In 2012, Martin Langecker and colleagues showed that designed DNA can be folded into a membrane-spanning pore. Their DNA-origami structure inserted into synthetic lipid membranes, conducted ions, opened and closed in response to molecular conditions, and distinguished individual DNA molecules in translocation experiments. It was a laboratory demonstration in lipid membranes—not a device tested in living cells or an established medical technology.

What the 2012 experiment actually demonstrated

The work appeared in Science on November 16, 2012, as “Synthetic lipid membrane channels formed by designed DNA nanostructures” by Martin Langecker and colleagues (volume 338, issue 6109, pages 932–936; DOI 10.1126/science.1225624). The central result was architectural: researchers designed DNA strands that self-assembled into a rigid nanoscale object capable of spanning a lipid bilayer.

That distinction matters. The experiment did not show that DNA channels could be put into a person, operate in an animal, or treat disease. Measurements were made in synthetic membrane systems, including lipid vesicles and other controlled bilayers. The Technical University of Munich (TUM) account explicitly states that the construct had not been tested with living cells.

How DNA origami becomes a membrane pore

Folding strands into a defined shape

DNA origami uses designed DNA sequences as construction material. Instead of relying on the molecule only as genetic information, researchers program complementary regions so multiple strands bind in a planned pattern. The resulting bundle can be shaped into a three-dimensional object with a cavity through its center.

For this project, the target shape was inspired by alpha-hemolysin, a natural membrane protein that forms a stem and a barrel-like cap. The synthetic version used a long DNA stem joined to a wider cap. The stem was intended to pass through the hydrophobic interior of a lipid membrane, while the cap remained at the membrane surface.

Anchoring the structure in lipid

DNA is not naturally comfortable inside the oily core of a membrane. To make insertion possible, the team attached cholesterol groups to the DNA framework. Cholesterol has an affinity for lipid bilayers, so these chemical anchors act as membrane handles. They help orient the construct and hold it in place once the stem has entered the membrane.

Design feature Reported value or description Why it matters
Construction method DNA origami made from designed DNA strands Allows a nanoscale pore to be specified by sequence and geometry
Overall inspiration Alpha-hemolysin’s stem-and-barrel membrane architecture Provides a model for spanning a bilayer while leaving an opening
Stem length 42 nanometres, reported by TUM Provides the membrane-spanning section and surface support
Inner diameter 2 nanometres, reported by TUM Defines the approximate passage through the channel
Membrane anchors 26 cholesterol groups, reported by Chemistry World Promote attachment and the intended orientation in lipid

Why orientation is important

A pore that merely sticks to a membrane is not equivalent to a channel that crosses it. The TUM description says experiments with lipid vesicles showed the device binding to a bilayer in the correct orientation: the stem penetrated the membrane while the cap held at the surface. That arrangement creates a continuous path from one side of the bilayer to the other.

What the electrical measurements showed

Researchers used single-channel electrophysiology to monitor ionic current through individual synthetic pores. The TUM research record reports conductances on the order of 1 nanosiemens. Conductance is a measure of how readily charged particles carry electrical current through the opening; observing a discrete channel-level signal is evidence that the DNA object was functioning as an ion-conducting pore in the test membrane.

The measurements also showed gating. In ion-channel terminology, gating means that a pore switches between conducting and less-conducting states rather than remaining a permanently open hole. The recorded changes in current indicated that the DNA structure could adopt different functional states under the experimental conditions.

Observed behavior What was measured Proper interpretation
Ion conduction Single-channel conductance on the order of 1 nanosiemens The construct formed an electrically active pore in a synthetic membrane
Gating Switching between conducting states The pore’s opening was responsive to structural or environmental conditions
Molecular translocation Experiments discriminated single DNA molecules The pore could be used as a single-molecule sensing platform in the test system

How the channel distinguished single DNA molecules

In a translocation experiment, a molecule moving through a nanopore changes the ionic current. The size, duration, and shape of that disturbance can carry information about the molecule. The TUM research record reports that the synthetic channels discriminated individual DNA molecules. This is a single-molecule result in an experimental membrane setup, not a demonstration of sequencing accuracy, clinical diagnostics, or operation inside a cell.

The important proof-of-concept is that a DNA framework could do more than provide a static hole. It created a controlled nanoscale passage through which researchers could observe individual molecular events electrically.

What the mutant channels revealed about gating

The contemporaneous account in Chemistry World described variants in which additional DNA strands protruded from the channel core. Those protrusions produced stronger gating than the corresponding unmodified design, and the reported gating times varied with protrusion length.

That result supports a structural explanation for the electrical behavior: details inside the pore can influence how readily molecules or ions pass and how long the channel remains in a given state. It does not mean every channel behaved identically, nor does it establish a universal gating law for all DNA-origami pores. The variants were an experimental way to connect a deliberate geometric change with a measurable change in channel behavior.

What was tested—and what was not

Demonstrated in the laboratory

  • Designed DNA strands assembled into a channel-like nanoscale structure.
  • Cholesterol modifications enabled binding to synthetic lipid bilayers.
  • The construct adopted an orientation in lipid vesicles in which its stem penetrated the membrane and its cap remained at the surface.
  • Single-channel electrical measurements showed ion-conducting behavior and gating.
  • Single-molecule experiments showed discrimination of individual DNA translocation events.

Not established by this paper

  • Insertion, stability, or safety in living cells.
  • Operation in an animal or human body.
  • Medical treatment, drug delivery, or clinical diagnosis.
  • Performance against natural channels in a head-to-head biological test.
  • A commercial product, consumer kit, or validated sequencing service based on this particular construct.

Professor Friedrich Simmel of TUM summarized the boundary clearly: “We have not tested this yet with living cells, but experiments with lipid vesicles show that our synthetic device will bind to a bilayer lipid membrane in the right orientation, so that the stem both penetrates the membrane and holds at the surface, forming a pore.”

How this synthetic channel compares with other pores

The 2012 report does not provide a product comparison or a head-to-head performance study. A useful comparison therefore has to stay at the level of design and evidence rather than imply rankings that were never measured.

The practical advantage suggested by the DNA approach is programmability: changing the sequence or adding strands can alter geometry. The practical limitation is that programmability does not automatically provide the robustness, selectivity, lifetime, or biological compatibility required for use outside a controlled membrane experiment.

Why the result mattered for nanopore research

Nanopore sensing depends on measuring how individual molecules alter current as they approach or pass through a nanoscale opening. Conventional biological pores are made from proteins; a DNA-origami pore offers a different construction strategy in which dimensions and internal features can be designed directly.

The channel’s demonstrated single-molecule discrimination made it relevant to sensing research. A future design might place recognition elements inside a pore, tune the opening for a target molecule, or alter the electrical signal through controlled structural changes. Those are possible directions, not results established by the 2012 experiment.

The same distinction applies to engineered transport. An artificial channel could, in principle, be designed to regulate movement across a membrane. Before that becomes a biological technology, researchers would need to establish reliable insertion into cell membranes, operation in the presence of cellular proteins and ions, resistance to degradation, control over unintended leakage, and compatibility with living systems. None of those requirements was resolved by the reported lipid-vesicle work.

How to read the dimensions and conductance

The reported dimensions describe one research construct, not a universal specification for DNA channels. The 42-nanometre stem and 2-nanometre opening were design and measurement details for this architecture. Likewise, conductance on the order of 1 nanosiemens came from the study’s single-channel electrophysiology conditions. Conductance can depend on membrane composition, salt concentration, voltage, temperature, channel orientation, and other experimental factors, so the value should not be treated as a guaranteed rating for every assembly.

The 26 cholesterol groups are also a feature of the reported design rather than a generally required number for all membrane-inserting DNA origami. Different architectures could use different anchor chemistries, counts, or locations.

Common misunderstandings about “DNA folds up into a channel”

It is not a DNA strand acting like a natural protein

The phrase describes DNA as structural material. The strands were designed to assemble into a three-dimensional object; they were not used as a genetic instruction inside a cell.

“Synthetic” does not mean artificial cells were created

The membrane was a laboratory lipid bilayer or vesicle. The experiment recreated a physical membrane environment for measurement, not a complete cell with metabolism, organelles, and regulatory systems.

An ion channel is not automatically a medical device

Ion conduction and gating establish a physical function under test conditions. Medical use would require separate evidence for delivery, targeting, toxicity, immune response, durability, and therapeutic benefit.

What a researcher would need to reproduce the concept

At a high level, reproducing this class of experiment requires designed DNA-origami strands, cholesterol-modified components, a lipid-bilayer or vesicle preparation, and single-channel electrophysiology. The work also depends on confirming assembly quality, membrane orientation, and electrical signals at the single-pore level. The source material identifies these as specialized research categories, not as an off-the-shelf consumer product or a standardized home experiment.

Because the published result concerns a particular construct, changing the strand design, anchor placement, membrane composition, or measurement conditions could change insertion and gating. A reproduction should therefore treat the reported architecture and conditions as an experimental starting point, not as a plug-and-play specification.

The bottom line for readers

The 2012 DNA-origami channel was a significant proof of concept: researchers designed a DNA nanostructure that anchored in a synthetic lipid membrane, conducted ions, gated, and distinguished single DNA molecules. Its importance lies in showing that a programmable nucleic-acid framework can perform a membrane-channel function normally associated with proteins.

Its limits are equally important. The evidence came from synthetic membranes and lipid vesicles, and TUM explicitly reported no living-cell testing. Nanopore sensing and engineered transport are reasonable future possibilities, but they remain prospective rather than demonstrated applications of this experiment.
