Electron paramagnetic resonance (EPR), also called electron spin resonance (ESR), can reveal what happens inside a porous crystal when a paramagnetic center—one with an unpaired electron—responds to a molecule entering or reacting in a pore. The signal may come from a metal ion in the framework, an adsorbed paramagnetic species, or a deliberately introduced spin probe. EPR is therefore a selective way to study local environments and host–guest interactions, not a universal detector for every molecule in a pore.
What EPR measures in a porous crystal
EPR detects paramagnetic species: atoms, ions, molecules or other centers that have one or more unpaired electron spins. Its signal is associated with the spin-active center, so the first question in an experiment is not simply “Which gas is in the pore?” but “Which center produces the signal, and how could the guest affect it?”
In a porous crystal, the signal-bearing center can be a transition-metal ion that is already part of the framework, a paramagnetic molecule adsorbed from the surrounding environment, or an organic radical or metal ion added as a probe. A probe can occupy a framework position or reside in a pore. When a guest adsorbs nearby, it may alter the probe’s local surroundings; EPR can then register changes associated with the local spin environment.

How adsorption becomes an EPR signal
Start with a spin-active center
A measurement needs a paramagnetic center that can generate an EPR signal. Some porous frameworks contain such centers naturally. If the framework and target guest are diamagnetic—that is, they do not supply an unpaired electron—there is no direct electron-spin signal from those components just because they are present together. An experiment may instead introduce a paramagnetic probe to make a selected pore environment observable.
Let the guest change the local environment
As gas molecules enter pores or leave them, a spin center in or near an adsorption site can experience a changed local environment. The EPR spectrum or signal can respond to that change. In this sense, the probe acts like a reporter: the measurement tracks what happens around the probe, rather than observing all molecules indiscriminately.

The 2025 review by Muhammad Fernadi Lukman and Andreas Pöppl focuses on dihydrogen-isotopologue adsorption in porous materials. It describes using in situ EPR during adsorption and desorption, and explains how pulsed approaches can provide higher-resolution information about host–guest interactions. Those applications illustrate the value of looking at the probe’s response as conditions change, rather than treating a single spectrum as a complete inventory of the pore contents.
Interpret the response at the right scale
A changed signal can support an inference about the probe’s local environment or a process taking place near it. It does not, by itself, establish that every pore site is occupied, quantify total loading, or uniquely identify an arbitrary adsorbed chemical. Those stronger conclusions require evidence that the particular signal and experimental design support them.

What the measurement can reveal
Local structure around a paramagnetic framework center
If an EPR-active metal center is part of the framework, its signal can be used to characterize its local structure and environment. This can help researchers investigate how the framework responds to its surroundings. Interpretation depends on which paramagnetic center is being measured: a signal from one metal site is not automatically representative of every chemically distinct site in the crystal.
Adsorption sites and host–guest interactions
A spin probe placed in a pore or framework can report changes in its surroundings as guests adsorb. In the 2025 review, CW and pulsed EPR are discussed as approaches for following adsorption and desorption and studying interactions between the host material and its guest. The useful inference is tied to the probe’s position and response; it should not be generalized beyond what the experiment establishes.

Gas uptake in a specific porous organic framework
A 2023 Nature Communications study demonstrated an embedded molecular spin sensor, Sc3C2@C80, in Py-COF. The study used EPR signal intensity and linewidth to probe adsorption of N2, CO, CH4, CO2, C3H6 and C3H8. It is an example of a sensor designed for a particular material and measurement, not evidence that any porous crystal can distinguish all gases by the same method.

EPR can also be relevant when paramagnetic species take part in chemistry inside a porous material. A 2023 review by Lena Marie Funke and co-authors cites EPR observation of NO2 dimerization in MFM-502 and work on paramagnetic atomic sites for catalysis. These examples show why EPR can contribute to studies of reaction species and active sites, while also underscoring the need to connect a signal to the specific center and reaction under investigation.

Choosing between continuous-wave and pulsed EPR
The method depends on the experimental question, the spin-bearing center and the guest or process of interest. The 2025 Lukman and Pöppl review describes both continuous-wave (CW) and pulsed EPR in porous-material adsorption studies.
| Approach | What it is used for in the reviewed adsorption work | What to keep in mind |
|---|---|---|
| Continuous-wave (CW) EPR | In situ monitoring of adsorption and desorption. | Best aligned with following signal changes as uptake or release proceeds; the signal still depends on the paramagnetic center being monitored. |
| Pulsed EPR | Higher-resolution investigation of host–guest interactions. | Useful when the aim is to resolve interaction details, rather than only track adsorption or desorption behavior. |
This distinction is not a rule that one technique can answer only one kind of question. It is a practical starting point: monitoring a process and resolving interaction details are different objectives, and the experiment should be designed around the one that matters.
How to plan and interpret an EPR adsorption experiment
- Identify the signal source. Establish whether the EPR-active center is already present in the framework, comes from an adsorbed paramagnetic species, or is an added metal-ion or organic-radical probe. If a probe is added, establish whether it is in a framework position or a pore.
- Define the claim you want to test. Decide whether the goal is to monitor adsorption or desorption, examine a local framework environment, investigate a host–guest interaction, or observe a reactive paramagnetic species. These are related but distinct questions.
- Choose a measurement approach that fits the goal. The reviewed work uses in situ CW EPR for monitoring adsorption and desorption, while pulsed EPR can provide higher-resolution insight into host–guest interactions.
- Connect the observable to the center. If signal intensity or linewidth changes, interpret that response in relation to the specific sensor and its local environment. The Sc3C2@C80 study is an example of those observables being used for a defined sensor in Py-COF; it does not make them universal measures of gas identity or total loading.
- Support broader structural or material claims with complementary evidence. Treat EPR as one part of porous-material characterization. Magnetic-resonance research on metal–organic frameworks combines it with other characterization approaches to address structure and function.
Limits that matter when reading an EPR result
- Only paramagnetic species provide the electron-spin signal. Diamagnetic framework components and guests do not automatically become visible to EPR. A spin probe can report on a nearby environment, but the conclusion concerns that probe and its local surroundings.
- The response is center-specific. A framework ion, an adsorbed radical and an introduced probe are not interchangeable reporters. Their locations and chemical environments determine what a measured change can support.
- A signal change is not automatically a chemical identification. The cited adsorption-sensor demonstration examined six named gases in one sensor–framework system. It does not establish universal identification of arbitrary guests.
- EPR alone does not necessarily measure total pore loading. A local spin response should not be presented as a direct measure of all molecules in the material unless the particular experiment validates that interpretation.
- No universal sensitivity or detection limit follows from these examples. The cited studies establish applications, not one performance figure that applies across different crystals, spin centers and gases.
How EPR fits into broader MOF characterization
EPR is one member of a wider magnetic-resonance and materials-characterization toolkit. In a 2023 review, Funke and co-authors describe magnetic resonance as useful for questions including monitoring crystal growth to investigate synthesis mechanisms, measuring adsorption isotherms with spectroscopically resolved site information, and contributing to the discovery of MOF glasses. Their description illustrates the breadth of magnetic-resonance methods; those capabilities should not all be attributed to EPR alone.
The same review reports that its Scopus and Web of Science searches, using stated filters, identified 124 filtered NMR/EPR studies of MOFs from 2017 through 2022, including 26 EPR studies. It separately reports 450 MOF solid-state NMR studies from January 2017 through July 2022. These are counts from the review’s search scope and filters, not totals for all published work or current cumulative counts.
For a reader evaluating a claim about a porous crystal, the key distinction is between observing a spin-bearing center and making a wider statement about a material. EPR can provide useful local and process-sensitive evidence, but adsorption capacity, framework structure or molecular identity may call for complementary measurements suited to those questions.
