Mammary glands likely evolved from skin glands associated with hair follicles in the ancestors of mammals. A leading hypothesis is that early secretions helped keep parchment-shelled eggs moist and may have offered protection from microbes; offspring could then consume the secretion, which gradually became a more specialized source of nourishment. The proposed sequence is a reconstruction from anatomy, development, milk biology and fossils—not a chain directly preserved in the fossil record.

Why the origin of milk is hard to see in the fossil record

Mammals are closely associated with lactation, but the glands that produce milk are soft tissues. They rarely fossilize, so researchers cannot usually point to an ancient fossil and see a mammary gland or a nursing event. The earliest stages of lactation must instead be inferred from evidence that survives: the arrangement and development of glands in living mammals, comparisons among milk constituents, and clues in fossil anatomy.

That distinction matters. There is a plausible account of how lactation could have begun, but no fossil sequence demonstrates each step from an ancestral skin gland to modern milk production. A review of lactation’s origins describes the evidence as a combination of comparative biology and paleobiology, rather than direct observation of the first mammary glands (“The origin and evolution of lactation,” 2009).

The broad evolutionary setting is the synapsid lineage, the group from which mammals arose. The hypothesis does not require the earliest secretions to have been milk in the familiar modern sense. It proposes that a skin secretion could have taken on useful roles before it became a substantial, nutrient-rich food for young.

How a skin secretion could have become milk

A leading explanation begins with glands associated with hair follicles. Reviews of mammary evolution propose that mammary glands arose from apocrine-like skin glands. This connection is consistent with the association between hair follicles and mammary structures in monotremes, and with developmental evidence from other mammals (Oftedal, “The mammary gland and its origin during synapsid evolution,” 2002; “Evo-devo of the mammary gland,” 2013).

One proposed early benefit of the secretion was egg protection. Under this hypothesis, secretions spread over parchment-shelled eggs could have helped limit drying. They may also have contained substances that reduced microbial risk. If hatchlings licked or otherwise consumed the secretion, it could have provided a further benefit as food. Over many generations, natural selection could have favored secretions that better supported offspring, with a gradual shift toward more nutrient-rich and specialized milk.

This is a plausible evolutionary pathway, not an established account of exactly what the first secretions did. The hydration and protective functions are proposed explanations, not preserved behaviors or molecules directly documented in an early synapsid fossil. Nor does the hypothesis mean that egg protection and nourishment must have appeared as cleanly separated stages. A secretion could potentially have served more than one role, and the transition would have involved changes over evolutionary time.

The idea helps explain a possible bridge between skin glands and lactation: glands do not have to begin as food-producing organs to become useful in feeding young. But the exact sequence—from ancestral gland type, to protective secretion, to food, to complex milk—remains a reconstruction. The evidence supports a likely skin-gland ancestry more strongly than it establishes the first function or the precise order of changes.

What does the 310-million-year estimate mean?

Olav T. Oftedal’s 2012 review suggests that milk’s origin as a glandular skin secretion in synapsids may reach back to the Pennsylvanian, about 310 million years ago (“The evolution of milk secretion and its ancient origins,” 2012). The date is an evolutionary estimate for possible early secretions, not a directly measured age for a fossil mammary gland, and not a date at which modern milk is known to have appeared.

It is therefore useful as a sense of how deep the proposed history may be, but not as a precise timestamp. The estimate concerns a possible precursor in the synapsid lineage. Mammary tissue itself is not directly visible in the reviewed fossil evidence, and the date does not tell us when every feature of modern lactation evolved.

How living mammals reveal different versions of lactation

Living monotremes, marsupials and eutherians—the major mammalian groups represented in comparative studies—help biologists examine how mammary anatomy and development vary. They are not unchanged ancestors or snapshots of a single evolutionary ladder. Each group has its own evolutionary history; together, their differences provide comparisons for studying an ancient shared system.

Group What the reviewed comparisons show What that can—and cannot—tell us
Monotremes A nipple-less mammary patch, with mammary structures associated with hair follicles. This living arrangement supports comparison with a skin-gland origin. It does not establish that the first mammal or the earliest lactating synapsid had the same anatomy.
Marsupials Comparative developmental work identifies growth patterns that differ from those described in monotremes and some eutherians. Developmental differences show that the mammary system diversified; they do not, by themselves, reveal the exact sequence of its earliest changes.
Eutherians Some eutherians show developmental patterns distinct from those in monotremes and marsupials. These patterns add a comparison among living mammals, but do not make any living group a direct stand-in for an ancestor.

The comparisons are valuable partly because the mammary system is more than the visible shape of a gland. Its development, the timing and dependence of offspring nutrition, and the composition of milk can all vary. The reviewed evo-devo work describes different growth patterns across monotremes, marsupials and some eutherians, while emphasizing comparative evidence rather than a direct record of the ancestral condition (“Evo-devo of the mammary gland,” 2013).

Reproductive strategy also matters to the comparison. Monotremes lay eggs, while marsupials and eutherians give birth to live young. Those differences make it possible to consider lactation in distinct reproductive contexts, but they do not prove that egg-laying mammals preserve the ancestral form of every mammary feature. The safe inference is that living groups exhibit different arrangements and developmental patterns within a system that has diversified.

Milk did not become complex all at once

Milk is not a single substance that appeared fully formed. Reviews discuss the evolution of caseins, lactose, milk lipids and oligosaccharides, as well as milk’s effects on offspring development beyond basic nourishment. These constituents have distinct evolutionary histories, so it would be misleading to imply that they all emerged simultaneously or followed one simple path (Lefèvre, Sharp and Nicholas, “Evolution of Lactation: Ancient Origin and Extreme Adaptations of the Lactation System,” 2010; Oftedal, “The Evolution of Lactation in Mammalian Species,” 2020).

This gradual, component-by-component view fits the proposed transition from a skin secretion to a more specialized resource. Changes in what a secretion contained, how it was produced and how offspring used it need not have happened together. The modern system reflects an accumulation of evolutionary adaptations, not one sudden invention of a complete formula.

Milk also contributes to offspring development in ways that exceed the simple delivery of calories and basic nutrients. The reviews treat lactation as a wider developmental system, while the detailed roles of individual constituents vary. That broader view helps explain why the evolution of lactation cannot be reduced to the question of when a secretion first became food.

What fossils can tell us about early milk consumption

Fossil evidence offers indirect clues. Soft mammary tissue rarely provides direct evidence in the reviewed account, but fossil anatomy and developmental patterns can support an inference that young mammaliaforms consumed milk. One example discussed in the literature is delayed tooth development. Such a clue can be consistent with milk intake during early life, but it does not directly show a gland, the composition of a secretion or the behavior of a parent and offspring (Oftedal, 2020; “The origin and evolution of lactation,” 2009).

That is the difference between evidence for lactation and evidence for a mammary gland. A fossil trait may support the conclusion that an animal’s young depended on milk or a milk-like secretion, while leaving the soft tissue that produced it unknown. Several kinds of evidence can make a reconstruction more persuasive, but they do not turn an inference into a directly preserved gland.

What is known, and what remains a hypothesis?

The strongest broad conclusion is that mammary glands likely have a skin-gland ancestry associated with hair follicles. Living anatomy and developmental comparisons support that connection. The origin of milk as a glandular secretion in early synapsids may be very ancient; one review places a possible origin as far back as about 310 million years ago, with the important qualification that this is an estimate rather than a fossil date for a modern mammary gland.

The idea that early secretions protected parchment-shelled eggs from drying and microbes, before offspring gained nutrition from them, is a plausible hypothesis. It offers a way to connect a skin secretion to lactation, but the first function and exact order of evolutionary changes are not directly established. Fossils can add indirect clues to milk consumption, and living mammals reveal diversity in gland arrangement and development, but neither evidence source supplies a complete record of the transition.

The most accurate answer, then, is that mammary glands probably evolved by modifying skin glands, while lactation developed gradually as secretions acquired new roles and components. The outline is supported by comparative evidence; the earliest stages remain inferred rather than directly observed.
