Crime-scene DNA testing can now be performed in or near the field with integrated Rapid DNA instruments, but “portable” does not mean instant, universally approved, or automatically eligible for a CODIS search. These systems automate extraction, amplification, separation, detection and allele calling. Published agency descriptions put the instrument workflow at under 90 minutes (National Institute of Standards and Technology) or one to two hours for a mouth-swab profile (Federal Bureau of Investigation). Those figures describe defined workflows, not a guaranteed result from every unknown scene sample.
What “Rapid DNA” means at a crime scene
Rapid DNA is an automated DNA-profile generation workflow, not a handheld detector that identifies any object immediately. A suitable sample is loaded into a closed system, and the instrument performs the major laboratory stages: DNA extraction, polymerase-chain-reaction amplification, separation of genetic fragments, detection and allele calling. The FBI describes this as a “swab in – profile out” process. The result is a DNA profile for interpretation within the governing forensic process; it is not, by itself, a complete identification or proof that a person committed a crime.
The technology was developed for rapid point-of-collection use, especially a known person’s buccal (inner-cheek) reference swab collected during booking. NIST also identifies potential applications such as mass-fatality operations, crime scenes, point-of-arrest reference sampling and human-trafficking investigations. A mobile instrument may therefore be useful at a scene, a booking location or a disaster facility, but the sample type, validation, quality system and intended database use determine what the result can legally and operationally support.
How the automated workflow operates
1. Collection and sample assessment
Personnel collect a swab or other validated input under an agency protocol. Before loading it, they assess whether the sample is suitable for the particular instrument and assay. A clean reference mouth swab is a much more predictable input than a touched object, a mixed stain, a degraded trace or material containing inhibitors. The instrument cannot correct for an unsuitable collection, contamination or an interpretation problem that began before the cartridge was closed.

2. Extraction
The system releases DNA from cells on the swab and prepares it for amplification. In a closed workflow, extraction reagents and disposable components are contained in a cartridge or chip. Operators still need documented lot controls, expiration checks, storage controls and procedures for spills, failed runs and waste.
3. Amplification
Polymerase-chain-reaction amplification copies selected forensic genetic markers. The marker set and chemistry are tied to the approved cartridge or chip and software configuration. A run can fail because the sample has too little DNA, contains inhibitors, is heavily degraded or is outside the validated sample type.
4. Separation and detection
The amplified fragments are separated and detected so that the instrument can measure allele signals. These steps replace several conventional bench-laboratory operations, which is why Rapid DNA is described as an integrated system rather than simply a faster collection kit.
5. Allele calling and review
Software converts detected signals into an allele profile. A trained, authorized reviewer and the applicable laboratory quality system determine whether the profile is technically acceptable, whether artifacts or mixtures are present and what use is permitted. A software-generated profile should never be treated as self-authenticating evidence.
How long a field result takes
Two official descriptions provide useful scale but should not be presented as a head-to-head test. NIST describes the integrated Rapid DNA typing process as completing in under 90 minutes. The FBI describes Rapid DNA analysis of a mouth swab in one to two hours. The sources frame these timings differently, so neither figure is a promise that an unknown crime-scene sample will produce a usable profile within that period.

- Instrument time: the automated run after a sample is accepted and loaded.
- Operational time: collection, labeling, security checks, setup, quality checks, review and reporting in addition to the run.
- Case turnaround: the time until an investigator or court can rely on the result, which may include repeat testing, laboratory confirmation, database procedures and interpretation.
A failed run, a mixed profile, a low-level sample or a need for confirmatory laboratory testing can make the overall process much longer. Agencies should publish a local service-level expectation only after validating their own workflow and documenting which steps are included.
Booking swabs and crime-scene evidence are different use cases
This distinction controls how a result may be used. A booking-station reference swab comes from a known person whose identity and collection circumstances are documented. The basic FBI Rapid DNA description is built around this buccal reference workflow. An unknown scene sample may be a single-source stain, a touch deposit, a mixture, an environmentally damaged trace or a sample with substances that interfere with the chemistry. It requires separate forensic validation and interpretation rules.

Carrying that instrument to a scene does not expand its validated sample menu, authorize every type of analysis or make the resulting profile admissible for every purpose.
CODIS eligibility and quality requirements
Forensic casework intended for CODIS searching or upload is governed by laboratory accreditation and quality requirements, not by the instrument’s portability. The FBI states that, effective July 1, 2025, forensic Rapid DNA casework must meet the applicable FBI requirements. The operation must fall under a CODIS laboratory’s ISO/IEC 17025 accreditation, and samples processed on or after that date must be handled under the 2025 Forensic Quality Assurance Standards.
The live FBI Rapid DNA requirements page identifies six criteria for CODIS search or upload eligibility. Only part of that checklist is exposed in the material available here, so an agency should consult the current page and its state or local CODIS administrator before treating a particular workflow as eligible. Do not infer the remaining criteria from a product brochure or from the fact that a profile was generated.

The FBI’s forensic DNA fact sheet explains that crime-scene evidence is sent to a forensic DNA laboratory for analysis and that CODIS participation is controlled by FBI Quality Assurance Standards. A mobile run therefore needs laboratory oversight, validated procedures, chain-of-custody records, documented review and a data-management process that can withstand audit and court scrutiny.

Rapid DNA listings are configuration-specific. Model name alone is not enough: the chip or cartridge and the system and expert software versions must match the current entry. The FBI list retrieved for this article identifies the following configurations, each marked for mouth swabs only.
| Instrument | Consumable | Software configuration listed | Sample scope shown |
|---|---|---|---|
| ANDE 6C Instrument | A-Chip (FlexPlex) | System Software 2.0.6; Expert System 2.0.5 | Mouth swabs only |
| Applied Biosystems RapidHIT ID | RapidHIT ID ACE GlobalFiler Express cartridge | Enumerated system-software and expert-system versions on the FBI entry | Mouth swabs only |
Software and consumable listings can change. Before procurement, deployment or a court filing, verify the live FBI page, the exact version identifiers and the intended forensic application. The FBI’s BioSpecs guidance advises agencies considering forensic DNA equipment and supplies to contact their state or local CODIS administrator.
What a managed mobile deployment requires
A field instrument is only one component of a managed forensic workflow. A practical deployment plan should assign responsibility for each stage below.
- Define the use case. Decide whether the operation is collecting a known reference, screening a scene for an investigative lead, supporting a mass-fatality response or producing forensic casework intended for a database or court. The answer determines validation, review and reporting requirements.
- Confirm sample suitability. Use only sample types covered by the current validated method. For an unknown stain or touch sample, establish in advance whether the instrument has a validated protocol; a mouth-swab-only listing is not sufficient.
- Control contamination. Use clean collection supplies, appropriate personal protective equipment, negative controls and physical separation between collection, loading and waste handling. Record who handled each item and when.
- Preserve chain of custody. Assign a unique identifier, photograph or otherwise document the item, seal it, log transfers and record the instrument, cartridge or chip lot and operator.
- Run performance checks. Follow the laboratory’s startup, calibration, control and maintenance schedule. A passing instrument check does not prove that a particular scene sample is suitable.
- Secure the data. Restrict access to raw files, allele calls and reports; synchronize clocks; back up records; and document any transfer to a laboratory information system or authorized database.
- Review and report. Have an authorized reviewer assess controls, signal quality, possible artifacts and mixture indications. State the sample type, method, software version, limitations and intended use in the report.
- Escalate when necessary. Send the item to a conventional forensic DNA laboratory when the run fails, the profile is mixed or partial, the sample falls outside validation, or confirmatory testing is required.
Field logistics that can determine success
Environment and power
Vehicles, temporary facilities and disaster sites need stable power, temperature control, secure work surfaces and protection from dust, moisture and unauthorized access. A plan should include charging or generator capacity, spare approved consumables, waste containment and a way to move evidence without breaking the chain of custody.

People and training
Operators need training on collection, contamination prevention, cartridge loading, error handling, data security and documentation. A separate qualified reviewer may be required under the governing laboratory’s quality system. “Easy to operate” does not eliminate competency assessment or continuing records.
Data and communications
A scene may have poor connectivity. The workflow should support secure local storage and later transfer, with access controls and audit logs. Never assume that a profile can be uploaded to CODIS directly from a scene device; authorization and laboratory procedures govern that step.
Disaster operations
The National Institute of Justice’s mass-disaster toolkit includes a Rapid DNA processing and data-management protocol. It addresses activities from performance checks through reporting and includes guidance on non-CODIS crime-scene use and courtroom considerations. Agencies can use that type of protocol to assign roles before a large incident rather than improvising evidence handling during one.
What a Rapid DNA result can and cannot establish
- It can provide: an allele profile from a suitable sample, an investigative comparison against an authorized reference, or a result that enters a validated laboratory workflow.
- It cannot automatically provide: a person’s identity from every trace, a distinction between legitimate contact and criminal activity, a complete answer for a mixture, or automatic CODIS eligibility.
- It does not replace context: investigators still evaluate how the sample was deposited, whether transfer is plausible, whether the reference was lawfully collected and what other evidence supports or contradicts the association.
No field success rate, false-match rate or case-resolution impact figure is established in the cited material. NIJ reports $500,000 in additional fiscal-year 2022 funding for an FBI collaborative evaluation of enhanced Rapid DNA limitations for crime-scene samples involving 12 laboratories. Those numbers describe study funding and design, not accuracy or effectiveness.
How to compare a Rapid DNA option
Agencies should compare complete, approved workflows rather than purchase prices or run-time claims alone.

| Decision axis | Questions to document |
|---|---|
| Intended use | Booking reference, disaster response, investigative scene lead or CODIS-eligible forensic casework? |
| Sample type | Which swabs, stains, mixtures or degraded samples are actually validated? |
| Configuration | What exact model, chip or cartridge, system software and expert software versions are listed? |
| Quality system | Which CODIS laboratory owns the method, and how does ISO 17025 accreditation cover the operation? |
| Timing | Does the quoted time cover only the instrument run or also setup, review and reporting? |
| Data management | How are raw data, allele calls, audit logs, backups and authorized transfers protected? |
| Failure handling | What triggers a repeat, referral to a conventional laboratory or exclusion from database use? |
| Field support | What power, environmental controls, training, maintenance, consumables and waste procedures are required? |
The available sources do not provide a current head-to-head trial that ranks the listed instruments. A procurement decision should therefore be based on the agency’s validated use case and quality obligations, not an assumed winner.
Common mistakes and how to avoid them
Calling any portable profile “FBI approved”
Approval is tied to a current configuration, sample scope and quality conditions. Check the live listing and avoid expanding a mouth-swab entry to crime-scene evidence.
Promising a result in 90 minutes
Report the NIST under-90-minute description or the FBI one-to-two-hour mouth-swab description with its source framing. Explain that field setup, unsuitable samples, review and confirmation can add time.
Recommended Free Tools
Uploading every scene profile to CODIS
Database searching requires the applicable FBI criteria, accredited laboratory operation and compliant quality procedures. A mobile run may be an investigative lead without being CODIS-eligible.
Treating a software call as a conclusion
Allele calls require quality review and interpretation. Mixed, partial or low-level profiles may need laboratory examination and cannot be reported as an unqualified match.
Using consumer collection supplies
Generic swabs or kits are not automatically suitable for evidentiary work. Use supplies approved by the agency protocol, document lot information and follow contamination controls.
A practical pre-deployment checklist
- Current FBI configuration and sample-scope verification completed.
- State or local CODIS administrator consulted for the intended use.
- ISO 17025 accreditation coverage and 2025 Forensic Quality Assurance Standards requirements documented where applicable.
- Validated collection, contamination, control, review and reporting procedures approved.
- Operator competency, maintenance, software-version and consumable-lot records established.
- Chain-of-custody, secure storage, backup and transfer procedures tested.
- Failure, referral and courtroom-disclosure procedures written before deployment.
Rapid DNA can shorten the path from collection to a usable profile, particularly for controlled reference swabs and planned disaster operations. Its value in crime-scene work depends on the entire managed process: validated sampling, contamination control, accredited oversight, secure data handling and careful interpretation.
