Donor material and process | Reported outcome | What the result establishes |
|---|---|---|---|
| 2009 mouse experiment | Immature Sertoli cells from neonatal male mice; somatic cell nuclear transfer | One female among 27 clones; the adult female had a 39,X karyotype and produced offspring by natural mating | A female clone arose after Y-chromosome loss, but the event was accidental |
| 2022 mouse study | Freeze-dried somatic cells; nuclear-transfer embryonic stem-cell lines used in further nuclear transfer | 75 cloned mice overall; clones from a male-derived cell line that lost its Y chromosome were female and fertile | A later process also produced fertile female mice from male-derived material; the timing of chromosome loss was unclear |
The headline numbers describe different things: the 2009 figure is one female among 27 clones in that experiment, while the 2022 paper’s 0.02% figure is its overall success rate for producing cloned mice from freeze-dried somatic cells through the reported process. They are not directly comparable rates.
Why the findings are not yet a conservation method
The conservation idea is most relevant to a hypothetical situation in which genetic material from only one sex remains. The authors of the 2009 paper said the finding “suggests the possibility of resuming sexual reproduction after a single male is cloned,” and proposed that this could be useful for reviving extinct or endangered species. That was a proposed implication of a mouse result, not a demonstration in wildlife.

Neither study establishes that the method works in an endangered species, is safe for conservation use, or can produce enough healthy, genetically useful animals to support a population. The experiments show laboratory outcomes in mice; they do not demonstrate that cloning can restore an endangered population.

The 2022 study reported an overall 0.02% success rate for producing cloned mice from freeze-dried somatic cells through its process. It also described DNA damage and chromosome abnormalities, and said the cells required storage at −30°C. Those constraints complicate any practical application beyond the laboratory. The reported Y-chromosome loss itself was not controlled: in the 2009 experiment it was inferred after an accidental female outcome, and in the 2022 study researchers could not pinpoint when it occurred.

- Sex outcome: Y-chromosome loss was associated with female clones, but the studies do not provide a dependable way to select the outcome.
- Efficiency: The 2022 process-wide success rate was low, making it a substantial technical hurdle.
- Cell quality and storage: DNA damage and chromosome abnormalities were reported, and freeze-dried cells required storage at −30°C in the 2022 study.
- Scope: Fertility was reported in mice; the evidence does not establish effectiveness or population-level value in endangered wildlife.
What the research means
These studies establish that fertile female mice can result from male-derived material after Y-chromosome loss, through two different cloning approaches. They make a narrow conservation possibility scientifically discussable when material from only one sex is available. They do not establish a reliable sex-reversal technique or a proven intervention for endangered species.
