A DNA origami flask is a hollow container assembled from DNA strands and engineered to look like a tiny, rounded-bottom flask. It is measured in nanometers, not millimeters, and it is a research nanocompartment rather than miniature glassware. A long single-stranded DNA scaffold is folded by many short staple strands; carefully positioned crossovers and nick points bend the resulting DNA helices into a controlled three-dimensional shape.
The flask-like design belongs to a broader class of DNA origami nanocompartments. A 2023 review describes the Yan group’s 2011 work as a method for modeling arbitrary three-dimensional shapes while controlling surface curvature. Images of flask-like and other curved containers were confirmed with atomic force microscopy and transmission electron microscopy.
What “DNA origami flask” actually means
The word flask describes the outline of the nanostructure, not its material, scale or everyday function. This object is made from folded DNA, with a hollow interior enclosed by an arrangement of DNA helices. Its dimensions are in the nanometer range, so it cannot be used to pour, store or measure macroscopic liquids.
DNA origami supplies the construction method. Instead of asking DNA to form a shape on its own, researchers design a target geometry and use complementary strands to fold a long scaffold into that geometry. The finished object can include flat panels, bends, cavities and rounded surfaces. In the flask example, the design creates a curved container whose profile resembles a laboratory flask when viewed at the nanoscale.

The underlying idea was reported by the Yan group in 2011. A later review by Jing Huang, Sabrina Gambietz and Barbara Saccà summarizes that work this way: “In 2011, the Yan group presented an innovative strategy to model arbitrary 3D shapes with control over the extent of surface curvature.” The statement is important because the achievement was not simply making a smaller version of a familiar bottle. It was demonstrating programmable curvature in a DNA-based structure.
A flask-shaped outline also does not prove that the object has a practical neck, stopper or pourable opening. Unless a particular design is reported with those features and a demonstrated loading method, “flask” should be read as a geometric description.
How DNA strands fold into a container
Scaffolded DNA origami uses two classes of strands. The scaffold is one long, single-stranded DNA molecule. Shorter strands called staples bind to selected sections of that scaffold. Each staple is designed to connect particular scaffold segments, holding neighboring DNA sections together and forcing the long strand to follow the planned route.
Do these 3 things before closing this tab:
1Fix the driver behind crashes, sound loss and screen glitches2Clear out junk files and repair common Windows errors3Scan for outdated or missing drivers - takes under a minute1. Define the target shape
Researchers first specify a three-dimensional container rather than starting with a random DNA mixture. The design identifies the walls, the hollow region and the degree of curvature required for a rounded-bottom profile. The scaffold length and the arrangement of DNA helices constrain what volume and shape are achievable.
2. Route the scaffold through the structure
The long scaffold is assigned a path through the planned helices. This routing determines which sections will sit next to one another and where short staples must bind. A larger or more elaborate container generally requires a different scaffold layout from a compact, simple compartment.

3. Add the staple strands
Each staple is a short, predesigned DNA strand with sequences complementary to selected scaffold regions. When the strands hybridize, the staples pull those regions into the intended arrangement. Collectively, thousands of these local connections can fold one long molecule into a defined three-dimensional object.
4. Place crossovers to create curvature
Crossovers connect neighboring DNA helices at selected positions. Moving a crossover changes how the helices track around one another and therefore changes the surface direction. A pattern that works for a flat panel will not automatically produce a smooth bend. The Yan group’s strategy used deliberate crossover placement to control how much a surface curved.
5. Use nick points to balance flexibility and stability
Nick points are breaks in one strand of a DNA duplex, while the overall assembly remains connected through the rest of the design. Their placement can let selected portions flex without abandoning the complete structure. The 2011 approach combined crossover placement and nick-point placement to obtain both controlled curvature and structural stability.
6. Confirm the assembled geometry
Once the designed strands have formed the intended object, imaging is needed to determine whether the particles actually resemble the model. The reviewed flask-like structures were examined with atomic force microscopy and transmission electron microscopy. Those methods provide visual evidence of the curved, hollow architecture; they do not turn the object into a macroscopic flask.
How small is the container?
The most defensible volume figure applies to the class of nanocompartments discussed in the 2023 review, not to this flask alone. Across the surveyed DNA origami containers, reported volumes are approximately 10 to 150 zeptoliters, with the value depending on scaffold size and geometry.

| Measurement or feature | What is established | How to interpret it |
|---|---|---|
| Volume of surveyed DNA origami nanocompartments | Approximately 10–150 zeptoliters, according to the 2023 review | This is a class-wide range, not a measured capacity for the specific nanoflask |
| Capacity of the rounded-bottom nanoflask | Not stated in the cited sources | Do not assign 10 or 150 zeptoliters to the flask without a flask-specific measurement |
| Physical scale | Nanometer-scale DNA structure | The object is a molecular container, not laboratory glassware |
| Shape | Flask-like, curved and hollow | The term describes its designed geometry |
| Structural confirmation | Atomic force microscopy and transmission electron microscopy images | Imaging verifies nanoscale form rather than consumer usability |
Using the range correctly matters. A zeptoliter value is easy to repeat as though it were a specification printed on a product label, but the cited evidence does not provide that specification for this particular flask. Scaffold length, helix arrangement and curvature all affect the enclosed space, so two DNA origami containers can look similar while holding different amounts.

What gives the nanoflask its rounded bottom?
DNA double helices are comparatively rigid over short distances, so a smooth bowl or rounded base cannot be obtained merely by drawing a curve around a collection of straight strands. The design has to distribute small directional changes through many connected helices.
Crossovers provide the main geometric control. By deciding where one helix connects to the next, a designer changes the relative orientation of neighboring sections. Repeating those changes across the lower portion of the object produces a controlled bend instead of a single sharp corner. Nick points then allow selected strand segments to accommodate the bend while preserving enough connections to keep the container intact.
This is why the flask is best understood as a programmed assembly. Nothing is blown, molded or carved like glass. The final outline emerges because the base sequence of every staple, its binding partners and its connection points were selected in advance.
How this design compares with other DNA nanocompartments
The flask is one geometry within a wider family of DNA-based containers. The available sources describe differences among designs but do not establish a performance ranking in which the flask is universally better or worse. Useful comparison criteria are therefore design and intended use rather than consumer-style product scores.

What researchers can use DNA nanocompartments for
DNA origami containers are valuable because a hollow, addressable structure can place molecules close together or confine them within a defined nanoscale environment. The review identifies several research directions.
Enzyme scaffolding
An origami compartment can serve as a framework for arranging enzymes at controlled positions. Bringing catalytic components into a planned arrangement may help researchers study how distance and organization affect a reaction. The flask shape itself is not proof of a particular catalytic result; the benefit depends on the attachments and experiment built into each design.
Single-molecule studies
A tiny enclosure can provide a defined setting for observing individual molecular events. Such experiments may use the compartment to hold or position components while optical or biochemical measurements are performed. The nanoflask is therefore a platform for an experiment, not a standalone measuring instrument.
Biosensing
DNA structures can be designed with locations where recognition molecules or reporter components are attached. A compartment may help organize those elements for sensing studies. A published flask geometry does not, by itself, establish a diagnostic device, a clinical test or a validated detection limit.

Biomedical research
DNA nanocompartments are also investigated as tools for biomedical research, including controlled molecular interactions and model delivery concepts. That research context should not be confused with an approved medicine, implant or treatment. The sources cited here establish a laboratory nanostructure, not a clinical product.
What the microscopy shows—and what it cannot show
Atomic force microscopy scans a surface and can reveal the outline and height of nanoscale particles. Transmission electron microscopy images objects after preparation for electron imaging and can show their projected morphology. Together, these methods support the claim that curved, flask-like DNA assemblies were made.
Images do not automatically establish the exact internal volume, fluid-tightness, loading capacity or lifetime of every particle. Those properties require separate measurements. The available description gives no flask-specific capacity, leakage test, storage lifetime or quantitative loading result, so those details should not be inferred from the shape alone.

Production is a separate challenge from design
Designing a DNA origami object and producing enough material for experiments are different problems. A 2017 Nature paper reported a method for scalable production of DNA strands and demonstrated production of a DNA origami nanorod in a litre-scale stirred-tank bioreactor. That result addressed strand manufacturing and a nanorod demonstration. It did not demonstrate litre-scale production of the rounded-bottom nanoflask, nor did it establish commercial availability of that flask.
For the same reason, a general DNA origami kit, a book about DNA nanotechnology or ordinary laboratory glassware should not be presented as the flask itself. The cited sources do not identify a catalog product or consumer supplier for this specific structure. Making and verifying one would require a designed set of DNA strands and laboratory methods for assembly and nanoscale imaging.
What remains unspecified
Several practical questions cannot be answered from the reported description:
- The exact internal capacity of the rounded-bottom flask is not stated.
- The source does not provide a universal recipe for loading, emptying or sealing the cavity.
- No flask-specific stability duration, temperature range or chemical compatibility result is reported.
- No production yield, retail price or consumer purchase route is established.
- No evidence here demonstrates that the flask is better than other DNA nanocompartment geometries for a particular experiment.
These limits do not undermine the core result. They define what the evidence supports: a deliberately curved, hollow DNA origami structure and a broader research platform whose dimensions and functions vary by design.
Bottom line
The DNA origami flask is a nanoscale container assembled by folding a long DNA scaffold with many short staple strands. Strategic crossover and nick placement let researchers control curvature well enough to create a rounded-bottom, flask-like cavity. The approximately 10–150 zeptoliter figure belongs to the wider class of reviewed nanocompartments, not to this flask as a measured specification. Its significance is as a programmable research architecture for molecular experiments—not as a tiny piece of glassware or a consumer product.
